These results suggested that the administration of CT improved BW gain and induced bacterial community changes in the rumen of steers grazing wheat forage.Safety and quality of compound feed for experimental animals in Costa Rica is unknown. Some contaminants, such as Salmonella spp. and mycotoxins, could elicit confounding effects in laboratory animals used for biomedical research. In this study, different batches of extruded animal feed, intended for laboratory rodents in Costa Rica, were analyzed to determine mycotoxin and microbiological contamination (i.e., Salmonella spp., Escherichia coli, total coliform bacteria, and total yeast and molds enumeration). Two methods for Salmonella decontamination (UV light and thermal treatment) were assessed. Only n = 2 of the samples were negative (representing 12.50%) for the 26 mycotoxins tested. Enniatins and fumonisins were among the most frequent toxins found (with n = 4+ hits), but the level of contamination and the type of mycotoxins depended on the supplier. None of the indicator microorganisms, nor Salmonella, were found in any of the tested batches, and no mold contamination, nor Salmonella growth, occurs during storage (i.e., 2-6 months under laboratory conditions). However, mycotoxins, such as enniatins and fumonisins tend to decrease after the fourth month of storage, and Salmonella exhibited a lifespan of 64 days at 17 °C even in the presence of UV light. The D-values for Salmonella were between 65.58 ± 2.95 (65 °C) and 6.21 ± 0.11 (80 °C) min, and the thermal destruction time (z-value) was calculated at 15.62 °C. Results from this study suggest that laboratory rodents may be at risk of contamination from animal feed that could significantly affect the outcomes of biomedical experiments. Thus, improved quality controls and handling protocols for the product are suggested.Two experiments were conducted to compare a supplemental blend of essential oils alone (EO) or combined with enzymes (EO + ENZ) versus virginiamycin (VM), on characteristics of growth performance (Exp. 1) and digestion (Exp. 2) in finishing lambs. Lambs were fed a high-energy finishing diet supplemented with (1) no supplement (control); (2) 150 mg supplemental EO; (3) 150 mg supplemental EO plus 560 mg alpha-amylase (EO + ENZ); and 4) 25 mg VM. Compared with the control, growth performance response to EO and VM were similar, enhancing (5.7%, p less then 0.05) feed efficiency and observed dietary net energy. Tosedostat ic50 Compared with control, supplementation with EO + ENZ tended (p = 0.09) to increase dry matter intake (6.8%), improving (p less then 0.05) weight gain and feed efficiency (10.4 and 4.4%, respectively). Dietary energy utilization was greater (2.7%, p less then 0.05) for EO and VM than EO + ENZ. Treatment effects on the carcass and visceral mass were small, but additive supplementation decreased (p ≤ 0.03) the relative weight of the intestines. link2 There were no treatment effects on measures of digestion nor digestible energy of the diet. Supplemental EO may be an effective alternative to VM in high-energy finishing diets for feedlot lambs. Combination EO + ENZ may further enhance dry matter intake, promoting increased weight gain.Cattle are the main reservoir of Shiga toxin-producing Escherichia coli (STEC), one of the world's most important foodborne pathogens. The pathogen causes severe human diseases and outbreaks. This study aimed to identify and characterize non-O157 STEC isolated from cattle feces from central and southern Chile. We analyzed 446 cattle fecal samples and isolated non-O157 STEC from 12.6% (56/446); a total of 93 different isolates were recovered. Most isolates displayed β-glucuronidase activity (96.8%; 90/93) and fermented sorbitol (86.0%; 80/93), whereas only 39.8% (37/93) were resistant to tellurite. A subgroup of 30 representative non-O157 STEC isolates was selected for whole-genome sequencing and bioinformatics analysis. In silico analysis showed that they grouped into 16 different sequence types and 17 serotypes; the serotypes most frequently identified were O116H21 and O168H8 (13% each). A single isolate of serotype O26H11 was recovered. One isolate was resistant to tetracycline and carried resistance genes tet(A) and tet(R); no other isolate displayed antimicrobial resistance or carried antimicrobial resistance genes. The intimin gene (eae) was identified in 13.3% (4/30) of the genomes and 90% (27/30) carried the stx2 gene. A phylogenetic reconstruction demonstrated that the isolates clustered based on serotypes, independent of geographical origin. These results indicate that cattle in Chile carry a wide diversity of STEC potentially pathogenic for humans based on the presence of critical virulence genes.Canine parvovirus (CPV) is a major pathogen in canines, with a high mortality rate in unvaccinated puppies. CPV is traditionally classified into three antigenic variants (CPV-2a, CPV-2b and CPV-2c) based on the amino acid sequence of the VP2 protein. Currently, various mutations are described in the receptor-binding area or in the regions of greatest antigenicity of the VP2 protein, giving rise to new viral variants that are capable of immunological escape, affecting the protective immunity of traditional vaccines. In the present study, a molecular characterization of the VP2 gene was performed, which included phylogenetic analysis, amino acid characterization and determination of selection pressures. Blood samples were initially collected from canine patients with clinical signs of gastrointestinal infection, of which 69 were positive for CPV as measured by means of PCR and 18 samples were selected for the amplification of the complete VP2 gene. The analysis revealed a higher rate of CPV-2c-positive patients compared to CPV-2b. link3 Furthermore, the amino acid characterization of VP2 indicated mutations in the regions of highest antigenicity previously described in the literature (CPV-2b 297 and 324; CPV-2c 440), as well as others not previously documented (CPV-2b 514; CPV-2c 188, 322, 379, 427 and 463). Our analysis of selection pressure showed that the VP2 gene is under negative selection. However, positive selection point sites were identified, both in CPV-2c (324, 426 and 440) and CPV-2b (297 and 324), at sites that have been associated with evasion of the immune response via antigenic drift, which possibly has implications for the protective immunity generated by traditional vaccines.This study aimed to determine the thyroid and lipid profiles in 30 Nicastrese goats, along different physiological periods before mating (nonpregnant goats), during the whole pregnancy (pregnant goats), and during postpartum and early lactation (milking goats). Blood samples were collected monthly from March 2020 to January 2021. Serum thyroid-stimulating hormone (TSH), total and free triiodothyronine (T3, fT3), and thyroxine (T4, fT4) concentrations were measured using immunoenzymatic assay kits and serum lipid panels (triglyceride (TG) and total cholesterol (tCho)) by enzymatic colorimetric method; very-low-density lipoprotein cholesterol (VLDL Cho) was calculated. Pregnant and milking goats showed the lower T3 (p less then 0.0002) and T4 (p less then 0.0005) concentrations, with lower BCS (p less then 0.001) only in pregnant ones. Milking goats showed tCho (p less then 0.006) concentrations lower than nonpregnant ones, and TG and VLDL Cho (p less then 0.001) lower than both pregnant and nonpregnant goats. T4T3 and T3fT3 were significantly and positively correlated in both pregnancy and lactation. Under similar environmental, nutrition, and management conditions, different physiological phases play a significant role in the thyroid and lipid profiles in Nicastrese goats. These endocrine and metabolic resources could contribute to the knowledge useful for the salvage of this endangered, local, native breed.The use of pea (Pisum sativum) has been recommended to replace soybean meal in the diet of ruminants, but it may affect meat quality. The aim of this study was to evaluate the effect of the proportion of pea (0%, 10%, 20% and 30%) in fattening concentrates fed to light lambs for 41 days on carcass colour and on the meat quality. Pea inclusion affected neither the colour of the lamb carcasses nor affected most of the parameters of the meat quality. However, the inclusion of pea affected the cholesterol content, and the 20%pea concentrate yielded meat with greater cholesterol contents than the 30%pea concentrate did (p less then 0.05). The inclusion of pea had minor effects on individual FAs but affected the total saturated fatty acids (p less then 0.01) and the thrombogenicity index (p less then 0.05). A greater total saturated fatty acid content was recorded for the 20%pea concentrate than for the rest of the concentrates, and a greater thrombogenicity index was recorded for the 20% concentrate than for the 10%pea concentrate. The results indicated the viability of the inclusion of pea in the fattening concentrate of light lambs without impairing meat quality, with the 30%pea concentrate being the most suitable to reduce the soya-dependency.Collaborative semen collection in monkeys is a valuable tool in research, animal collection management, and conservation efforts. To obtain samples, monkeys are often restrained in open restraint chairs (ORC) with the "pole and collar" technique. While commonly used, this restraint is not tolerated by all individuals; some become anxious or aggressive towards the poles and people. In an effort to refine this procedure and improve welfare of the monkeys, we examined the use of a "closed box chair" (CBC), a clear, plexiglass box in which the monkey is trained to sit for sperm collection. The CBC does not require pole and collar, and although legs are secured, the arms and neck are not restrained. The use of CBCs has increased in recent years; however, there are few studies demonstrating its effects on scientific outcomes. We used positive reinforcement techniques to train 34 adult male rhesus macaques (Macaca mulatta) to provide semen samples using either the ORC or the CBC. While all CBC monkeys (n = 14) were reliably trained for this procedure, only 75% of ORC (n = 20) males completed the training (p = 0.04). It took significantly less time to train animals in the CBC than the ORC (201.0 vs. 412.4 min; p less then 0.001). In a controlled subset, males restrained with ORC (n = 7) produced a significantly lower ejaculatory volume than those collected by CBC (n = 10) (297.6 µL vs. 522.1 µL respectively; p = 0.04) and had a lower concentration of sperm (186.0 × 106/mL vs. 367.5 × 106/mL respectively; p = 0.017), although there were no differences with respect to sperm motility (p = 0.15). Our data suggest the closed box chair technique reduces stress on the animals while enhancing semen quality, supporting the use of the CBC as an important refinement.More than 61% of all human pathogens are zoonotic, representing 75% of all emerging pathogens during the past decade. In this Special Issue, we approach a myriad of zoonotic infectious diseases and their complex mechanisms. This Special Issue is composed of three reviews on zoonotic diseases of African Lions, hemogregarine classification, and hepatitis E virus in Brazil, followed by one letter and one opinion piece that broadens the spectrum of disease emergence to mechanistic aspects of emerging non-communicable diseases.
