All recovered colonies were presumptively identified according to colony color described by manufacturer, and the definitive identification was performed by mass spectrometry at 48 h. A total of 134 C. auris isolates were obtained (101 from patients' surveillance samples, and 33 from environmental samples). Sensitivity, specificity, and predictive positive and negative values were 99.5%, 100%, 100%, and 99.1%, respectively, for the main clinical Candida species, showing that CC-Plus is comparable to CC, with the advantage of being able to differentiate C. auris from C. parapsilosis. Furthermore, CC-Plus was able to detect one C. albicans, one C. glabrata, and eight C. auris that did not grow in CC. Additionally, the yeast colonies were generally larger, suggesting that this novel medium could be a richer medium, and suitable for surveillance and environmental cultures of C. auris and other clinically relevant Candida species.Secondary metabolites of actinomycetes are a potential source of bioactive compounds in the agricultural sector. This study aimed to determine the fungicidal properties of extracts of marine organism-derived actinomycetes. Actinomycetes were isolated from marine organisms using agar media with 1% colloidal chitin in artificial seawater. Then, the isolates were cultured on liquid media with 1% colloidal chitin in artificial seawater under static conditions for 14 days. The culture was extracted, the fungicide properties were evaluated using the microtiter 96-well plate method, and the influence of inhibition was visualized using apotome and SEM. Finally, the active extract was analyzed using LCMSMS. In the present study, 19 actinomycetes were isolated from marine organisms, and the isolates were examined with regard to their antifungal activities. Of these nineteen isolates, the isolate 19C38A1 was picked out from the rest. Hence, it showed significant control towards F. oxysporum. The prospective strain 19C38A1 was determined to be Kocuria palustris 19C38A1. The extract 19C38A1 was shown to cause damage to cell integrity, indicated by the shrinking form, and inhibited germination in the F. oxysporum; subsequently, the chemical characteristics of the compound produced by the potential isolate 19C38A1 indicated the presence of benzimidazole compounds in the active fraction of C38BK2FA. These results indicate that actinomycetes derived from marine organisms near the coast of Oluhuta, Tomini Bay, Gorontalo, related to strain 19C38A1, are not widely known as sources of valuable fungicides. This preliminary information is important, as it can be used as a basis for further development in the search for fungicides derived from marine actinomycetes.Onychomycosis is the most common nail disease encountered in clinical practice. The Boletaceae-wide and Xerocomoideae-wide phylogenetic analyses based on four-gene data sets (atp6, cox3, rpb2, and tef1) support Rubinosporus as monophyletic and places it in Boletaceae subfamily Xerocomoideae. Alternaria species are saprobic-largely involved in the decomposition of organic material-but they can also act as animal pathogens, causing disease in humans and animals, developing infections, toxicosis and allergic diseases. The current study examined the mechanism of said degradation and tested therapeutic and food-grade oils (instead of finishing oils) for their potential to carry draconin red, the pigment from Scytalidium cuboideum, without color loss over time. FTIR analysis from finishing oils indicated that oxidation was not likely the cause of color loss as the pigment could not be distinguished from the oils in the IR spectra. SEM was employed to determine if crystal degradation was contributing to color loss and indicated, surprisingly, that the crystals of draconin red formed rather than degraded over time. This suggested crystal breakdown was also not likely the cause of color loss. The pigment did not show degradation in hemp oil, flaxseed oil, and cold-pressed linseed oil when treated with β-carotene. Further in-depth chemical studies are needed to determine the mechanism of color loss in pigmented natural oils; however, food-grade oils appear to be a promising alternative to carry draconin red, without degradation of the color.With rare exceptions, the shape and appearance of lichen thalli are determined by the fungal partner; thus, mycobiont identity is normally used for lichen identification. However, it has repeatedly been shown in recent decades that phenotypic data often does not correspond with fungal gene evolution. Here, we report such a case in a three-species complex of red-fruited Cladonia lichens, two of which clearly differ morphologically, chemically, ecologically and in distribution range. We analysed 64 specimens of C. bellidiflora, C. polydactyla and C. umbricola, mainly collected in Europe, using five variable mycobiont-specific and two photobiont-specific molecular markers. All mycobiont markers exhibited very low variability and failed to separate the species. In comparison, photobiont identity corresponded better with lichen phenotype and separated esorediate C. bellidiflora from the two sorediate taxa. These results can be interpreted either as an unusual case of lichen photomorphs or as an example of recent speciation, in which phenotypic differentiation precedes the separation of the molecular markers. We hypothesise that association with different photobionts, which is probably related to habitat differentiation, may have triggered speciation in the mycobiont species.Nowadays, climate change is not the only threat facing our planet. For this reason, our article is dedicated to offering biofriendly solutions to decrease wastes, give them a positive meaning, such as a substrate for an edible oyster fungus with nutritive and biological properties usefully for humans. Three types of wastes such as coconut coir, pine sawdust, and paper waste-representative symbols of pollution in Ecuador-have been tested as suitable growing substrate for the edible fungi Pleurotus ostreatus (Jacq. ex Fr.) P. Kumm by analyzing parameters such as Biological Efficiency, Mushroom Yield, and Productive Rate. The influence of these "waste" substrates on the nutritive (protein content), biological characteristic (antioxidant activity), and the content of human-health-sustaining compounds (phenols, flavonoids) were also evaluated using the Kjeldahal, DPPH, ABTS, FRAP, and Folin-Ciocalteu methods. The results indicate that all the waste products represent desirable substrates for growing the edible fungi, with more focus on coconut coir waste (one of the principal pollution problems in Ecuador), but that also achieved the increase in the fungi's desirable characteristics. Coconut coir waste could be an environmentally friendly solution that also offers for humans additional nutritive and healthy benefits.Rates of invasive aspergillosis (IA) among COVID-19 ICU patients seem to reach over 30% in certain settings. At Vienna General Hospital (VGH), all rooms in COVID-19 ICUs were put under negative pressure as a protective measure, thus increasing the risk of exposure to environmental pathogens for patients. Even though all ICU patients are surveilled for healthcare-associated infections (HAI), there were concerns that the routine protocol might not be sufficient for IA detection. We reviewed the electronic patient charts of all patients with COVID-19 admitted to ICUs between 1 March 2020 and 31 July 2021 for fungal co- or superinfections, comparing four diagnostic algorithms based on different recommendations for the diagnosis of IA (according to EORTC/MSG, BM-AspICU, IAPA and CAPA) to our routine surveillance protocol. We found that out of 252 patients who were admitted to the ICU during the study period, 25 (9.9%) fulfilled the criteria of probable or possible IA of at least one algorithm. The IAPA definitions detected 25 and the CAPA definition 23 probable and 2 possible cases, out of which only 16 were classified as hospital-acquired IA by routine surveillance. In conclusion, adjustment of the routine protocol using a classification system especially designed for respiratory viral illness seems useful for the surveillance of IA in a highly vulnerable patient cohort.
