The emergence of 5-Fluorouracil (5-FU) resistance is the barrier to effective clinical outcomes for colorectal cancer (CRC) patients. Autophagy was found to be involved in protecting tumor cells from 5-FU. However, the specific role of autophagy-related genes in CRC 5-FU resistance remains unclear. In this study, HSPB8 among 34 differentially expressed ARGs in CRC was identified to be the hub ARGs in 5-FU resistant which was down-regulated in CRC samples when compared with normal samples but up-regulated in CRC samples with relatively higher lymphatic invasion, later stages and poor prognosis of CRC. Mechanistic analysis demonstrated that due to the recruitment of CAFs, HSPB8 expression was enhanced in CRC cells so that HSPB8 could act together with its co-chaperone BAG3 in autophagy drived 5-FU resistance. Furthermore, the augmented expression level of HSPB8 was found to be significantly correlated to the immune cell infiltration such as Treg cells, macrophages, monocyte and dendritic cells and so on. Our results suggested CAFs driving HSPB8 induced CRC 5-FU resistance by promoting tumor autophagy would provide a new strategy in seeking potential CRC therapeutic target.The functionally redundant ubiquitin E3 ligases SIAH1 and SIAH2 have been implicated in the regulation of metabolism and the hypoxic response, while their role in other signal-mediated processes such inflammatory gene expression remains to be defined. Here we have downregulated the expression of both SIAH proteins with specific siRNAs and investigated the functional consequences for IL-1α-induced gene expression. The knockdown of SIAH1/2 modulated the expression of approximately one third of IL-1α-regulated genes. These effects were not due to changes in the NF-κB and MAPK signaling pathways and rather employed further processes including those mediated by the coactivator p300. Most of the proteins encoded by SIAH1/2-regulated genes form a regulatory network of proinflammatory factors. Thus SIAH1/2 proteins function as variable rheostats that control the amplitude rather than the principal activation of the inflammatory gene response.
Delay in seeking emergency obstetric care contributes to high maternal mortality and morbidity in developing countries. One of the major factors contributing to maternal death in developing countries is a delay in seeking emergency obstetric care. This study aimed to assess the proportion and associated factors of delay in deciding to seek emergency obstetric care on institutional delivery among postpartum mothers in the South Gondar zone hospitals, Ethiopia, 2020.
An institution-based cross-sectional study design was conducted from September to October 2020. A total of 650 postpartum mothers were recruited using a systematic random sampling technique. We collected the data through personal interviews with pretested semi-structured questionnaires. We used a logistic regression model to identify statistically significant independent variables, and entered the independent variables into multivariable logistic regression. The Adjusted Odds Ratio was used to identify associated variables with delay in decidiny area. Therefore, stakeholders must address them to reduce the proportion of delay in deciding to receive on-time obstetric care as per the standards.

Our research aims to analyse the impact of COVID-19 on SCs and enable organisations to prioritise solutions based on their relative importance. The research consists of two main phases. Phase I analyses the impact of the pandemic on SCs in terms of challenges, concerns, steps, and solutions with the goal of SC resilience. The second phase proposes a merged Analytic Network Process-Technique for Order Preference by Similarity to Ideal Solution framework (ANP-TOPSIS) to prioritise the solutions that considers the complicated interrelationships between the factors involved in decision-making. This can be ascribed to a variety of factors, e.g., increasing Internet use, the Open Access movement, academic pressure to publish, and the emergence of publishers with questionable interests that cast doubt on the reliability and the scientific rigor of the articles they publish. All this has transformed the scholarly and scientific publishing scene and has opened the door to the appearance of journals whose editorial procedures differ from those of legitimate journals. These publishers are called predatory, because their manuscript publishing process deviates from the norm (very short publication times, non-existent or low-quality peer-review, surprisingly low rejection rates, etc.). The object of this article is to spell out the editorial practices of these journals to make them easier to spot and thus to alert researchers who are unfamiliar with them. It therefore reviews and highlights the work of other authors who have for years been calling attention to how these journals operate, to their unique features and behaviors, and to the consequences of publishing in them. The most relevant conclusions reached include the scant awareness of the existence of such journals (especially by researchers still lacking experience), the enormous harm they cause to authors' reputations, the harm they cause researchers taking part in promotion or professional accreditation procedures, and the feelings of chagrin and helplessness that come from seeing one's work printed in low-quality journals. Future comprehensive research on why authors decide to submit valuable articles to these journals is also needed. However, the material of shells and spines is thought to be rich in natural bioactive molecules. The current study used liquid chromatography-electrospray mass spectrometry to extract summer quinones pigment present in spines and shells of the burrowing sea urchin 'black' type Echinometra mathaei from the natural Qeshm Island echinoculture. Then, the biochemical, antioxidant, anti-inflammatory, antidiabetic, antibacterial, and cytotoxic activities of sea urchin quinones pigment were investigated. In terms of bioactivity, both shell and spine pigments demonstrated strong radical scavenging activity (antioxidant). The shell pigment exhibited maximum albumin denaturation inhibition (IC50 = 9.62 μg/ml) (anti-inflammatory), as well as α-amylase inhibition (92.28 percent 4.77) (antidiabetic). Pigments were discovered to have a low antibacterial effect against positive gramme bacteria, as well as low cytotoxic and embryotoxic effects when compared to Artemia salina and zebrafish (Danio rerio). This paper aims to determine the cost-optimal nZEB reform strategy for Spanish university buildings located in the Mediterranean climate, evaluating the influence of the building's orientation on nZEB cost optimality. The present work carries out a case study on three similar buildings at the University of Málaga. The optimal cost-efficiency nZEB reform strategy is determined, rigorously maintaining the formal design of the building through energy simulations with Sefaira Systems software under the EPBD framework. Six reform options were proposed to determine the cost-optimal strategy meeting nZEB requirements for each building. The results show that the most profitable strategy is to improve the efficiency of the thermal envelope and the HVAC system, reducing the energy consumption of the studied buildings by 85-93%. Although a general strategy for nZEB renovations was identified, building orientation proved to be a governing factor in energy performance. As such, the cost-optimal reform strategy was found to be specific to each particular building.Mucopolysaccharidosis type II (MPS II), also known as Hunter syndrome, is a rare X-linked recessive disease caused by a deficiency of the lysosomal enzyme iduronate-2-sulfatase (IDS), which activates intracellular accumulation of nonmetabolized glycosaminoglycans such as heparan sulfate and dermatan sulfate. This accumulation causes severe damage to several tissues, principally the central nervous system. Previously, we identified 187 IDS-protein interactions in the mouse brain. To validate a subset of these interactions, we selected and cloned the coding regions of 10 candidate genes to perform a targeted yeast two-hybrid assay. The results allowed the identification of the physical interaction of IDS with LSAMP and SYT1. Although the physiological relevance of these complexes is unknown, recent advances allow us to point out that these interactions could be involved in vesicular trafficking of IDS through the interaction with SYT1, as well as to the ability to form a transcytosis module between the cellular components of the blood-brain-barrier (BBB) through its interaction with LSAMP. These results may shed light on the role of IDS on cellular homeostasis and may also contribute to the understanding of MPS II physiopathology and the development of novel therapeutic strategies to transport recombinant IDS through the brain endothelial cells toward the brain parenchyma.Intranasal drug transport through the olfactory route to the brain is an effective drug route for increased absorption and bioavailability of the drug. The objective of this study was to increase the penetration of valproic acid as an anticonvulsant into a delivery system comprising liposomes. Valproic acid liposomes were prepared by a thin-layer hydration technique using soybean phosphatidylcholine and cholesterol as the main ingredients. The formulations were evaluated for diameter size, entrapment efficiency (EE), zeta potential, polydispersity index, and morphology. ex vivo permeation using sheep nasal mucosa and in vivo efficacy were assessed by performing a pharmacokinetic study in Wistar albino rats following intranasal administration of the formulations in comparison with pure drug. The mean size particle of optimized liposomes ranged from 90 to 210 nm with a low polydispersity index ( less then 0.5). The EE of optimized liposomes was between 60% and 85%, increasing the concentration of phosphatidylcholine added to the formula.
