mRNA expression was assayed by qRT-PCR, and protein expression was determined by Western blot. Our results indicate that CRTC2 is overexpressed in CRC cells. Knockdown of CRTC2 inhibits the proliferation, migration, and invasion of CRC cells. When the phosphorylation of CRTC2 Ser238 decreases due to the lack of ERK2, the phosphorylation of Ser171 site increases. The proliferation, migration and invasion of CRC cells were inhibited, the nuclear aggregation of CRTC2 in the nucleus was reduced, and the interaction between CRTC2 and CREB was weaken. It is shown that the phosphorylation of CRTC2 Ser238 is important for CREB transcriptional activity. These findings may help in the identification of potentially new targets for CRC therapy.In the population of Primary Health Care Nursing (PHC) professionals, the association between Burnout Syndrome (BS) and Metabolic Syndrome (MS) has not been investigated. The objective was to evaluate the association between BS and MS among PHC Nursing Professionals. A cross-sectional, multicenter, population-based study was conducted in the state of Bahia, Brazil, with 1,125 professionals. The prevalence of BS and MS corresponded to 18.3% and 24.4%, respectively. The prevalence in women of BS was 16.4% and of MS 23.7%, in men 31.6% for BS and 29.4% for MS. Men with BS are 3.23 times more likely to develop MS, and women 1.48 times more. BS was associated between men and women and exhibited a good discriminatory predictive power.Chitin synthase (CHS) is one of the crucial enzymes that play an essential role in chitin synthesis during the molting process, and it is considered to be the specific target to control insect pests. Currently, there are no potent inhibitors available in the market, which specifically target this enzyme. Pyrimidine nucleoside peptide, nikkomycin Z, binds to nucleotide-binding sites of fungal and insect CHS. But, their mode of action is still fragmentary due to the lack of a 3Dstructure of CHS. Chilo partellus is a severe pest insect of major food crops such as maize and sorghum, in an attempt to target integument expressed cuticular CpCHS. The CpChsA cDNA was cloned, and subsequently, their developmental and tissue-specific expression was studied. The 3D structure of the CHS catalytic domain was modeled, after which natural compounds were screened using a virtual screening workflow and resulted in the identification of five hit molecules. Molecular dynamics simulations were performed to investigate the dynamics and interactions of hits with CpCHS. The obtained results revealed that the compounds kasugamycin, rutin and robinin could act as potent inhibitors of CpCHS. All three molecules were observed to significantly reduce the chitin production as validated using in vitro and in vivo studies. Thus, this study aims to provide a set of novel inhibitor molecules against CpCHS for controlling the pest population. Communicated by Ramaswamy H. Sarma.The present study explored the links between the quality of adolescents' attachment to their mother and their father, separation anxiety and suicidal ideation. Self-report questionnaires were completed by French adolescents (n = 455; Mage = 15.78 years, SD = 1.61) to probe their attachment styles, separation anxiety (related to school phobia, a secure base, and family members), and suicidal ideation. Avoidantly attached adolescents displayed greater school phobia than securely attached adolescents. Female adolescents with an avoidant attachment to their father tended to report less fear related to family members than their secure counterparts. Male adolescents who were insecurely attached to their mother reported more separation anxiety than securely attached males. The structural models were significant for both boys and girls (R2 = .38 and .44, p less then .001) with good fit indices. Mediation analyses revealed that separation anxiety partially mediated the negative effect of attachment to mother (especially alienation) on suicidal ideation for both girls and boys, and partially mediated the negative effect of attachment to father (especially alienation) on suicidal ideation for boys. These results support an individuated conception of attachment patterns, with a differential influence of dyadic relationships, and could provide a better understanding of adolescents' suicidal ideation.Opportunities for refugees to engage in occupations within host countries can be constrained by factors such as governmental policies or language barriers (Smith). Female refugees with physical disabilities may experience compounding barriers to participation related to their identity markers. The main aim of this study is to understand the experiences of female refugees with physical disabilities in seeking, preparing for, and obtaining employment. This study used an instrumental case study using semistructured interviews with five primary participants and four caregivers. Thematic analysis (Clarke & Braun) identified five key barriers to securing employment (a) stigma and discrimination, (b) restrictive traditional labor market, (c) inaccessible and inadequate housing, (d) lack of cohesion and information across services, and (e) English language predominance. Findings highlight complex challenges experienced by the women who faced intersecting systems of oppression and can enable occupational therapists to address barriers associated with occupational injustice by improving services to better meet their needs.ABSTRACT Glioma is a common type of brain tumour that is associated with neurocognitive and psychosocial impairments. This study primarily aimed to investigate the feasibility and acceptability of a videoconferencing format of the Making Sense of Brain Tumour (Tele-MAST) programme. Recruited from a multidisciplinary brain tumour clinic, participants were randomly allocated to the 10-session Tele-MAST programme or standard care, with the latter group offered Tele-MAST after re-assessment. Semi-structured interviews explored participants' experiences of receiving psychological support via the videoconferencing platform. Measures of mental health and quality of life were administered at pre-intervention and post-intervention assessments. Of the 35 patients deemed eligible for the study, 14 (40% accrual) with high grade (71%) or low grade (29%) glioma participated. KI696 They were randomized to the Tele-MAST condition (n = 8) or standard care/waitlist (n = 6). Ten individuals commenced the Tele-MAST programme, of whom eight completed ≥8 sessions.
