Two months of post-transplant survival were achieved in an end-stage heart disease patient with a porcine heart from a pig whose genome had been modified in 10 of its genes. This first-ever life-saving cardiac xenotransplantation was the result of decades of work and close coordination by researchers in genetic engineering, animal cloning, immunology, ex vivo organ perfusion, and thoracic surgery.The functional groups in porous carbon generally suffer a severe loss during the high-temperature carbonization. Instead, the low-temperature synthesis of carbon featuring porous structures and abundant functional groups is not only a solution that evades the pitfalls of pyrolysis but also is of significance for the development of synthetic methodology. Herein, a liquid metal interfacial engineering strategy is reported for the synthesis of porous carbon using CCl4 as the carbon precursor and sodium-potassium alloy (NaK) as the reducing agent, which is superior to traditional synthetic methods because it enables the engineering of a highly active liquid metal alloy microemulsion to directly generate porous carbon at ambient temperature. As synthesized porous carbon featured abundant carbon-chlorine bonds can be tandem-grafted with imidazole and 1,2-dibromoethane to achieve a CO2 cycloaddition catalyst, which exhibits excellent catalytic activity, in addition to exceptional stability.An analytical formula is presented here for the electrophoresis of a dielectric or perfectly conducting fluid droplet with arbitrary surface potentials suspended in a very dilute electrolyte solution. In other words, when the Debye length (κ-1 ) is very large, or κa ≪ $ll $ 1, where κ is the electrolyte strength and a stands for the droplet radius. This formula can be regarded as an extension of the famous Hückel solution valid for weakly charged rigid particles to arbitrarily charged fluid droplets. The formula reduces successfully to the ones obtained by Booth for a dielectric droplet, and Ohshima for a perfectly conducting droplet, both under Debye-Hückel approximation valid for weakly charged droplets. Moreover, the formula is valid for a gas bubble and a rigid solid particle as well. Classic results obtained by Hückel for a rigid particle are reproduced as well. We found that for a dielectric droplet, the more viscous the droplet is, the faster it moves regardless of its surface potential, contrary to the intuition based on the purely hydrodynamic consideration. For a perfectly conducting liquid droplet, on the other hand, the situation is reversed The less viscous the droplet is, the faster it moves. The presence or absence of the spinning electric driving force tangent to the droplet surface is found to be responsible for it. As a result, an axisymmetric exterior vortex flow surrounding the droplet is always present for a dielectric liquid droplet, and never there for a conducting liquid droplet.
A switch in pollinator can occur when a plant lineage enters a new habitat where the ancestral pollinator is less common, and a novel pollinator is more common. Because pollinator communities vary according to environmental tolerances and availability of resources, there may be consistent associations between pollination mode and specific regions and habitats. Such associations can be studied in lineages that have experienced multiple pollinator transitions, representing evolutionary replicates.
Our study focused on a large clade of Penstemon wildflower species in western North America, which has repeatedly evolved hummingbird-adapted flowers from ancestral bee-adapted flowers. For each species, we estimated geographic ranges from occurrence data and inferred environmental niches from climate, topographical, and soil data. Using a phylogenetic comparative approach, we investigated whether hummingbird-adapted species occupy distinct geographic regions or habitats relative to bee-adapted species.
Hummingbons and habitats associated with novel pollinator communities.Interleukin-4 (IL-4) is a signature cytokine pivotal in Type 2 helper T cell (Th2) immune response, particularly in allergy and hypersensitivity. Interestingly, IL-4 increases endogenous levels of prostaglandin D2 (PGD2 ) and its metabolites, Δ12 -prostaglandin J2 (Δ12 -PGJ2 ) and 15-deoxy-Δ12,14 -prostaglandin J2 (15d-PGJ2 ), collectively called cyclopentenone PGs (CyPGs). However, the therapeutic role of IL-4 in hematologic malignancies remains unclear. Here, we employed a murine model of acute myeloid leukemia (AML), where human MLL-AF9 fusion oncoprotein was expressed in hematopoietic progenitor cells, to test the effect of IL-4 treatment in vivo. Daily intraperitoneal treatment with IL-4 at 60 µg/kg/d significantly alleviated the severity of AML, as seen by decreased leukemia-initiating cells (LICs). The effect of IL-4 was mediated, in part, by the enhanced expression of hematopoietic- PGD2 synthase (H-PGDS) to effect endogenous production of CyPGs, through autocrine and paracrine signaling mechanisms. Similar results were seen with patient-derived AML cells cultured ex vivo with IL-4. Use of GW9662, a peroxisome proliferator-activated receptor gamma (PPARγ) antagonist, suggested endogenous CyPGs-PPARγ axis mediated p53-dependent apoptosis of LICs by IL-4. Taken together, our results reveal a beneficial role of IL-4 treatment in AML suggesting a potential therapeutic regimen worthy of clinical trials in patients with AML.
Adherence to evidence-based guidelines in gastric cancer is low. We aimed to evaluate adherence to National Comprehensive Cancer Network (NCCN) Guidelines for gastric cancer at both patient- and hospital-levels and examine associations between guideline adherence and treatment outcomes, including overall survival (OS).
We applied stage-specific, annual NCCN Guidelines (2004-2015) to patients with gastric cancer treated with curative-intent within the National Cancer Database and compared characteristics of patients who did and did not receive guideline-adherent care. Hospitals were evaluated by guideline adherence rate. We identified associations with OS through multivariable Cox regression.
Of 37 659 patients included, 32% received NCCN Guideline-adherent treatment. OS was significantly associated with both guideline adherence (51 months for patients receiving guideline-adherent treatment vs. 22 for patients receiving nonadherent treatment, p < 0.001). Treatment at a hospital with higher adherence was associated with longer OS (21 months for patients treated at lowest adherence quartile hospitals vs. 37 months at highest adherence quartile hospitals, p < 0.001), regardless of type of treatment received.
Guideline-adherent treatment was strongly associated with longer median OS. Guideline adherence should be used as a benchmark for focused quality improvement for physicians taking care of patients with gastric cancer and institutions at large.

Macrophage-expressed gene 1 (Mpeg1), a pore-forming protein sometimes known as perforin-2, is reported to be essential for bacterial killing following phagocytosis. Mice homozygous for the mutant allele Mpeg1tm1Pod succumb to bacterial infection and exhibit deficiencies in bacterial killing in vitro. Here we describe a new Mpeg mutant allele Mpeg1tm1.1Pib on the C57BL/6J background. Mice homozygous for the new allele are not abnormally susceptible to bacterial or viral infection, and irrespective of genetic background show no perturbation in bacterial killing in vitro. Potential reasons for these conflicting findings are discussed. In further work, we show that cytokine responses to inflammatory mediators, as well as antibody generation, are also normal in Mpeg1tm1.1Pib/tm1.1Pib mice. We also show that Mpeg1 is localized to a CD68-positive endolysosomal compartment, and that it exists predominantly as a processed, two-chain disulfide-linked molecule. It is abundant in conventional dendritic cells 1, and mice lacking Mpeg1 do not present the model antigen ovalbumin efficiently. We conclude that Mpeg1 is not essential for innate antibacterial protection or antiviral immunity, but may play a focused role early in the adaptive immune response.

Homozygous familial hypercholesterolemia (HoFH) is a rare, life-threatening, inherited condition characterized by extremely elevated levels of low-density lipoprotein cholesterol (LDL-C). Patients are at high risk of atherosclerotic cardiovascular disease, adverse cardiovascular events, and associated early mortality. Liver transplant is sometimes used with curative intent. PLX-4720 The objective of the current case series was to evaluate the follow-up of a range of patients who have undergone liver transplant for the treatment of HoFH.
Patients with clinical and/or genetic diagnoses of HoFH were treated according to local practices in four units in Europe and the Middle East. All patients underwent liver transplantation. Baseline and long-term follow-up data were collected, including LDL-C levels, DNA mutations, lipid-lowering medications, and complications due to surgery and immunosuppressive therapy.
Nine patients were included with up to 22years' follow-up (mean ± SD 11.7 ± 11.7years; range 0.5-28years). Three of the patients died as a result of complications of transplant surgery (mortality rate 33%).
