Sphingosine-1-phosphate (S1P) is a unique lipid ligand binding to S1P receptors to transduce various cell survival or proliferation signals via small G proteins. S1P lyase (S1PL) is the specific enzyme that degrades S1P to phosphoethanolamine and (2E)-hexadecenal and therefore regulates S1P levels. S1PL also degrades dihydrosphingosine-1-phosphate (Sa1P), with a higher affinity to produce hexadecanal. Here, we developed a newly designed assay using a C17-Sa1P substrate that degrades into pentadecanal and phosphoethanolamine. For higher sensitivity in pentadecanal analysis, we developed a quantitative protocol as well as a 5,5-dimethyl cyclohexanedione (5,5-dimethyl CHD) derivatization method. The derivatization conditions were optimized for the reaction time, temperature, and concentrations of the 5,5-dimethyl CHD reagent, acetic acid, and ammonium acetate. The S1PL reaction in the cell lysate after spiking 20 µM of C17-Sa1P for 20 min was linear to the total protein concentrations of 50 µg. The S1PL levels (4 pmol/mg/min) were readily detected in this HPLC with fluorescence detection (λex = 366 nm, λem = 455 nm). The S1PL-catalyzed reaction was linear over 30 min and yielded a Km value of 2.68 μM for C17-Sa1P. This new method was validated to measure the S1PL activity of mouse embryonal carcinoma cell lines of the standard cell (F9-0), S1PL knockdown cells (F9-2), and S1PL-overexpressed cells (F9-4). Furthermore, we treated F9-4 cells with different S1PL inhibitors such as FTY720, 4-deoxypyridoxine (DOP), and the deletion of pyridoxal-5-phosphate (P5P), an essential cofactor for S1PL activity, and observed a significant decrease in pentadecanal relative to the untreated cells. In conclusion, we developed a highly sensitive S1PL assay using a C17-Sa1P substrate for pentadecanal quantification for application in the characterization of S1PL activity in vitro.Glioblastoma (GB) (grade IV astrocytoma) is the most malignant type of primary brain tumor with a 16 months median survival time following diagnosis. Despite increasing attention regarding the development of targeted therapies for GB that resulted in around 450 clinical trials currently undergoing, radiotherapy still remains the most clinically effective treatment for these patients. Nevertheless, radiotherapy resistance (radioresistance) is commonly observed in GB patients leading to tumor recurrence and eventually patient death. It is therefore essential to unravel the molecular mechanisms underpinning GB cell radioresistance in order to develop novel strategies and combinational therapies focused on enhancing tumor cell sensitivity to radiotherapy. The subjects of this study were 41 consecutive patients who underwent video-assisted lung lobectomy for lung cancer. Rehabilitation was performed both before and after surgery. Lung function testing, physical function testing (timed up and go test (TUG) and the 30-s chair-stand test (CS-30)), and QOL (EORTC QLQ-C30) were measured before and 1 week after surgery.
Postoperative VC recovered to 76.3% ± 15.6% 1 week after surgery. TUG, CS-30, and QOL were significantly worse after surgery (
< 0.05). Lung function and physical function were found to affect QOL. Postoperative complications included pneumonia in 1 patient. There were no patients who discontinued rehabilitation.

Compared with the cloud computing platform, VEC has several new features, such as the higher network bandwidth and the lower transmission delay. Recently, vehicular computation-intensive task offloading has become a new research field for the vehicular edge computing networks. However, dynamic network topology and the bursty computation tasks offloading, which causes to the computation load unbalancing for the VEC networking. To solve this issue, this paper proposed an optimal control-based computing task scheduling algorithm. Then, we introduce software defined networking/OpenFlow framework to build a software-defined vehicular edge networking structure. The proposed algorithm can obtain global optimum results and achieve the load-balancing by the virtue of the global load status information. Besides, the proposed algorithm has strong adaptiveness in dynamic network environments by automatic parameter tuning. Experimental results show that the proposed algorithm can effectively improve the utilization of computation resources and meet the requirements of computation and transmission delay for various vehicular tasks.Laser additive manufacturing is a promising technique for the preparation of complex-shaped SiC composites. In this work, core-shell Cf @phenolic resin (PR) composites for selective laser sintering of carbon fiber reinforced silicon carbide (Cf/SiC) composites were fabricated by surface modification using 3-aminopropyltriethoxy silane coupling agent (KH550) in combination with planetary ball milling. PR coated uniformly on the fiber surface to form a core-shell structure. At 1 and 7 days post-inoculation, the expression profiles of eight genes related to immunity, transport of nutrients in the intestine, and metabolism were examined. Cecal microbiome composition and blood biochemical/immunological indices were also explored and egg production traits recorded. As a result, the SE challenge of laying hens and Intebio® administration had either a suppressive or activating effect on the expression level of the studied genes (e.g., IL6 and BPIFB3), the latter echoing mammalian/human tissue-specific expression. Within this review, we present a brief discussion of the fibrinolytic pathway, its components, and their contribution to injury progression. We review how local derangements of fibrinolysis, resulting from increased coagulation and reduced plasminogen activator activity, promote extravascular fibrin deposition. Further, we describe how pleural mesothelial cells contribute to lung scarring via the acquisition of a profibrotic phenotype. We also discuss soluble uPAR, a recently identified biomarker of pleural injury, and its diagnostic value in the grading of pleural effusions. Finally, we provide an in-depth discussion on the clinical importance of single-chain urokinase plasminogen activator (uPA) for the treatment of loculated pleural collections.A new optical fiber sensor based on the fluorescence lifetime was prepared for specific detection of sulfate ion concentration, where 1,1'-(anthracene-9,10-diylbis(methylene))bis(3-(dodecylcarbamoyl)pyridin-1-ium) acted as the sulfate fluorescent probe. The probe was immobilized in a porous cellulose acetate membrane to form the sensitive membrane by the immersion precipitation method, and polyethylene glycol 400 acted as a porogen. The sensing principle was proven, as a sulfate ion could form a complex with the probe through a hydrogen bond, which led to structural changes and fluorescence for the probe. The signals of the fluorescence lifetime data were collected by the lock-in amplifier and converted into the phase delay to realize the detection of sulfate ions. Based on the phase-modulated fluorometry, the relationship between the phase delay of the probe and the sulfate ion concentration was described in the range from 2 to 10 mM. The specificity and response time of this optical fiber sensor were also researched.
