The aim of this study was to investigate the potential role of transient receptor potential vanilloid 6 (TRPV6) in E2-mediated osteoclast function and to characterize the relevant underlying mechanisms. Here, we found that Trpv6 is drastically decreased in ovariectomy operation animals and the administration of E2 results in an increased expression of Trpv6 in osteoclasts. In contrast, Trpv6 depletion significantly blocked the inhibitory effects of E2 on bone resorption activity, and silencing Trpv6 alleviated E2-induced osteoclast apoptosis. In addition, we found that E2 regulates the transcription of Trpv6 through ERα, by interacting with C/EBPβ and NF-κB. Chip assay analysis indicated that C/EBPβ regulates Trpv6 transcription by binding to Trpv6 promoter fragments -1,866 nt to -1,761 nt and -2,685 nt to -2,580 nt, whereas NF-κB binds to the -953 nt to -851 nt region. We conclude that TRPV6 has a significant effect on E2-mediated osteoclast function.The only representative of the MYC superfamily transcription factors in C. elegans, MML-1 (Myc and Mondo-like 1), was shown to promote extended lifespan in a variety of models and to regulate some aspects of C. elegans development. This previous research did not involve molecular characterization of MML-1. Here we use available mml-1 mutant alleles and other reagents to demonstrate that MML-1 is modified by O-GlcNAc, binds to promoters of some genes directly regulated by the DOT-1.1 histone methyltransferase complex, and has a role in promoting neuronal migration. Surprisingly, we found that the deletion allele mml-1(ok849), which was considered a null, produces an internally truncated protein resulting from an in-frame deletion. Localization of this truncated product to MML-1 target promoters was not impaired. The deleted region of MML-1 is proline-rich, and its function is poorly understood in mammalian homologs of MML-1. This study aims to develop a screening tool that will help first responders identify patients with proximal femur fractures, commonly referred to as hip fractures, on site and direct these patients to hospitals with orthopaedic surgery services.

Literature and expert opinion defined parameters for the Collingwood Hip Fracture Rule (CHFR) which predict a patient's likelihood of hip fracture. The study population included adults presenting to Collingwood General and Marine Hospital with lower extremity injuries between December 1, 2019 and March 10, 2020. Excluded patients had previous hip replacement, previous hip fracture on the side of the injury, or a high energy mechanism of injury. Patients were assessed with the CHFR before receiving x-ray imaging. The parameters were scored based on their predictive powers and analyzed by a receiver operating characteristic curve.

The study included 101 patients (mean age 66.3 years), and 25.7% had a hip fracture confirmed on imaging. Wearable surface electromyography (sEMG) sensors have been used to track recovery in research; however, sEMG is rarely used within acute and subacute clinical settings. link2 The purpose of this case study was to describe the use of wireless sEMG sensors to examine changes in muscle activity during acute and subacute phases of stroke recovery, and understand the participant's perceptions of sEMG monitoring. Beginning three days post-stroke, one stroke survivor wore five wireless sEMG sensors on his involved arm for three to four hours, every one to three days. Muscle activity was tracked during routine care in the acute setting through discharge from inpatient rehabilitation. link3 Three- and eight-month follow-up sessions were completed in the community. Activity logs were completed each session, and a semi-structured interview occurred at the final session. The longitudinal monitoring of muscle and movement recovery in the clinic and community was feasible using sEMG sensors. The participant and medical team felt monitoring was unobtrusive, interesting, and motivating for recovery, but desired greater in-session feedback to inform rehabilitation. While barriers in equipment and signal quality still exist, capitalizing on wearable sensing technology in the clinic holds promise for enabling personalized stroke recovery.
This consensus aims to clarify the role of Dipeptidyl Peptidase-4 inhibitors (iDPP-4) in managing patients with diabetes during the COVID-19 pandemic.

A PubMed bibliographic search was carried out (December 2019-February 2021). Oxford methodology was used for the evaluation of evidence and possible recommendations were established by consensus.

Diabetes appears to be an independent factor in COVID-19 disease (evidence 2b). No increased risk of contagion with iDPP-4 is demonstrated (evidence 2b), and its use has been shown to be safe (evidence 2b). The use of this drug may present a specific benefit in reducing mortality, particularly in in-hospital use (evidence 2a), reducing admission to intensive care units (evidence 2b) and the need for mechanical ventilation (evidence 2b).

The rise of platforms dependent on largely expendable labour relations, with significant migrant involvement, must be seen as connected, and as replicating larger social processes rather than merely technological changes. This perspective paper urgently calls for an intersectional perspective to better understand social-technical relations crossing the digital-urban interface of platform urbanism in contemporary European cities. Critics of platforms and gig work, to date, have mainly focused on algorithms-based social control, degraded working conditions, problematic employment relations and precariousness of gig work. The ongoing Covid-19 pandemic has both disrupted and amplified these issues, intensifying the vulnerability of gig workers. For example, in Sweden, migrant groups and gig workers were separately identified as being hardest hit by Covid, but with little attention to the interconnectivity between these categories, nor to how these groups are co-positioned vis-a-vis larger socio-economic inequalities. Thus, we argue for a deeper understanding of the social processes underlying platforms and for active investigation of how inequalities are being produced and/or maintained in/by these processes. Urban planners, designers and policy makers will need to actively address the hybrid (digital and physical) urban spaces produced in platform urbanism in order to prevent spatial and economic inequalities. Cities produce less greenhouse gas emissions per capita than suburbs. Meanwhile, the US and the world have been wrestling with the public health emergency of the COVID-19 pandemic. But as US cities recover from the pandemic, they are unlikely to add more population than their suburbs because of changes in favor of working from home, online shopping, and the search for more affordable housing and green space, as well as concerns about population density and contagious diseases. So, the challenge is how to design and redevelop suburbs to make them more environmentally, socially, and economically sustainable to address both climate change and the threat of future pandemics. A sustainable suburb scenario offers an alternative to the sprawling development, separation of land uses and income classes, and automobile dependence that characterizes the typical American suburban landscape. This essay reviews the literature on re-designing suburbs and describes and evaluates both a business-as-usual suburb scenario and a sustainable suburb scenario. Though challenges exist, sustainable suburbs will be needed in order to reduce US greenhouse gas emissions, to achieve greater resilience in adapting to the effects of climate change, and to guard against future pandemics while providing economic opportunities and greater equity over the long run.In Africa, climate change impacts including, but not limited to, erratic rainfall and prolonged droughts are already affecting farmers' productivity and disrupting households' livelihoods. Following this realization are recommendations for implementing climate-smart agriculture (CSA) as adaptation and resilience pathways to address the negative ramifications of climate change impacts. While CSA mainstreaming is strong at the global and national levels, it remains a challenge at the local level. To understand CSA mainstreaming at the local level, this paper utilizes mixed-content analysis to deconstruct eleven local development plans for the 2018-2021 plan period for the Upper West Region, a semi-arid region of Ghana. Results show that CSA mainstreaming is a challenge, despite a general awareness of climate change impacts on agriculture. The plans lacked adequate data on local climate change trends and impacts leading to discrepancies among CSA problematization, development goals, objectives, and strategies-raising serious concerns about ownership and localization of CSA in semi-arid Ghana.
