Although, to date, specific molecular mechanisms have not been elucidated, at the cellular level, it is mainly due to pathological changes in the life process of nucleus pulposus (NP) cells in the intervertebral disc (IVD). These changes are closely related to cell proliferation, apoptosis, senescence, autophagy, inflammation, and extracellular matrix (ECM) remodeling. Long noncoding RNAs (lncRNAs) have gradually become a focus of scientific research because of their functional complexity and local tissue specific expression. Moreover, they mediate a series of cellular signaling pathways in NP cells by competing for microRNA (miRNA) or directly targeting gene expression by mRNA adsorption, thereby regulating cell life activities that play a vital role in the mechanism underlying IDD. In-depth studies on lncRNAs can help identify new therapeutic targets or aid in developing IDD treatment strategies at the gene level and those based on regenerative medicine, thus providing new ideas for researchers. This article reviews the classification, biological functions, mechanisms of action, and therapeutic potential of lncRNAs in IDD.
This study aimed to investigate the clinicopathological and molecular characteristics of ELOC(TCEB1)-mutant renal cell carcinoma.
Sanger sequencing was used to assess 32 cases originally diagnosed as clear cell renal cell carcinoma with CK7 positive and/or fibromyomatous stroma. Of these, 4 patients with ELOC(TCEB1) gene mutation were screened, and their clinicopathological data were collected for histomorphological observation, immunohistochemical staining, and follow-up, and relevant pieces of literature were reviewed.
The 4 patients with ELOC(TCEB1) mutations were all males and aged between 57 and 64 years (median age 59 years old). The tumor was located in the renal cortex, with a diameter of 2-3.5cm. The cross-section was grayish-yellow and grayish brown, solid and nodular, and clearly demarcated from the surrounding tissues. Of the 4 patients, 3 harbored a thick fibrous pseudocapsule rich in smooth muscle and were separated from the surrounding normal renal tissue, and 2 of them showed focal invasSanger sequencing method is helpful for the diagnosis of this type of RCC. Most patients harbor tumors exhibiting low nuclear grade and inert clinical behavior, and a few tumors exhibit high nuclear grade and aggressive characteristics.
Phytotherapeutic modulation of the immune system to mitigate infectious ailments has been in vogue all over the world.
The present work has been designed to scientifically explore the immunomodulatory potential of Nyctanthes arbortristis stem bark using mice models.
Methanolic (MNA) and aqueous (ANA) extracts of N.arbortristis stem bark were evaluated for possible modulation in humoral immunity through serum immunoglobulin estimation. The variation in cellular immunity was assessed using neutrophil adhesion test, carbon clearance assay, and cyclophosphamide-induced neutropenia.
Administration of MNA and ANA (both at 200mg/kg, p.o.) significantly augmented the levels of serum immunoglobulins (humoral antibody), neutrophil adhesion, and phagocytic index (a measure of carbon clearance). Extracts also guarded the animals against cyclophosphamide-induced leukopenia, especially neutropenia.
Results indicate that cellular and humoral immune responses were aroused by pretreatment of the animal with methanol and aqueous extract of N.arbortristis. Thus, the methanol and aqueous extract of N.arbortristis stem bark possesses a significant immunostimulant activity and can be used to uplift the immune system in the infectious condition.

The current study investigated how social and monetary reward anticipation affected cognitive control in 39 children, 40 adolescents, and 40 adults. We found that cognitive control performance improved with age in a Simon task, and the reaction time (RT) was modulated by the reward magnitude. The conflict monitoring process (target N2 amplitudes) of adolescents and the attentional control processes (target P3 amplitudes) of adolescents and adults could be adjusted by reward magnitude, suggesting that adolescents were more sensitive to rewards compared to children. Reward magnitudes influenced the neural process of attentional control with larger P3 in congruent trails than that in incongruent trials only in low reward condition. The result of hierarchical drift-diffusion model indicated that children had slower drift rates, higher decision threshold, and longer non-decision time than adolescents and adults. Adolescents had faster drift rates in monetary task than in social task under the high reward condition, and they had faster drift rates under high reward condition than no reward condition only in the monetary task. The correlation analysis further showed that adults' non-decision time and decision threshold correlated with conflict monitoring process (N2 responses) and attentional control process on conflicts (P3 responses). Adolescents' drift rates associated with neural process of attentional control. The current study reveals that reward magnitude and reward type can modulate cognitive control process, especially in adolescents.

This study adopted a qualitative design. Twentysix final (6th) semester nursing students were recruited through purposive sampling at a Swedish university. Data were collected through semi-structured, face-to-face, focus group interviews in September 2015. The transcribed interviews were analyzed using qualitative content analysis.

Nursing students adapt to the prevailing venous blood sample collection practice culture and become copycats.The aim of this study was to evaluate the topography and the fatigue performance of lithium disilicate glass-ceramic after surface grinding through different laboratory protocols used to simulate the Computer-aided design/Computer-aided manufacturing (CAD/CAM) milling. Ceramic discs (IPS e.max CAD, Ø = 13.5 mm × 1.2 mm of thickness) were produced through different methodologies milling in CAD/CAM system (CAD/CAM group); produced in-lab with a polished surface (POL group); or produced through in-lab methods and randomly distributed into five groups according to different grinding protocols to simulate the CAD/CAM milling [grinding with a CAD/CAM bur coupled to a mandrel (CAD/CAM Bur group); fine diamond bur using oscillatory movements (DBO group); fine diamond bur in x and y axes of the disc (DBXY group); #60-grit silicon carbide sandpaper (SiC group); and #60-grit wood sandpaper (WS group)]. The specimens were fatigue tested (n = 15) according to the step-stress method (initial load 60 N; step-size 20 N; 10,000 cycles/step; 20 Hz frequency). A roughness analysis was performed on all specimens, while fractal dimension (FD) and fractography were performed on representative samples. The Kaplan-Meier analysis showed that the POL (293.3 N) group presented better fatigue performance (higher load and number of cycles for failure) (p 0.05). The surface created by CAD/CAM milling and POL group exhibited more complexity (FD) higher values than the experimental groups. For the ceramic surface topography images, the CAD/CAM milling visibly produced a uniform surface compared to the other groups; however, the POL group was the smoothest. The DBO, DBXY, SiC, and WS groups resulted in similar characteristics of surface topography. Therefore, although the SiC and WS groups showed similar roughness to the control group (CAD/CAM), no in-lab simulation method was fully capable to mimic the mechanical performance of the CAD/CAM-milled lithium disilicate glass-ceramic.Metal deficiencies in synthetic hydroxyapatite (Hap), is one factor among others behind its poor mechanical reliability. Hence, compensating these deficiencies has been suggested as a possible way to improve these properties. Several studies have shown that the incorporation of metals or metal oxides appear to increase the fracture toughness and lower the brittleness of synthetic Hap. In this study, we examined the resultant effect of two different methods of incorporating metals into the Hap structure on its mechanical properties. The first method consisted of incorporating metals by replacing some calcium in the Hap structure via substitution. The second method consisted of modifying Hap by preparing (metal oxide)/Hap composites. Thus Nano powders of pure Hap, substituted Hap (Ca10-xMx(PO4)6(OH)2), and (MO)x /Hap composites (M = Mg, Zn) with x = 0.0, 0.1, and 0.3 were prepared by a simple low temperature technique. The (ZnO)x/Hap nanocomposites were prepared by precipitating Zinc oxide nanoparticles in a nano Hap slurry, while the (MgO)x/Hap composites were prepared by precipitating Hap nanoparticles in a diluted nano MgO slurry. XRD, FTIR, and SEM analysis of all prepared materials identified Hap as the only crystalline phase present, exhibiting a uniform morphology with particles in the 40-100 nm size range. EDAX analysis clearly shows the presence of Ca, P, and O in pure hydroxyapatite, Ca, P, O, Mg and Zn in both substituted Hap and (MO)x/Hap composites. The mechanical testing consisted of evaluating Breaking force, work of fracture, and brittleness/ductility of Hap, substituted Hap, and (MO)x/Hap composites. Our study clearly shows that the mechanical properties of Hap are much more improved by modifying Hap with metal oxides than by calcium substitution for the same quantity and type of incorporated metal. Optimum mechanical strength was obtained for the (MO)x/Hap composites with a 0.11.0 MOHap mole ratio (M = Mg, Zn).
