Lowest values of MDA (348.5 nmol g-1 FW) and H2O2 (3539.4 mmol/L) were observed in 7.70 g Si/kg soil followed by in 3.85 g Si/kg soil (MDA 392.6 nmol g-1 FW and H2O2 3134.6 mmol/L) than that of the control. Whereas, PAL enzyme activity (50.8 mmol/L), JA (230.2 mmol/L) and SA (2.7 ug mL-1) contents were significantly higher in 7.70 g Si/kg soil followed by in 3.85 g Si/kg soil (PAL 46.3 mmol/L, JA 182.7 mmol/L and SA 2.4 ug mL-1) as compared to control. The lower MDA, H2O2 level and higher enzymatic activities were associated with the highest expression levels of their metabolic pathway associated genes i.e., ShMAPK1, ShLOX, ShPAL, ShAOS, ShAOC, ShC4H, ShCAT, Sh4CL and ShNPR1 (22.08, 15.56, 10.42, 3.35, 2.54, 2.14, 1.82, 1.67 and 1.22 folds, respectively) in 7.70 g Si/kg soil as compared to other experimental units and control. In the present research, 4-weeks old healthy C57BL/6 mice were randomly assigned into four experimental groups (n = 12), viz., CON, DON 3 mg/kg BW, Rb1 50 mg/kg BW and DON 3 mg/kg + Rb1 50 mg/kg BW (DON + Rb1). Feed intake and body weight gain were monitored during the entire experiment (15 d). Our results demonstrated that Rb1 markedly increased the ADG (30%) and ADFI (25.10%) of mice compared with DON group. Furthermore, Rb1 alleviated the DON-induced immune injury by relieving the splenic histopathological alteration, enhancing the T-lymphocytes subsets (CD4+, CD8+), the levels of cytokines (IL-2, IL-6, IFN-γ, and TNF-α), as well as production of immunoglobulins (IgA, IgM, and IgG). Moreover, Rb1 ameliorated DON-inflicted oxidative stress by reducing the ROS, MDA and H2O2 contents and boosting the antioxidant defense system (T-AOC, T-SOD, CAT, and GSH-Px). Additionally, Rb1 significantly reversed the DON-induced excessive splenic apoptosis via modulating the mitochondria-mediated apoptosis pathway in mice, depicting the decreased percentage of splenocyte apoptotic cells by 26.65%, down-regulated the mRNA abundance of Bax, caspase-3, caspase-9, and protein expression of Bax, cleaved caspase-3, and Cyt-c. The results showed that the application of BCM significantly increased the soil fertility, the biomass of wheat seedling roots increased by more than 50%, and soil dehydrogenase (DHA) and catalase (CAT) activities increased by 369.23% and 12.61%, respectively. In addition, with the application of BCM, the diethylenetriaminepentaacetic acid extractable (DTPA-extractable) Cd, Pb, Cu and Zn in soil were reduced from 2.34 to 0.38 mg/kg, from 49.27 to 25.65 mg/kg, from 3.55 mg/kg to below the detection limit and from 4.05 to 3.55 mg/kg, respectively. Correspondingly, the uptake of these metals in wheat roots and shoots decreased by 62.43% and 79.83% for Cd, 73.21% and 66.32% for Pb, 57.98% and 68.92% for Cu, and 40.42% and 43.66% for Zn. Furthermore, BCM application decreased the abundance and alpha diversity of soil bacteria and changed the soil bacterial community structure dramatically.
