The present study investigates the insecticidal effect of plant extract such as Mentha pulegium and Rosmarinus officinalis essential oils and some of their major compounds; these plants are well known for their many biological activities. The fumigant toxicity was evaluated, using glass jars, against female adults of Culex pipiens that constitute a mosquito vector of important diseases such as the West Nile virus. The adulticidal test showed that both essential oils and monoterpenes presented an insecticidal effect better than the chemical insecticide (Deltamethrin). The highest mortality percentages for the two essential oils have occurred at 312.5 µL/L air (between 56.14 ± 1.7% and 97.71 ± 3.03% after 24 h and 48 h of treatment). Moreover, all tested monoterpenes (carvone, R(+)-pulegone, 1,8-cineole, camphor and α-pinene) have produced high mortalities that varied depending on the time of the treatment and the concentrations used. Lethal concentrations (LC50) obtained for the essential oils and the main compounds have also varied according to the exposure time. M. pulegium and R. officinalis essential oil exhibited the lowest LC50 values after 24 h (72.94 and 222.82 µL/L air, respectively) and after 48 h (25.43 and 55.79 µL/L air, respectively) while the pure molecules revealed the lowest LC50 values after 48 h (between 84.96 and 578.84 µL/L air). This finding proves that the two essential oils and their main compounds have an insecticidal potential, which could help to develop natural toxic fumigants that may be used as an eco-friendly alternative in integrated and sustainable vector management.Sesquiterpene lactone (STL) and natural rubber (NR) are characteristic isoprenoids in lettuce (Lactuca sativa). Both STL and NR co-accumulate in laticifers, pipe-like structures located along the vasculature. NR-biosynthetic genes are exclusively expressed in laticifers, but cell-type specific expression of STL-biosynthetic genes has not been studied. Here, we examined the expression pattern of germacrene A synthase (LsGAS), which catalyzes the first step in STL biosynthesis in lettuce. VH298 solubility dmso Quantitative PCR and Illumina read mapping revealed that the transcripts of two GAS isoforms (LsGAS1/LsGAS2) are expressed two orders of magnitude (~100-200) higher in stems than laticifers. This result implies that the cellular site for LsGAS1/2 expression is not in laticifers. To gain more insights, promoters of LsGAS1/2 were cloned and fused to β-glucuronidase (GUS), followed by transformations of lettuce with these promoter-GUS constructs. In in situ GUS assays, the GUS expression driven by the LsGAS1/2 promoters was tightly associated with vascular bundles. High-resolution microsections showed that GUS signals are not present in laticifers but are detected in the vascular parenchyma cells neighboring the laticifers. These results suggest that expression of LsGAS1/2 occurs in the parenchyma cells neighboring laticifers, while the resulting STL metabolites accumulate in laticifers. It can be inferred that active metabolite-trafficking occurs from the parenchyma cells to laticifers in lettuce.Unlike typical negative gravitropic curvature, young hypocotyls of Brassica rapa and other dicots exhibit positive gravitropism. This positive curvature occurs at the base of the hypocotyl and is followed by the typical negative gravity-induced curvature. We investigated the role of auxin in both positive and negative hypocotyl curvature by examining the transcription of PIN1, PIN3, IAA5 and ARG1 in curving tissue. We compared tissue extraction of the convex and concave flank with Solid Phase Gene Extraction (SPGE). Based on Ubiquitin1 (UBQ1) as a reference gene, the log (2) fold change of all examined genes was determined. Transcription of the examined genes varied during the graviresponse suggesting that these genes affect differential elongation. The transcription of all genes was upregulated in the lower flank and downregulated in the upper flank during the initial downward curving period. After 48 h, the transcription profile reversed, suggesting that the ensuing negative gravicurvature is controlled by the same genes as the positive gravicurvature. High-spatial resolution profiling using SPGE revealed that the transcription profile of the examined genes was spatially distinct within the curving tissue. The comparison of the hypocotyl transcription profile with the root tip indicated that the tip tissue is a suitable reference for curving hypocotyls and that root and hypocotyl curvature are controlled by the same physiological processes.Arabinogalactan proteins (AGPs) are members of a family of proteins that play important roles in cell wall dynamics. AGPs from inclined pines were determined using JIM7, LM2, and LM6 antibodies, showing a higher concentration in one side of the stem. The accumulation of AGPs in xylem and cell wall tissues is enhanced in response to loss of tree stem verticality. The differential gene expression of AGPs indicates that these proteins could be involved in the early response to inclination and also trigger signals such as lignin accumulation, as well as thicken cell wall and lamella media to restore stem vertical growth. A subfamily member of AGPs, which is Fasciclin-like has been described in angiosperm species as inducing tension wood and in some gymnosperms. A search for gene sequences of this subfamily was performed on an RNA-seq library, where 12 sequences were identified containing one or two fasciclin I domains (FAS), named PrFLA1 to PrFLA12. Four of these sequences were phylogenetically classified in group A, where PrFLA1 and PrFLA4 are differentially expressed in tilted pine trees.This paper reviews the history of herbicide-resistant (HR) traits in U.S. cotton since the beginning, highlighting the shortcomings of each trait over time that has led to the development of their successor and emphasizing the importance of integrated weed management (IWM) going forward to ensure their long-term sustainability. Introduction of glyphosate-resistant cropping systems has allowed for expansion of no-till systems more reliant on herbicides, favored less diverse crop rotations, and heavily relied on a single herbicide mode of action (MOA). With repeated applications of glyphosate over the years, biotypes of glyphosate-resistant (GR) A. palmeri and other weeds became economically damaging pests in cotton production systems throughout the U.S. Moreover, the reported cases of weeds resistant to different MOA across various parts of the United States has increased. The dicamba- (XtendFlex®) and 2,4-D-resistant (Enlist®) cotton traits (with stacks of glyphosate and glufosinate resistance) were introduced and have been highly adopted in the U.S. to manage HR weeds. Given the current rate of novel herbicide MOA discovery and increase in new HR weed cases, the future of sustainable weed management relies on an integrated approach that includes non-herbicidal methods with herbicides to ensure long-term success.The aim of this study was to extract and identify the chemical compounds of Diplosthephium juniperinum essential oil (EO) from Ecuador and to assess its anticholinesterase and antioxidant properties. The EO chemical composition was determined by GC-MS. A total of 74 constituents of EO were identified, representing 97.27% in DB-5ms and 96.06% in HP-INNOWax of the total EO. The major constituents (>4.50%) identified were α-pinene (21.52, 22.04%), geranyl acetate (10.54, 7.78%), silphiper-fol-5-ene (8.67, 7.38%), α-copaene (8.26, 8.18%), 7-epi-silphiperfol-5-ene (4.93, 5.95%), and germacrene D (4.91, 6.00%). Enantioselective analysis of the volatile fraction of D. juniperinum showed (+)-α-pinene as a pure enantiomer and 5 pairs of enantiomeric compounds. Among them, (-)-β-Pinene and (-)-Germacrene D presented a high enantiomeric excess of 93.23 and 84.62%, respectively, while (-)-α-Thujene, (-)-Sabinene and (S)-4-Terpineol with a lower enantiomeric excess of 56.34, 47.84 and 43.11%, respectively. A moderate inhibitory effect was observed for Acetylcholinesterase (AChE) and Butyrylcholinesterase (BuChE) enzymes with IC50 values of 67.20 ± 7.10 and 89.00 ± 9.90 µg/mL, respectively. A lower antioxidant potential was observed for the EO measured through DPPH and ABTS radical scavenging assays with SC50 values of 127.03 and >1000 µg/mL, respectively. To the best of our knowledge, this is the first report of the chemical composition, enantiomeric distribution and, anticholinesterase and antioxidant potential of the EO of D. juniperinum. As future perspective, further in-vivo studies could be conducted to confirm the anticholinesterase potential of the EO.Developing high-yielding maize hybrids is essential under the fast-growing global population and abrupt global climate change. Planting density is one of the imperative components for enhancing maize productivity. This study assessed newly developed maize hybrids under three planting densities on two sowing dates. The evaluated hybrids were 40 maize genotypes comprised of 36 F1-developed hybrids and 4 commercial high-yielding check hybrids. The developed hybrids were generated from selected maize inbred lines according to their adaptive traits to high planting density, such as prolificacy, erect leaves, short plants, early silking, anthesis-silking interval, and small tassel size. The applied planting densities were high, intermediate, and low, with 95,000, 75,000, and 55,000 plants/ha, respectively, under timely and late sowing. The high planting density displayed the uppermost grain yield compared with the intermediate and low densities at both sowing dates. The developed hybrid G36 exhibited the highest agronomic performance under high planting density at timely and late sowing. Additionally, G38, G16, G37, G23, G5, G31, G18, G7, G2, G20, G29, and G17 displayed high agronomic traits at both sowing dates. Joint regression and AMMI analyses revealed significant genotype, agro-environment, and genotype × agro-environment interaction effects for grain yield. The AMMI biplot displayed that G39 was closest to the ideal stable hybrid, and the hybrids G36, G18, G38, G17, G2, and G37 were considered desirable stable hybrids. Moreover, the GGE biplot indicated that a high planting density at an optimal sowing date could be considered a representative environment for discriminating high-yielding maize hybrids. The designated promising hybrids are recommended for further inclusion in maize breeding due to their stability and high yields.In tropical forests of southern Ecuador, artisanal gold mining releases heavy metals that become xenobiotic with indefinite circulation and eventual bioaccumulation. Restoration and rehabilitation of degraded mining sites represent a major ecological, technological and economic issue. In this study, we estimate the capacity of two native woody plants to accumulate cadmium (Cd), lead (Pb), zinc (Zn) and mercury (Hg), with the goal of developing effective strategies for phytoremediation of mining sites. Individuals of Erato polymnioides and Miconia sp., as well as their rhizospheric soils, were sampled from a natural zone (NZ) of montane cloud forest, used as a control, and a polluted zone (PZ) subjected to active gold mining. Concentrations of the four heavy metals were analyzed using atomic absorption spectrophotometry. Cd, Zn and Hg concentrations were higher in soils of PZ than NZ.
