Anthocyanins from Clitoria ternatea were obtained using ultrasound-assisted extraction with glycerol-water (6040 w/v). The anthocyanins in the extract were measured at wavelength 550 and 700 nm using a UV-visible spectrophotometer and expressed in terms of total anthocyanins content (TAC). Taguchi method was employed to optimize the extraction parameters that include the extraction time (30 to 50 min), extraction temperature (40 to 60 °C), and solvent to solid ratio (101 to 301), with TAC as the response. The obtained data showed the optimum extraction parameters as 30 min extraction time, 50 °C extraction temperature, and 101 solvent to solid ratio. The anthocyanin's storage stability was analyzed for 7 days at varying storage temperatures and exposure to light. The present dataset in this article indicated the glycerol-water system as a green alternative for anthocyanin extraction and acted as a storage medium. Furthermore, our methodology can be applied to optimize the anthocyanins extraction process, manipulate storage conditions and improve the extract quality.In this article, we present a multicenter aortic vessel tree database collection, containing 56 aortas and their branches. The datasets have been acquired with computed tomography angiography (CTA) scans and each scan covers the ascending aorta, the aortic arch and its branches into the head/neck area, the thoracic aorta, the abdominal aorta and the lower abdominal aorta with the iliac arteries branching into the legs. For each scan, the collection provides a semi-automatically generated segmentation mask of the aortic vessel tree (ground truth). The scans come from three different collections and various hospitals, having various resolutions, which enables studying the geometry/shape variabilities of human aortas and its branches from different geographic locations. Furthermore, creating a robust statistical model of the shape of human aortic vessel trees, which can be used for various tasks such as the development of fully-automatic segmentation algorithms for new, unseen aortic vessel tree cases, e.g. by training deep learning-based approaches. Hence, the collection can serve as an evaluation set for automatic aortic vessel tree segmentation algorithms.The sago palm (Metroxylon sagu Rottboll) is a tropical halophytic starch-producing, economically important crop palm mainly located in Southeast Asian countries. Recently, a genome survey was conducted on this palm using the Illumina sequencing platform, with a very low (21.5%) BUSCO genome completeness score, and most of them (∼78%) are either fragmented or missing. Thus, in this study, the sago palm genome completeness was further improved with the utilization of the Nanopore sequencing platform that produced longer reads. A hybrid genome assembly was conducted, and the outcome was a much complete sago palm genome with BUSCO completeness achieved at as high as 97.9%, with only ∼2% of them either fragmented or missing. The estimated genome size of the sago palm is 509,812,790 bp in this study. A sum of 33,242 protein-coding genes was revealed from the sago palm genome and around 96.39% of them had been functionally annotated. An investigation on the carbohydrate metabolism KEGG pathways also unearthed that starch synthesis was one of the major sago palm activities. The genome data obtained from this work is indispensable for future molecular evolutionary and genome-wide association studies on the economically important sago palm.The data presented in this article- originally reported by Soto and colleagues (Soto et al., in press)- assess social, emotional, and behavioral (SEB) skills, indexed by the Behavioral, Emotional, and Social Skills Inventory (BESSI), across seven independent samples (N = 6,309). Four of the datasets (N = 5000) were collected using an online survey housed on PersonalityLab.org. In two of these internet datasets, participants provided their responses to sociodemographic items, subsets of BESSI items (45 - 102 items), and the Big Five Inventory-2 (BFI-2, 60 items). In the other two internet datasets, participants answered the same sociodemographic items and the full BESSI questionnaire (192 - 200 items). The fifth dataset was collected via an online survey sponsored by the Character Lab Research Network and included responses from 499 high school students. The High School Student Sample completed sociodemographic items, the full BESSI (192 items), and measures of academic engagement, occupational interests, peerter strengths, and (3) explore the associations between SEB skills and consequential outcomes.The effective use of human-derived cells that are difficult to freeze, such as parenchymal cells and differentiated cells from stem cells, is crucial. A stable supply of damage-sensitive cells, such as differentiated neuronal cells, neurons, and glial cells can contribute considerably to cell therapy. We developed a serum-free freezing solution that is effective for the cryopreservation of differentiated neuronal cells. The quality of the differentiated and undifferentiated SK-N-SH cells was determined based on cell viability, live-cell recovery rate, and morphology of cultured cells, to assess the efficacy of the freezing solutions. The viability and recovery rate of the differentiated SK-N-SH neuronal cells were reduced by approximately 1.5-folds compared to that of the undifferentiated SK-N-SH cells. The viability and recovery rate of the differentiated SK-N-SH cells were remarkably different between the freezing solutions containing 10% DMSO and that containing 10% glycerol. Cryoprotectants such as fetal bovine serum (FBS), antifreeze proteins (sericin), and sugars (maltose), are essential for protecting against freeze damage in differentiated neuronal cells and parenchymal cells. Serum-free alternatives (sericin and maltose) could increase safety during cell transplantation and regenerative medicine. Considering these, we propose an effective freezing solution for the cryopreservation of neuronal cells.
In cartilage regenerative medicine, transplanted chondrocytes contain a mixture of populations, that complicates the regeneration of uniform cartilage tissue. Our group previously reported that chondrocytes with higher chondrogenic ability could be enriched by selection of rapidly growing cells. In this study, the detailed properties of rapidly growing chondrocytes were examined and compared to slowly growing cells.
Human auricular chondrocytes were fluorescently labeled with carboxyfluorescein succinimidyl ester (CFSE) and analyzed using flow cytometry, focusing on division rates as indicated by fluorescence intensity and cell morphology according to the forward scatter and side scatter. Mature adipocyte-derived dedifferentiated fat cells (DFATs) are mesenchymal stem cell (MSC)-like cells with high proliferative ability and multilineage differentiation potential. In this study, we first examined whether DFATs can be prepared from infrapatellar fat pad (IFP) and then compared phenotypic and functional properties of IFP-derived DFATs (IFP-DFATs) with those of subcutaneous adipose tissue (SC)-derived DFATs (SC-DFATs).
Mature adipocytes isolated from IFP and SC in osteoarthritis patients (n=7) were cultured by ceiling culture method to generate DFATs. Obtained IFP-DFATs and SC-DFATs were subjected to flow cytometric and microarray analysis to compare their immunophenotypes and gene expression profiles. Cell proliferation assay and adipogenic, osteogenic, and chondrogenic differentiation assays were performed to evaluate their functional properties.
DFATs could be prepared from IFP and SC with similar efficiency. IFP-DFATs and SC-DFATs exhibited similar immunophenotypes (CD73
, CD90
, CD10ce for chondrogenic regeneration.
The BMP signaling pathway plays a key role in growth, differentiation and patterning during neural development. Recent work on the generation of a self-organization of three-dimensional retinal organoid (3D-retina) from human pluripotent stem cells (hPSCs) revealed that addition of recombinant human BMP4 (rhBMP4) promotes retinal differentiation in the early neural differentiation stage. For clinical application, efficient differentiation from hPSCs to retinal cells with minimal numbers of off-target non-retinal cells is desirable. We therefore aimed to further improve an efficient retinal differentiation method for future up-scaling of cell production.
hPSCs were differentiated into 3D-retina using a modified SFEBq method. The effect of rhBMP4 with or without Checkpoint kinase 1 (Chk1) inhibitor (PD407824), a modulator of BMP signaling pathway, at day 3 was compared by characterizing the differentiating 3D-retina by the use of the hPSCs and immunohistochemical analysis.

The fundamental treatment of calcinosis has not yet been established. Here, we report for the first time that calcinosis at the nasal spine in patients with MERR can be replaced by cancellous bone-like tissue. Patients 1 and 2 were a 62-year-old Japanese female and a 45-year-old Japanese female (respectively) with MERR who had been previously treated for SSc (Patient 1 limited type, positive for anti-centromere antibody; Patient 2 diffuse type, positive for anti-Scl70 and anti-SS-A antibodies). Patient 3 was a 57-year-old female with MERR who had been previously treated for SSc (diffuse type, positive anti-Scl-70 antibody) and underwent denosumab injection for osteoporosis. Cone-beam computed tomography (CBCT) and CT images in the calcifications at the nasal spine in Patient 1 and 2 were replaced with cancellous bone-like tissue, but not in Patient 3. Serum laboratory examination was performed to assess the systemic bone disease. All three patients had normal clinical data within the references, apart from slightly higher 1,25-dihydroxyvitamin D levels in Patient 1. SSc patients with calcinosis in the maxillofacial area need to be examined carefully for bone replacement using CBCT or CT.
