Wheat is one of three major food crops in China. Alternaria species can cause spoilage of wheat with consequent mycotoxin accumulation. Alternariol (AOH), alternariol monomethyl ether (AME), and tenuazonic acid (TeA) are the most common and frequently studied mycotoxins. There are limited regulations placed on Alternaria mycotoxin concentrations worldwide due to the lack of toxicity data available. More data on the levels of mycotoxin contamination are also needed. It is also important to reduce the risks of Alternaria mycotoxins.

One hundred and thirty-two wheat samples were collected from Hebei Province, China, and analyzed for AOH, AME, and TeA. Tenuazonic acid was found to be the predominant Alternaria mycotoxin, especially in flour samples. Studying Alternaria species that cause black-point disease of wheat indicated that Alternaria alternata and Alternaria tenuissima were the dominant species. Most of the Alternaria strains studied produced more than one mycotoxin and TeA was produced at the highest in wheat, or as a natural alternative food preservative based on its strong antifungal and antimycotoxigenic activity against Alternaria strains. © 2020 Society of Chemical Industry.
Bee pollen is considered as a treasure trove of human and animal nutrients as a result of its extensive nutritional and therapeutic properties. However, the sophisticated pollen wall can largely limit the digestibility and bioavailability of these nutrients.

An ultrasonication and high shear technique was used to break the walls of five species of bee pollen, including rape bee pollen, lotus bee pollen, camellia bee pollen, wuweizi bee pollen and apricot bee pollen. We compared the digestibilities of bee pollen with or without wall-disruption. After in vitro and in vivo digestion, unbroken bee pollen grains were still intact and the fragments of wall-disrupted bee pollen still remained as fragments. Mouse in vivo digestion results suggested that the wall-disrupted bee pollen was more easily emptied from the gastrointestinal tract than unbroken bee pollen. After dynamic in vitro digestion, the digestibilities of protein and crude fat in wall-disrupted bee pollen significantly increased to more than 80%; similarly, the release rates of amino acids and reducing sugars in all wall-disrupted samples were almost 1.5 and 2 times as much as those of unbroken samples.

Acclimatization to hypoxia leads to a reduction in plasma volume (PV) that restores arterial O
content. Findings from studies investigating the mechanisms underlying this PV contraction have been controversial, possibly as experimental conditions were inadequately controlled. We examined the mechanisms underlying the PV contraction evoked by 4 days of exposure to hypobaric hypoxia (HH) in 11 healthy lowlanders, while strictly controlling water intake, diet, temperature and physical activity. Exposure to HH-induced an ∼10% PV contraction that was accompanied by a reduction in total circulating protein mass, whereas diuretic fluid loss and total body water remained unchanged. Our data support an oncotically driven fluid redistribution from the intra- to the extravascular space, rather than fluid loss, as the mechanism underlying HH-induced PV contraction.

But researchers know little about how children learn to use everyday artifacts. We encouraged forty-four 12- to 60-month-old children to unzip a vinyl pouch during a single 60-s trial. Although unzipping a pouch may seem simple, it is not. Unzipping requires precise role-differentiated bimanual actions-one hand must stabilize the pouch while the other hand applies a pulling force on the tab. Moreover, kinematic data from six adults showed that the tolerance limits for applying the forces are relatively narrow (pulling the tab within 63° of the zipper teeth while stabilizing the pouch within 4 cm of the slider). check details Children showed an age-related progression for the unzipping action. The youngest children did not display the designed pulling action; children at intermediate ages pulled the tab but applied forces outside the tolerance limits (pulled in the wrong direction, failed to stabilize the pouch in the correct location), and the oldest children successfully implemented the designed action. GIRK2-null mice have been widely investigated for their distinct behaviour and altered depotentiation following long-term potentiation (LTP), whereas GIRK1 mice are less well characterized. Here we utilize a novel knockin mouse strain in which the GIRK1 subunit is fluorescently tagged with yellow fluorescent protein (YFP-GIRK1) and the GIRK1-null mouse line to investigate the role of GIRK1 in neuronal processes such as spatial learning and memory, locomotion and depotentiation following LTP. Neurons dissected from YFP-GIRK1 mice had significantly reduced potassium currents and this mouse line phenotypically resembled GIRK1-null mice, making it a 'functional knockdown' model of GIRK1-containing channels. YFP-GIRK1 and GIRK1-null mice had increased locomotion, reduced spatial learning and memory and blunted depotentiation following LTP.

Traditionally, consistent cell density is difficult to achieve, owing to the inaccurate regulation of cell density with manual feedback. A novel cell culture method with automatic feedback is proposed for real-time regulation of cell density based on microfluidic chip in this paper. Here, an integrated microfluidic system combining cell culture, density detection, and control of proliferation rate was developed. Interdigital electrode structures were sputtered on the microchannel automatically to provide the real-time feedback information of impedance. The most sensitive frequency was studied to improve the detection resolution of the sensing chip. Cells were cultured on the chip surface and cell density was detected by monitoring the alternation of the impedance. The feedback controller was established by the least squares support vector machines. Then, the cell proliferation rate was automatically controlled using the feedback controller to achieve the desired cell density in the repeated assays. The results show that the standard error of this method is 2.8% indicating that the method can keep a consistency of cell density in the repeated assays. This study provides a basis for improving the accuracy and repeatability in the further assays of finding the optimal drug concentration.To determine the role of the transplantation of bone marrow-derived mesenchymal stem cells (MSCs) in podocyte renewal, we studied BALB/C mice with or without adriamycin-induced acute kidney injury. MSCs were transplanted ectopically under the capsule of the left kidney or into the peritoneal cavity after the onset of kidney injury to test testing their local or systemic paracrine effects, respectively. Adriamycin produced increases in urine protein creatinine ratios, blood urea nitrogen, and blood pressure, which improved after both renal subcapsular and intraperitoneal MSCs transplants. The histological changes of adriamycin kidney changes regressed in both kidneys and in only the ipsilateral kidney after intraperitoneal or renal subcapsular transplants indicating that the benefits of transplanted MSCs were related to the extent of paracrine factor distribution. Analysis of kidney tissues for p57-positive parietal epithelial cells (PECs) showed that MSC transplants restored adriamycin-induced decreases in the abundance of these cells to normal levels, although after renal subcapsular transplants these changes did not extend to contralateral kidneys. Moreover, adriamycin caused inflammatory activation of PECs with coexpression of CD44 and phospho-ERK, which was normalized in both or only ipsilateral kidneys depending on whether MSCs were transplanted in the peritoneal cavity or subcapsular space, respectively.The aim of this study was to isolate and characterize marine bacterial strains capable of converting selenite to elemental selenium with the formation of Se nanoparticles (SeNPs). For the first time, a novel marine strain belonging to Bacillus amyloliquefaciens (GenBank accession no. MK392020) was isolated from the coast of the Caspian Sea and characterized based on its ability for transformation of selenite to SeNPs under aerobic conditions. The preliminary formation of SeNPs was confirmed via color changes and the products characterized by UV-Vis spectroscopy. The field-emission scanning electron microscopy (FESEM) together with energy-dispersive X-ray (EDX) analysis showed the presence of the spherical SeNPs on both the surface of the bacterial biomass and in the supernatant solution. Dynamic light scattering (DLS) analysis showed the SeNPs to have an average particle size (Z-average) around 45.4-68.3 nm. The X-ray diffraction (XRD) studies substantiated the amorphous nature of the biosynthesized SeNPs. Fourier-transform infrared spectroscopic (FTIR) studies of the SeNPs indicated typical proteinaceous and lipid-related bands as capping agents on the SeNPs. Different effective parameters corresponding the yield of SeNPs by B. amyloliquefaciens strain SRB04 were optimized under resting cell strategy. Results showed that the optimal process conditions for SeNP production were 2 mM of selenite oxyanion, 20 g/L of cell biomass, and 60 h reaction time. The synthesized SeNPs had a remarkable antibacterial activity on Staphylococcus aureus compared with chloramphenicol as a broad-spectrum antibiotic.
