We show that the lack of phylogenetic resolution is not necessarily problematic for all analyses and that its effect will depend on the chosen metric and on the solutions used to deal with the problem. Based on our results, we suggest that ecologists should prefer the Operational tree solution to remove polytomies in the phylogenetic tree and take careful consideration while designing experiments, and analyzing and interpreting the results of phylogenetic metrics.
Localizing the source of ectopic adrenocorticotropic hormone secretion (EAS) is challenging. This study compared the diagnostic value of
Ga-DOTATATE PET/CT and
F-FDG PET/CT in tumors with EAS.
Thirty-six patients with a suspicion of EAS were enrolled to undergo both
Ga-DOTATATE PET/CT and
F-FDG PET/CT within 4weeks for comparison. Twenty-three underwent surgical resection or biopsy. Immunohistochemical staining for SSTR2 and Ki-67 was performed to correlate with
Ga-DOTATATE uptake and
F-FDG uptake, respectively.
EAS tumors were observed in 20/23 patients. Among the 20 patients with histologically proven EAS tumors,
Ga-DOTATATE PET/CT correctly identified the tumor in 15 (75.0%), with an SUV
ranging from 1.4 to 20.7 (6.7 ± 5.5).
F-FDG PET/CT correctly identified the tumor in 12 (60.0%) patients, with an SUV
ranging from 1.8 to 10.0 (4.0 ± 2.1). Moreover,
Ga-DOTATATE PET/CT unmasked the sources of EAS in 6 patients with negative
F-FDG uptake, and
F-FDG PET/CT unmasked the sources in 3 patients with negative
Ga-DOTATATE uptake, resulting in EAS tumors being identified in 18 (90%) patients by combining
Ga-DOTATATE PET/CT and
F-FDG PET/CT.
Ga-DOTATATE PET/CT and
F-FDG PET/CT are complementary in localizing and discriminating the source of EAS.
Ga-DOTATATE PET/CT combined with
F-FDG PET/CT had higher detection rate than each alone.

Therefore, the development and validation of audiovisual educational material with the participation of health specialists and patients could lead to an improved level of compliance with treatment, while favoring its acceptance. To design and validate audiovisual educational material generated by a multidisciplinary and participative group of patients and health specialists. This study was carried out using a sequential methodology, including qualitative and quantitative techniques (1) a non-participative observational study with patients and a non-systematic literature search were performed to identify conceptual topics. (2) Pilot videos were qualitatively assessed by patients and health specialists (focus groups and expert committees). (3) Improved versions of seven videos were quantitatively evaluated by patients and specialists following qualitative criteria of attraction, understanding, involvement, acceptance and induction of action. 74 patients with RA, 10 rheumatologists, 4 pharmacists and 2 medical anthropologists participated in the different phases of validation. A total of seven videos lasting 3 min each were generated, incorporating the most relevant suggestions by patients and healthcare professionals. The final version of the videos led to a mean compliance of 96.04 ± 5.2%, according to a representative group of patients and a mean 89.6 ± 9.4%, according to health professionals. With the participation of both patients and health specialists, seven audiovisual educational video recordings were developed and validated, reaching high levels of compliance in accordance with international criteria.
This study aimed to investigate the possibility of UCP-2 inhibitor in reducing acquired resistance of trastuzumab to improve the outcome of patients receiving trastuzumab therapy by exploring the relationship between UCP-2 expression and HER2 signaling pathway and examining whether UCP-2 expression was modulated by trastuzumab treatment.
32 women diagnosed with primary HER2-positive breast cancer were recruited in this study. Needle biopsy was obtained from patients before they received at least four cycles neoadjuvant therapy containing trastuzumab in combination with chemotherapy. Surgical tumor biopsy was obtained during surgical procedure after the neoadjuvant therapy. Levels of HER2 phosphorylation and UCP-2 expression were detected by immunohistochemistry (IHC) and compared between tumor needle biopsy tissue and surgical tumor samples of these patients, as well as in BT474 breast cancer cells before and after trastuzumab treatment. The study involved examining the intraspecific variability and demographic structure of Echinococcus granulosus sensu stricto (s.s.) in common hosts of this parasite. Molecular surveillance included the fragment amplification of a partial mitochondrial gene, cox1 (750 bp), for a total of 69 isolates derived from sheep (n = 52), cattle (n = 11), pigs (n = 4), and goats (n = 2). It was ascertained that E. granulosus s.s. was the primary agent of infection among these ungulates and G1 genotype was highly prevalent (79.71%). Considerable intraspecific variation was found, revealing the existence of 22 haplotypes with relatively high haplotype (0.8555 ± 0.033) and low nucleotide diversities (0.00281 ± 0.00030). Population demographics indicated an expanding parasitic population signifying negative deviation from neutrality indices. Little genetic differentiation was found between the subpopulations of E. granulosus s.s. in the island. Moreover, the geographic dispersal of genotypes G1 and G3 also indicated similarity between Sardinian and mainland Echinococcus granulosus s.s. populations reaffirming the sympatric occurrence and efficient transmission of G1 and G3 genotypes. Molecular survey of CE has the potential to yield baseline information on the infective genotypes among the intermediate hosts and helps in devising suitable control strategies for curtailing the disease.Poultry coccidiosis causes considerable economical losses to the livestock industry. Eimeria parasites are responsible for this disease. On a global scale, E. acervulina and E. tenella are amongst the most common Eimeria spp. infecting broilers. E. tenella is commonly used as infection model in in vivo and in vitro studies. On the other hand, E. acervulina has barely been studied under in vitro conditions. A well established and widely used in vitro model for E. tenella infection is the Madin-Darby bovine kidney cell line (MDBK); however, little is known regarding suitability of MDBK cells as host cells for E. acervulina. We infected MDBK monolayers with two different doses, 5 × 104 and 2 × 105, of E. acervulina sporozoites and evaluated cultures at 24 and 96 h post infection (hpi). For comparison, we ran an identical infection assay using E. tenella sporozoites. To assess parasite reproduction, the number of DNA copies of E. acervulina SCAR marker and E. tenella ITS-1 gene was quantified using real-time quantitative PCR. We found that the number of E. acervulina copies increased significantly at 24 hpi in comparison to E. tenella (p less then 0.05). After 96 hpi, E. acervulina gene copies were considerably reduced while E. tenella continued to multiply (p less then 0.05). Our results show that MDBK monolayers could be used for in vitro research aimed to study E. acervulina sporozoite cell invasion. Nevertheless, modifications of in vitro cultivation appear necessary to allow qualitative and quantitative studies over longer periods of parasite reproduction.Appraisal theories of emotion sustain that stimuli containing high biological relevance preferentially capture our attention, regardless of their valence. In this work, we study the priming effect of both cute and threatening pictures on global/local visual processing. Seventy-eight males and 168 females in different menstrual cycle phases performed the global/local Navon letter task immediately after observing cute (infants and baby animals), threatening (animals in aggressive poses) or neutral pictures. Hierarchical stimuli were made up of global and local letters that could be either congruent (e.g. global H, localH) or incongruent (e.g. global H, localS). While we observed no interaction between affective primes and sex/menstrual cycle phase, each of these variables did have an independent effect on the global/local task. Thus, letter identification was significantly slower in the global task (only) after cute vs. neutral primes. Relevantly, the local-minus-global RT index revealed an attentional narrowing after both cute and threatening primes (vs. neutral primes) in incongruent trials. As for sex effects, a facilitation of global vs. local processing was observed in both sexes. However, women registered slower RTs than men, whereas women in the luteal phase showed faster RTs than those in follicular phase in the local task. This suggests that women, mainly those in their luteal menstrual phase, tend to rely on a more analytical style of processing while attending to hierarchical stimuli. check details Most importantly, stimuli containing high biological significance drive narrowing of the attentional focus in global/local visual processing, especially in conditions of higher attentional demand.
To discuss the potential utility of newer imaging modalities including micro-ultrasound and PSMA-PET for the detection of clinically significant prostate cancer, technologies that may gain roles as adjuncts to multiparametric magnetic resonance imaging (mpMRI) in the active surveillance (AS) setting.
Narrative review of two new imaging modalities used for primary prostate cancer through April 2021. A targeted search was performed to identify current relevant literature on the role of new imaging modalities for primary prostate cancer using search terms "micro-ultrasound," "molecular imaging," "prostate cancer," "active surveillance," "multiparametric MRI," "PI-RADS," "PRI-MUS," and "detection rate." In addition, references of included articles were screened for further relevant publications.
Micro-ultrasound (micro-US) and prostate-specific membrane antigen-positron emission tomography (PSMA-PET) are increasing in their use and applicability to prostate cancer imaging. Micro-US is used for cancer detection and may identify higher grade cancers more accurately than conventional ultrasound, despite technical hurdles in its initial launch. PSMA-PET is highly sensitive and specific for high-grade and metastatic prostate cancer, though costly and not easily available. Though data are sparse, it may have an emerging role in cancer diagnosis in select localized cases, and in some men considering (or currently on) AS who have indications of more aggressive disease.
There are very limited data on micro-US and PSMA-PET in AS patients. However, given the ability of these modalities to identify high-grade cancer, their judicious use in AS patients may be of utility in the future.
