A microtubule hollow structure is one type of cytoskeletons which directs a number of important cellular functions. When recapitulating biological events in a cell-free system, artificial frames are often required to execute similar cytoskeletal functions in synthetic systems. Here, I report a prototypical microtubular assembly using a DNA origami nanostructuring method. Through structural design at the molecular level, 32HB (helices bundle)-based DNA origami objects can form micrometers long tubular structures via shape-complementary side patterns engagement and head-to-tail blunt-end stacking. Multiple parameters have been investigated to gain optimized polymerization conditions. Conformational change with an open vs closed hinge is also included, rendering conformational changes for a dynamic assembly. When implementing further improved external regulation with DNA dynamics (DNA strand displacement reactions or using other switchable non-canonical DNA secondary structures) or chemical stimuli, the DNA origami-based microtubule analogue will have great potential to assemble and disassemble on purpose and conduct significantly complicated cytoskeletal tasks in vitro.Chiral interfaces provide a new platform to execute quantum control of light-matter interactions. One phenomenon which has emerged from engineering such nanophotonic interfaces is spin-momentum locking akin to similar reports in electronic topological materials and phases. While there are reports of spin-momentum locking with combination of chiral emitters and/or chiral metamaterials with directional far field excitation it is not readily observable with both achiral emitters and metamaterials. Here, we report the observation of photonic spin-momentum locking in the form of directional and chiral emission from achiral quantum dots (QDs) evanescently coupled to achiral hyperbolic metamaterials (HMM). Efficient coupling between QDs and the metamaterial leads to emergence of these photonic topological modes which can be detected in the far field. We provide theoretical explanation for the emergence of spin-momentum locking through rigorous modeling based on photon Green's function where pseudo spin of light arises from coupling of QDs to evanescent modes of HMM.
To compare the effect of intact umbilical cord milking (MUC) and delayed cord clamping (DCC) on venous hematocrit at 48 (±6) hours in late preterm and term neonates (350/7- 426/7 wk).

Randomized trial.

All late preterm and term neonates (350/7 - 426/7 wk) neonates born in the labor room and maternity operation theatre of tertiary care unit were included.

We randomly allocated enrolled neonates to MUC group (cord milked four times towards the baby while being attached to the placenta; n=72) or DCC group (cord clamped after 60 seconds; n=72).

Primary outcome was venous hematocrit at 48 (±6) hours of life. Additional outcomes were venous hematocrit at 48 (±6) hours in newborns delivered through lower segment caesarean section (LSCS), incidence of polycythemia requiring partial exchange transfusion, incidence of hyperbilirubinemia requiring phototherapy, and venous hematocrit and serum ferritin levels at 6 (±1) weeks of age.

The mean (SD) hematocrit at 48 (±6) hours in the MUC group was higher than in DCC group [57.7 (4.3) vs. #link# 55.9 (4.4); P=0.002]. Venous hematocrit at 6 (±1) weeks was higher in MUC than in DCC group [mean (SD), 37.7 (4.3) vs. 36 (3.4); mean difference 1.75 (95% CI 0.53 to 2.9); P=0.005]. Other parameters were similar in the two groups.

To assess efficacy and safety of High flow nasal cannula therapy (HFNC) as primary mode of treatment for children with respiratory distress.

Consecutive patients (1 mo-16 years) with respiratory distress were assessed for respiratory clinical score, COMFORT score and saturation to FiO2 (SF) ratio.

A total of 188 (91.7%) patients out of 205 responded to HFNC alone. The respiratory clinical score and COMFORT score were lower with higher SF ratio in these than 17 patients who required ventilation (P<0.001). Median (IQR) time to failure was 2 (1.75-24) hours. link2 Air leak was seen in 2 (1%) patients.

HFNC is an effective and safe primary mode of respiratory support in children with respiratory distress. Children who succeed on HFNC show a favorable clinical response within first few hours.

Published Indian studies on the economic burden of juvenile idiopathic arthritis (JIA) are lacking.

A prospective observational study recruited pediatric patients aged from 1 to 12 years with JIA in the pediatric rheumatology clinic of a public sector tertiary care hospital. Direct healthcare costs and indirect costs for transportation, accommodation of the caregivers, and productivity loss for work absenteeism were assessed.

The proportions of direct annualized cost assessed in 60 patients (mean (SD) age 8.46 (2.24) year) spent on outpatient visits, blood tests, imaging investigations, other tests, medications and hospitalization were 0.85%, 12.8%, 9.0%, 2.9%, 41.7% and 32.7%, respectively. Direct healthcare costs for blood tests and medicine were lowest in oligoarticular JIA and highest in systemic onset JIA and (P=0.043 and 0.001 respectively). The direct and indirect costs were higher with the use of biologic agents (n=9) than in those without (n=51).

To compare the pulmonary function in adolescents exposed to different concentrations of air pollutants in two different zones.

Two zones based on monitoring of environmental pollutant concentration as high (zone 1) and low (zone 2) were chosen. The lung functions of apparently healthy adolescents (12-15 years) residing in two zones were measured for forced vital capacity (FVC), forced expiratory volume in first second (FEV1), FEV1/FVC ratio, and forced expiratory flow (FEF)25-75.

A total of 302 adolescents (142, zone 1 and 160, zone 2) resided in the study area, with higher than permissible concentrations of PM10 and ozone at both places. Abnormal lung functions were seen in a higher proportion of adolescents in zone 1 than zone 2 (23% and 14%; P=0.04). A significantly lower mean (SD) FEV1 was seen in adolescents in zone 1 than zone 2 [2.9 (0.5) vs. 3.2 (0.4) L, P = 0.04]. A higher proportion of abnormal FEV1/FVC ratio% was seen in zone 1 than 2 (12% vs. 6%, P=0.04), suggestive of an obstructive pattern on spirometry. Higher risk (b 95% CI) for abnormal lung functions was seen with the zone [2.2 (1.1-4.2)], diagnosis of asthma [5.74 (2.4-13.2)], and living within 500 meters from a brickwork [1.8 (1.0-2.5)].

Intravital microscopy (IVM) allows the real-time, direct visualization of microscopic blood vessels. This pilot clinical trial will elucidate the physical and functional characteristics of vessels associated with solid tumors.

The main objective of this study is to determine the feasibility of performing IVM in patients with solid tumors during the standard course of surgical resection. IVM will also be performed when vasopressors or fluid boluses are administered during the standard course of the operation.

This is an open-label, nonrandomized, single-center, pilot study of IVM observation in subjects with solid tumors undergoing surgical resection.

This study was active on January 1, 2019 (NCT03823144) and funded by the Mayo Clinic Florida Cancer Focused Research Team Award. As of September 27, 2020, we had enrolled 20 patients. Accrual period is expected to end by December 31, 2021.

This trial will support the development of interventions to improve patient treatment by extending the application of IVM to the tumor microenvironment. IVM observations during volume and pressor management at the time of surgery may aid in the development of strategies to augment responses to systemic treatments.

However, less is known about how their interplay might influence behavior and physiology. Here we focused on social hierarchy status, a major component of the social environment in mice, and whether it influences behavioral adaptation to chronic stress in a sex-specific manner. We used a high-throughput automated behavioral monitoring system to assess social dominance in same-sex, group-living mice. We found that position in the social hierarchy at baseline was a significant predictor of multiple behavioral outcomes following exposure to chronic stress. Crucially, this association carried opposite consequences for the two sexes. This work demonstrates the importance of recognizing the interplay between sex and social factors and enhances our understating of how individual differences shape the stress response.Connections from the ventral hippocampus (vHPC) to the prefrontal cortex (PFC) regulate cognition, emotion, and memory. link3 These functions are also tightly controlled by inhibitory networks in the PFC, whose disruption is thought to contribute to mental health disorders. However, relatively little is known about how the vHPC engages different populations of interneurons in the PFC. Here we use slice physiology and optogenetics to study vHPC-evoked feed-forward inhibition in the mouse PFC. We first show that cholecystokinin (CCK+), parvalbumin (PV+), and somatostatin (SOM+) expressing interneurons are prominent in layer 5 (L5) of infralimbic PFC. We then show that vHPC inputs primarily activate CCK+ and PV+ interneurons, with weaker connections onto SOM+ interneurons. CCK+ interneurons make stronger synapses onto pyramidal tract (PT) cells over nearby intratelencephalic (IT) cells. However, CCK+ inputs undergo depolarization-induced suppression of inhibition (DSI) and CB1 receptor modulation only at IT cells. Moreover, vHPC-evoked feed-forward inhibition undergoes DSI only at IT cells, confirming a central role for CCK+ interneurons. Together, our findings show how vHPC directly engages multiple populations of inhibitory cells in deep layers of the infralimbic PFC, highlighting unexpected roles for both CCK+ interneurons and endocannabinoid modulation in hippocampal-prefrontal communication.We inoculated 6 cattle with severe acute respiratory syndrome coronavirus 2 and kept them together with 3 uninoculated cattle. We observed viral replication and specific seroreactivity in 2 inoculated animals, despite high levels of preexisting antibody titers against a bovine betacoronavirus.
