coelicolor compared to S. lividans. In condition of phosphate limitation, PhoR/PhoP is known to control positively and negatively, respectively, phosphate and nitrogen assimilation and our study revealed that it might also control the expression of some genes of central carbon metabolism. The tuning down of the regulatory role of PhoR/PhoP in S. coelicolor is thus expected to be correlated with low and high phosphate and nitrogen availability, respectively and with changes in central carbon metabolic features. These changes are likely to be responsible for the observed differences between S. coelicolor and S. lividans concerning energetic metabolism, triacylglycerol biosynthesis and antibiotic production. Furthermore, a novel view of the contribution of the bio-active molecules produced in this context, to the regulation of the energetic metabolism of the producing bacteria, is proposed and discussed.Vascular calcification occurs in various diseases including atherosclerosis, chronic kidney disease and type 2 diabetes but the mechanism underlying mineral deposition remains incompletely understood. Here we examined lower limb arteries of type 2 diabetes subjects for the presence of ectopic calcification and mineral particles using histology, electron microscopy and spectroscopy analyses. While arteries of healthy controls showed no calcification following von Kossa staining, arteries from 83% of diabetic individuals examined (19/23) revealed microscopic mineral deposits, mainly within the tunica media. Mineralo-organic particles containing calcium phosphate and proteins such as albumin, fetuin-A and apolipoprotein-A1 were detected in calcified arteries. Ectopic calcification and mineralo-organic particles were observed in a majority of diabetic patients and predominantly in arteries showing hyperplasia. While a low number of subjects was examined and information about disease severity and patient characteristics is lacking, these calcifications and mineralo-organic particles may represent signs of tissue dysfunction.Dimethyl ether (DME) is an advanced second-generation biofuel produced via methanol dehydration over acid catalysts such as γ-Al2O3, at temperatures above 240 °C and pressures above 10 bar. Heteropolyacids such as tungstosilicic acid (HSiW) are Brønsted acid catalysts with higher DME production rates than γ-Al2O3, especially at low temperatures (140-180 °C). In this work, we show that the performance of supported HSiW for the production of DME is strongly affected by the nature of the support. TiO2 and SiO2 supported HSiW display the highest DME production rates of ca. 50 mmolDME/h/gHSiW. Characterization of acid sites via 1H-NMR, NH3-isotherms and NH3-adsrobed DRIFT reveal that HSiW/X have Brønsted acid sites, HSiW/TiO2 showing more and stronger sites, being the most active catalyst. Methanol production increases with T until 200 °C where a rapid decay in methanol conversion is observed. This effect is not irreversible, and methanol conversion increases to ca. 90% by increasing reaction pressure to 10 bar, with DME being the only product detected at all reaction conditions studied in this work. The loss of catalytic activity with the increasing temperature and its increasing with reaction pressure accounts to the degree of contribution of the pseudo-liquid catalysis under the reaction conditions studied.Alterations in DNA methylation occur during development, but the mechanisms by which they influence gene expression remain uncertain. There are few examples where modification of a single CpG dinucleotide directly affects transcription factor binding and regulation of a target gene in vivo. Here, we show that the erythroid transcription factor GATA-1 - that typically binds T/AGATA sites - can also recognise CGATA elements, but only if the CpG dinucleotide is unmethylated. We focus on a single CGATA site in the c-Kit gene which progressively becomes unmethylated during haematopoiesis. We observe that methylation attenuates GATA-1 binding and gene regulation in cell lines. In mice, converting the CGATA element to a TGATA site that cannot be methylated leads to accumulation of megakaryocyte-erythroid progenitors. Thus, the CpG dinucleotide is essential for normal erythropoiesis and this study illustrates how a single methylated CpG can directly affect transcription factor binding and cellular regulation.Development of multicellularity was one of the major transitions in evolution and occurred independently multiple times in algae, plants, animals, and fungi. However recent comparative genome analyses suggest that fungi followed a different route to other eukaryotic lineages. To understand the driving forces behind the transition from unicellular fungi to hyphal forms of growth, we develop a comparative model of osmotrophic resource acquisition. This predicts that whenever the local resource is immobile, hard-to-digest, and nutrient poor, hyphal osmotrophs outcompete motile or autolytic unicellular osmotrophs. This hyphal advantage arises because transporting nutrients via a contiguous cytoplasm enables continued exploitation of remaining resources after local depletion of essential nutrients, and more efficient use of costly exoenzymes. The model provides a mechanistic explanation for the origins of multicellular hyphal organisms, and explains why fungi, rather than unicellular bacteria, evolved to dominate decay of recalcitrant, nutrient poor substrates such as leaf litter or wood. This work aimed to use artificial intelligence to predict subjective refraction from wavefront aberrometry data processed with a novel polynomial decomposition basis. Subjective refraction was converted to power vectors (M, J0, J45). Three gradient boosted trees (XGBoost) algorithms were trained to predict each power vector using data from 3729 eyes. Selleck LY-188011 The model was validated by predicting subjective refraction power vectors of 350 other eyes, unknown to the model. The machine learning models were significantly better than the paraxial matching method for producing a spectacle correction, resulting in a mean absolute error of 0.301 ± 0.252 Diopters (D) for the M vector, 0.120 ± 0.094 D for the J0 vector and 0.094 ± 0.084 D for the J45 vector. Our results suggest that subjective refraction can be accurately and precisely predicted from novel polynomial wavefront data using machine learning algorithms. We anticipate that the combination of machine learning and aberrometry based on this novel wavefront decomposition basis will aid the development of refined algorithms which could become a new gold standard to predict refraction objectively.
