T cell recognition of a cognate peptide-major histocompatibility complex (pMHC) presented on the surface of infected or malignant cells is of the utmost importance for mediating robust and long-term immune responses. The advent and success of cancer immunotherapies, however, have opened new perspectives for the investigation of the role of inherited genetic variation in codetermining outcome and safety. It is increasingly likely that the entirety of germline genetic variation involved in regulating immune responses accounts for a significant fraction of the observed variability in responses to cancer immunotherapies. Although germline genetic data from patients treated with cancer immunotherapies are still scarce, this line of research benefits from a vast body of knowledge derived from studies into autoimmune and infectious disease phenotypes, thus not requiring a start from a blank slate. Here, we discuss how a thorough investigation of genomic variation relevant for individuals' variability in (auto)immune responses can contribute to the discovery of novel treatment approaches and drug targets, and yield predictive biomarkers to stratify cancer patient populations in precision and personalized medicine settings.

Plasmonic nanomaterials have become an integral part of numerous technologies, where they provide important functionalities spanning from extraction and harvesting of light in thin film optical devices to probing of molecular species and their interactions on biochip surfaces. More recently, we witness increasing research efforts devoted to a new class of plasmonic nanomaterials that allow for on-demand tuning of their properties by combining metallic nanostructures and responsive hydrogels. This review addresses this recently emerged vibrant field, which holds potential to expand the spectrum of possible applications and deliver functions that cannot be achieved by separate research in each of the respective fields. It aims at providing an overview of key principles, design rules, and current implementations of both responsive hydrogels and metallic nanostructures. We discuss important aspects that capitalize on the combination of responsive polymer networks with plasmonic nanostructures to perform rapid mechanical actuation and actively controlled nanoscale confinement of light associated with resonant amplification of its intensity. The latest advances towards the implementation of such responsive plasmonic nanomaterials are presented, particularly covering the field of plasmonic biosensing that utilizes refractometric measurements as well as plasmon-enhanced optical spectroscopy readout, optically driven miniature soft actuators, and light-fueled micromachines operating in an environment resembling biological systems.
Reliable markers to predict the response to neoadjuvant chemoradiotherapy (nCRT) in locally advanced rectal cancer (LARC) are lacking. We aimed to assess the ability of 18F-FDG PET/MRI to predict response to nCRT among patients undergoing curative-intent surgery.

Patients with histological-confirmed LARC who underwent curative-intent surgery following nCRT and restaging with 18F-FDG PET/MRI were included. Statistical correlation between radiomic features extracted in PET, apparent diffusion coefficient (ADC) and T2w images and patients' histopathologic response to chemoradiotherapy using a multivariable logistic regression model ROC-analysis.

Overall, 50 patients were included in the study. A pathological complete response was achieved in 28.0% of patients. Considering second-order textural features, nine parameters showed a statistically significant difference between the two groups in ADC images, six parameters in PET images and four parameters in T2w images. Combining all the features selected for the three techniques in the same multivariate ROC curve analysis, we obtained an area under ROC curve of 0.863 (95% CI, 0.760-0.966), showing a sensitivity, specificity and accuracy at the Youden's index of 100% (14/14), 64% (23/36) and 74% (37/50), respectively.

We report on the environmentally friendly and efficient syntheses of a homologous series of [(RSO2)ZnR]n complexes from reactions involving homoleptic R2Zn (R = Me, tBu, Ph) compounds and SO2. Diffusion ordered spectroscopy experiments indicate that the resulting compounds predominately occur as solvated dimers, [(RSO2)ZnR(THF)]2, in THF solution irrespective of the character of the group bonded to the zinc centres. In turn, these organozinc sulfinates exhibit structurally diversified molecular and supramolecular arrangements in the solid state, as evidenced by single-crystal X-ray diffraction studies. The methyl compound crystallises as a one-dimensional polymer, [(MeSO2)ZnMe]n, and the use of tBu2Zn and Ph2Zn leads to molecular aggregates, a tetramer [(tBuSO2)ZntBu]4, and a solvated [(PhSO2)ZnPh]2·2THF dimer, respectively. The energy barriers of the ˙OH-induced H atom abstractions were simulated by the density functional theory (DFT) calculation performed with GAUSSIAN using the method of M06-2X and the basis set of 6-311++G(3df,2p). According to the calculation results, the -COOH and -NH3+ groups with strong negative inductive effects increase the energy barriers and thus decrease the ˙OH reaction rate constants. In contrast, the presence of a -OH or -CH3 group with weak negative or positive inductive effects can reduce energy barriers and hence increase the ˙OH reaction rate constants. To improve the previous structure-activity relationship, the contribution factors of -NH3+ at Cα-atom and Cβ-atom are determined as 0.07 and 0.15, respectively. We analyzed factors that might be associated with WLST, including procedure type, age, sex, race, insurance, Glasgow Coma Scale score, mechanism of injury, geographic region, and institutional size and teaching status. Adjusted logistic regression was performed to examine factors associated with WLST. Sixty-nine thousand fifty-three patients were identified 66% male, 77% with isolated TBI, and 7.8% had WLST.
