To understand the immunological mechanisms that mediate long-lasting clinical remission after RTX treatment, we analyzed the phenotype of DSG-specific memory B cells and DSG-specific T follicular helper cells by flow cytometry and measured antibody-secreting cells by enzyme-linked immune absorbent spot in patients treated with corticosteroids alone or RTX. This post hoc analysis of the RITUX3 trial showed that RTX induced a significant decrease of IgG-switched DSG-specific memory B cells. Accordingly, anti-DSG antibody-secreting cells were no longer detected in patients in complete remission after RTX. In contrast, corticosteroids did not modify the frequency or the phenotype of DSG-specific memory B cells, and anti-DSG antibody-secreting cells were still detected after treatment, even in patients in remission. Using peptide-HLADRB1∗0402 tetramer staining, we identified DSG-3-specific T follicular helper cells, which dramatically decreased after RTX, while remaining stable after corticosteroid treatment. Our findings suggest that long-lasting response to RTX in pemphigus relies on the decrease of DSG-specific circulating T follicular helper cells, which correlates with a sustained depletion of IgG-switched memory autoreactive B cells, leading to the disappearance of anti-DSG antibody-secreting cells.Pemphigus is a group of autoimmune bullous diseases characterized by the presence of autoantibodies against adhesion molecules, desmogleins (Dsg) and desmocollins (Dsc). Pathogenicity of anti-Dsc3 antibodies in pemphigus has been demonstrated, however its characteristics have not yet been elucidated. We aimed to analyze the characteristics of anti-Dsc3 antibodies using Dsc3 domain-swapped Dsg2 molecules in which the prosequence and five extracellular (EC) domains of Dsg2 were replaced with the corresponding domains of human Dsc3. Using these proteins, we established an enzyme-linked immunosorbent assay (ELISA) and analyzed sera from 56 patients with pemphigus. In 34 pemphigus sera positive for Dsc3 full EC domains, 15 sera (44.1 %) were positive for EC2 domain, whereas other domains were rarely positive. We assessed the reactivity to calcium-dependent epitope in Dsc3 by ELISA with ethylenediaminetetraacetic acid (EDTA). The reactivity with EC2 domain was mostly compromised in the presence of EDTA. In the in vitro assay, IgG from patient with paraneoplastic pemphigus pre-adsorbed with EC2 prevented both reduction of Dsc3 and keratinocyte dissociation as compared to that with EDTA-treated EC2. In this study, we identify miR-130a to be downregulated in cSCC compared to healthy skin and precancerous lesions (actinic keratosis). Moreoever, we show that its expression is regulated at the transcriptional level by HRAS and MAPK signaling pathway. We demonstrate that overexpession of miR-130a suppresses long-term capacity of growth, cell motility and invasion ability of human cSCC cell lines. We report that miR-130a suppresses the growth of cSCC xenografts in mice. Mechanistically, miR-130a directly targets ACVR1 (ALK2), and changes in miR-130a levels result in the decreased activity of the BMP/SMAD pathway through ACVR1.
