Advancing bioethics in medical research in Africa and steadily improving its global networks and infrastructures, CERMEL serves as a reference centre for several international consortia. In close collaboration with national authorities, CERMEL has become one of the main training hubs for medical research in Central Africa. It is hoped that CERMEL and its leitmotiv "to improve medical care for local populations" will serve as an inspiration to other institutions in sub-Saharan Africa to further increase African capacity to advance medicine.
To compare the posterior condylar angle measured with Kanekasu radiograph and 2D-CT with the gold standard 3D-CT following primary total knee arthroplasty (TKA).
Eighty-two knees with pain following TKA were included in this retrospective study. Two independent raters measured the anatomical and surgical posterior condylar angles twice on each Kanekasu radiograph and 2D-CT. These measurements were compared against the 3D-CT measurement. buy Veliparib The intra- and interrater reliability of the Kanekasu radiograph and 2D-CT and the correlation with 3D-CT were calculated.
The intra- and interrater reliability for measurements of the anatomical posterior condyle angle for the Kanekasu radiograph and the 2D-CT were excellent for both raters (0.85-0.92). For the less experienced rater 1, the intrarater reliability was significantly better for 2D-CT than Kanekasu radiograph for measuring both the surgical (p < 0.01) and anatomical posterior condyle angles (p < 0.05). For the experienced rater 2, the intrarater relialable, it should be preferred over 2D-CT and Kanekasu view radiograph for femoral component rotation measurements.
In a mouse model of diabetic cardiomyopathy (DCM) the expression of the circular RNA circHIPK3 was found to be significantly increased. This study aimed to discover the molecular mechanisms linking circHIPK3 to the pathogenesis of DCM.
The diabetic mouse model was established by i.p. injection of streptozotocin, which led to the development of DCM. Echocardiographic measurements were used to evaluate cardiac structure and function, and histological staining was applied to detect myocardial fibrosis in mice. 5-Ethynyl-2'-deoxyuridine incorporation was performed to determine cell proliferation and RNA fluorescent in situ hybridisation was employed to examine circHIPK3 expression in cardiac fibroblasts. RNA immunoprecipitation and luciferase reporter assay were conducted to explore the pathological mechanism of circHIPK3 in myocardial fibrosis.
Knockdown of circHIPK3 was found to attenuate myocardial fibrosis and enhance cardiac function in DCM mice. In addition, silencing of circHIPK3 could suppress proliferation of cardiac fibroblasts treated with angiotensin II. Furthermore, RNA immunoprecipitation and luciferase reporter assay revealed a circHIPK3-miR-29b-3p-Col1a1-Col3a1 regulatory network in the pathogenesis of myocardial fibrosis.
circHIPK3 contributes to increased myocardial fibrosis during DCM by functioning as a competing endogenous RNA that upregulates Col1a1 and Col3a1 expression through suppressing miR-29b-3p.

Although many studies have shown enhancements of target-related electroencephalographic activity (EEG) with synthetic cues, there have been fewer studies demonstrating such enhancements during search with scene context and objects in real world scenes. Here, observers covertly searched for a target in images of real scenes while we used EEG to measure the steady state visual evoked response to objects flickering at different frequencies. The target appeared in its typical contextual location or out of context while we controlled for low-level properties of the image including target saliency against the background and retinal eccentricity. A pattern classifier using EEG activity at the relevant modulated frequencies showed target detection accuracy increased when the target was in a contextually appropriate location. A control condition for which observers searched the same images for a different target orthogonal to the contextual manipulation, resulted in no effects of scene context on classifier performance, confirming that image properties cannot explain the contextual modulations of neural activity. Pattern classifier decisions for individual images were also related to the aggregated observer behavioral decisions for individual images. Together, these findings demonstrate target-related neural responses are modulated by scene context during visual search with real world scenes and can be related to behavioral search decisions.Light touch of a stable reference reduces sway during standing. link= buy Veliparib However, unexpected displacement of a light touch reference leads to short-latency reactions in ankle muscles consistent with a balance reaction, that are replaced by responses in arm muscles on subsequent trials. We anticipated that the excitability of sensorimotor pathways arising from finger cutaneous afferents would reflect these changes in behavior. We hypothesized that (1) interlimb cutaneous reflexes in muscles of the ipsilateral leg, derived from median nerve (MED) stimulation would be facilitated when touch was stable, but reduced when touch was unreliable, (2) intralimb MED reflexes in muscles of the homonymous arm would be facilitated when touch was unreliable and participants tracked the touch reference with arm movements, and (3) radial nerve (RAD) evoked reflexes would be unaffected, given that the RAD innervation territory is not involved in the light touch task. Cutaneous reflexes were evoked using a transcutaneous train of pulsesSickle cell disease (SCD) is a severe hereditary blood disorder caused by a mutation of the beta-globin gene, which results in a substantial reduction in life expectancy. Many studies are focused on various novel therapeutic strategies that include re-activation of the γ-globin gene. Among them, expression therapy caused by the fetal hemoglobin (HbF) at a later age is highly successful. The induction of HbF is one of the dominant genetic modulators of the hematological and clinical characteristics of SCD. In fact, HbF compensates for the abnormal beta chain and has an ameliorant effect on clinical complications. Erythropoiesis is a multi-step process that involves the proliferation and differentiation of a small population of hematopoietic stem cells and is affected by several factors, including signaling pathways, transcription factors, and small non-coding RNAs (miRNAs). miRNAs play a regulatory role through complex networks that control several epigenetic mechanisms as well as the post-transcriptional regulation of multiple genes. In this review, we briefly describe the current understanding of interactions between miRNAs, their molecular targets, and their regulatory effects in HbF induction in SCD.Magnetic resonance (MR) imaging is considered the gold standard for non-invasive evaluation of carotid artery plaque morphology and composition. A number of studies have demonstrated the clinical utility of MR plaque imaging in the risk stratification of carotid atherosclerotic disease, determination of stroke etiology, and identification of surgical and endovascular candidates for carotid revascularization procedures. The MR plaque imaging also provides researchers and clinicians with valuable insights into the pathogenesis, natural history and composition of carotid atherosclerotic disease. link2 Nevertheless, the field of MR plaque imaging is complex, and requires a thorough knowledge of the histologic basis for how various plaque features appear on imaging. link2 This article details the pathogenesis and histology of atherosclerosis, reviews the expected appearance of different plaque components, and describes how MR imaging features may be related to symptomatology or predict future ischemic events. The exact visual mechanisms underpinning the approximate number system are still debated. Recent evidence suggests that numerosity is extracted on segmented visual objects, at least for a moderate numerical range (e.g., less then 100 items), whereas alternative models rather propose that numerosity is derived from low-level features (e.g., power spectrum) of an unsegmented image, independently from the range. Here, to disentangle these accounts, we generated stimuli that were equalized for spatial frequency amplitude spectrum and luminance across sets of moderate range numerosities (e.g., 9-15 dots), while independently manipulating the perceived item segmentation by connecting dots with illusory contours (ICs). In Experiment 1, participants performed a numerical discrimination task, in which they had to select the numerically larger between two stimuli a reference stimulus (always 12 dots) and a test stimulus (from 9 to 15 dots) containing 0, 2 or 4 pairs of dots grouped by ICs lines. In Experiment 2, participants were presented only the test stimulus and performed an estimation task. link3 Results clearly showed that in both experiments participants' performance followed well-known numerical signatures (e.g., distance effect and scalar variability), with numerosity that was underestimated as the illusory connections increased. Crucially, this was found despite spatial frequencies and luminance were kept constant across all the experimental stimuli and these variables were thus uninformative about numerosity. Taken together, these findings indicate that power spectrum in its own cannot explain numerical processing. Rather, visual segmentation mechanisms may be crucial in such processing at least for a moderate numerosity range.Sense of agency refers to the sense that I am controlling an action or outcome, and is associated with a perceived compression of the temporal interval between these events known as intentional binding (IB). Though most IB studies focus on individually performed actions, some reveal the existence of shared implicit agency among joint actors, suggesting the formation of a shared "we" identity that may be sensitive to the intentions and actions of all actors. buy Veliparib While current understanding of this shared agentic identity is far from clear, the present study used variants of the IB paradigm involving truly co-operative actions to investigate (1) the relative strength of individual and joint agency, and (2) whether different complementary roles modulate the sense of agency among joint actors. link3 Experiment 1 compared the strength of agency for actions performed alone or with a partner, by instructing pairs of participants to press a key simultaneously to produce an auditory tone. Here, binding was weaker when performing jointly, but this may have been influenced by the demands of temporally co-ordinating actions. Experiment 2 removed these task demands by implementing joint actions that were not identical but complementary, and assessed the impact of varying roles (moving versus clicking a mouse) on agency. Here, binding was similar for independent and joint actions, and across all roles. These results support the notion of a shared "we" identity, suggesting that the joint sense of agency is as strong as individual agency and represents the intentions and actions of all co-actors.
