Novel nanocomposite hydrogels were successfully prepared by blending and crosslinking sodium alginate (SA), poly(vinyl alcohol) (PVA) and cellulose nanofibers (CNFs) in the presence of a fertilizer formulation containing nitrogen (N), phosphorus (P) and potassium (K). The hydrogels had a macroporous flexible core and a microporous semi- interpenetrating polymer network (IPN) shell. The crystalline nature of the NPK chemicals was retained in the hydrogel nanocomposite network. Furthermore, the SA/CNF/PVA-based hydrogels showed a higher water-retention capacity in both deionized water and mixed soil. The swelling behavior in various physiological pH, salt and alkali solutions exhibited good sensitivity. The NPK release from SA/CNF/NPK and SA/CNF/PVA/NPK hydrogels was controlled by Fickian diffusion in both water and soil based on the Korsmeyer-Peppas release kinetics model (n less then 0.5). Therefore, the prepared hydrogels have the potential for applications in drought-prone and/or fertilizer-loss regions for future development of precision agriculture and horticulture. The aim of the present study was to develop soy protein isolate (SPI) and κ-carrageenan (KC) composite hydrogels as a delivery system for hydrophilic compounds. The pigment of monascus yellow was used as a model. A systematic study was performed to characterize the rheological, textural, microstructural properties and in vitro digestion release profile of monascus yellow of the composite gels. The results of power law modeling, electrophoresis patterns and fourier transform infrared spectroscopy (FTIR) confirmed that non-covalent interactions were involved in the formation of SPI/KC composite hydrogels. Compared to pure κ-carrageenan hydrogels, the incorporation of SPI could promote the formation of tougher, more uniform and compact composite gels with sustained-release property. In addition, the release behaviors of monascus yellow entrapped in the hydrogel network can be well described by the Ritger-Peppas mathematical model. Overall, our study provided a promising strategy to enhance the sustained release performance of hydrogels in digestive conditions. In insects, the cytochrome P450 CYP6B family plays key roles in the detoxification of toxic plant substances. However, the function of CYP6 family genes in degrading plant toxicants in Tribolium castaneum, an extremely destructive global storage pest, have yet to be elucidated. In this study, a T. castaneum CYP gene, TcCYP6BQ7, was characterized. TcCYP6BQ7 expression was significantly induced after exposure to essential oil of the plant Artemisia vulgaris (EOAV). Spatiotemporal expression profiling revealed that TcCYP6BQ7 expression was higher in larval and adult stages of T. castaneum than in other developmental stages, and that TcCYP6BQ7 was predominantly expressed in the brain and hemolymph from the late larval stage. TcCYP6BQ7 silencing by RNA interference increased larvae mortality in response to EOAV from 49.67% to 71.67%, suggesting that this gene is associated with plant toxicant detoxification. Combined results from this study indicate that the CYP6 family gene TcCYP6BQ7 likely plays a pivotal role in influencing the susceptibility of T. castaneum to plant toxicants. These findings may have implications for the development of novel therapeutics to control this agriculturally important pest.Photodynamic inactivation (PDI) is a novel sterilization technology that has proven effective in medicine. This study focused on applying PDI to food packaging, where chitosan (CS) films containing photosensitizing riboflavin (RB) were prepared via solution casting. The CS-RB composite films exhibited good ultraviolet (UV)-barrier properties, and had a visually appealing highly transparent yellow appearance. Scanning electron microscopy (SEM) confirmed even dispersion of RB throughout the CS film. The addition of RB led to improved film characteristics, including the thickness, mechanical properties, solubility, and water barrier properties. The CS-RB5 composite films produced sufficient singlet oxygen under blue LED irradiation for 2 h to inactivate two food-borne pathogens (Listeria monocytogenes and Vibrio parahaemolyticus) and one spoilage bacteria (Shewanella baltica). The CS-RB composite films were assessed as a salmon packaging material, where inhibition of bacterial growth was observed. The film is biodegradable, and has the potential to alleviate the issues associated with the excessive use of petrochemical materials, such as environmental pollution and limited resources. The CS-RB composite films showed potential as a novel environmentally friendly packaging material for shelf-life extension of refrigerated food products.Microsporidia are obligate single-celled eukaryote parasites. Microsporidian infection can cause large economic losses to beneficial insects such as silkworms and honey bees. Identification of resistance biomacromolecules and breeding of transgenic lines resistant to the microsporidian Nosema bombycis are important for disease management. We previously used transcriptome analysis to identify a guanylate binding protein family BmAtlastin-n gene that was significantly upregulated after Nosema bombycis infection, and we determined that the molecule was highly expressed in resistance-related tissues such as the midgut, fat body and the epidermis. selleckchem The transgenic silkworm line overexpressing BmAtlastin-n biomolecules had economic characters similar to those of non-transgenic lines. The transgenic OE-BmAtlastin-n lines had significantly improved survival after microspore infection. We used RT-PCR and H&E staining to show that the number of spores in the transgenic lines was significantly lower than in the control lines. In this study, we identified a BmAtlastin-n macromolecule with resistance to N. bombycis and developed a transgenic line. The results improved understanding of the GBP protein family and provided biomacromolecule material for the treatment and prevention of microsporidia. The recovery of gold from wastewater has received significant attention in the last years due to its high economic value and low availability. A novel chitosan-based adsorbent (CS-GTU) was successfully synthesized by using formaldehyde as a crosslinker between chitosan and guanylthiourea, and applied for selective adsorption of AuIII from an aqueous medium. Through batch experiments, the maximum adsorption capacity of CS-GTU for AuIII could reach up to 695.63 mg/g at pH 5.0, and the adsorption process followed the Pseudo-second-order kinetic and Langmuir isotherm models, indicating that the monolayer chemisorption possibly occurred on the adsorbent surfaces. The adsorption was an enthalpy driven and spontaneous chemical process based on thermodynamic analysis. Furthermore, the adsorbent has demonstrated outstanding selectivity toward AuIII from multi-metallic solutions, and five cycled experiments of adsorption-desorption showed that CS-GTU could be efficiently regenerated. Experimental breakthrough curves were successfully simulated by using the Thomas model, which can fit the experimental data with the correlated curve (R2 > 0.9) well. This improvement in adsorption was a consequence of the complexation and electrostatic attraction of gold ions with the abundant sulfur/nitrogen-containing groups. The CS-GTU beads can be considered as a suitable and efficient adsorbent for gold ions in aqueous solutions. This study is the first to evaluate the effects of Iron oxides (FeOx) species and their decoration on graphene oxide/chitosan (GO/CS) composites for Cr(VI) removal and the possibility of Fe secondary pollution. Results show that Fe(III) is a better decoration material than Fe(II) and decoration through immersion-evaporation shows a higher adsorption capacity of Cr(VI) (Qe) than co-precipitation. Fe2O3-GO/CS as the only eco-friendly composite for enhanced Cr(VI) removal is further used for batch adsorption experiments, characterization, kinetics, isotherms, and thermodynamic studies. It is found that Cr(VI) removal mainly includes electrostatic attraction between Cr(VI) oxyanions and surface -NH3+ and -OH2+, and the adsorbed Cr(VI) partially reduces to Cr(III). Qe increases with the increasing initial Cr(VI) concentration, contact time, and temperature, while decreases with the increasing pH and mass and volume ratio (m/v). The coexisting ions (Cl-, NO3-, SO42-, PO43-, As, Fe, and Pb) can cause an obvious decrease of Qe. The removal efficiency (Re) and Qe are 94.3% and 83.8 mg/g, respectively under the optimal conditions. After five times of regeneration, Re is still as high as 84% and Qe drops about 2.6%. Cr(VI) adsorption is spontaneous and endothermic, which is best fitted with the Sips model, and the fitted maximum Qe is 131.33 mg/g.Advanced glycation end products (AGEs) play an important role in oxidative stress and inflammation, processes implicated in the development and progression of kidney dysfunction. In the present study, we investigated the participation of the pro-oxidant protein thioredoxin-interacting protein (TXNIP) and of epigenetic mechanisms on kidney tissue (in vivo, in non-diabetic rats) and on terminally differentiated glomerular podocytes (in vitro) chronically exposed to AGEs. AGEs induced total kidney and glomerular TXNIP expression and decreased H3K27me3 content. Concomitant treatment with the antioxidant N-acetyl-cysteine (NAC) reversed only the increased TXNIP expression. TXNIP expression positively correlated with proteinuria and negatively correlated with H3K27me3 content. In vitro studies in podocytes showed that 72 h exposure to AGEs decreased nephrin expression and increased Txnip, Nox4, Col4a1, and epithelial-to-mesenchymal transition (EMT) markers (Acta2, Snail1, and Tgfb1). Podocytes treatment with NAC reversed Nox4, Col4a1, Acta2, and Tgfb1 increased expression but did not abrogate the reduced expression of nephrin. MiR-29a expression was downregulated by AGEs in vivo, but not in vitro. In conclusion, treatment of non-diabetic rats with AGEs induced TXNIP expression and decreased the contents of the repressive epigenetic mark H3K27me3 and of miR-29a, potentially driving injury to glomerular filtration barrier and podocytes dysfunction.Identifying miRNAs involved in cancer and devising strategies to control their expression is a new therapeutic approach. Valproic acid (VPA) has attracted a lot of interest in cancer research. We evaluated the impact of VPA on the expression of miR-34a, miR-520h, and their target gene histone deacetylase 1 (HDAC1), as well as their relationship with apoptosis in breast cancer. First, through bioinformatics analyses, the possible target genes of miR-34a and miR-520h and their roles in apoptosis regulation were investigated. Then, miR-34a, miR-520h, and HDAC1 gene expression in tissues of breast cancer patients were determined using the qRT-PCR method. The anticancer impact of VPA on apoptosis and the expression levels of miR-34a, miR-520h, and HDAC1 gene were measured in MCF-7 and MDA-MB-231 cell lines. The bioinformatics analyses indicated that miR-34a and miR-520h might make a unique contribution in regulating the apoptosis pathway. The relative expression of miR-34a and miR-520h significantly decreased in cancer tissues, while the relative expression of HDAC1 increased.
