This work resolves the challenges of the conventional ETL based wide-field microscopic systems viz. variable magnification, field-of-view (FOV), an inconsistent lateral and axial resolution. The proposed combination performs the tuning of the object plane by changing ETL current and effectively modifies the object space numerical aperture using an aperture controller to provide a stationary telecentric image plane. Moreover, with the proposed combination, the lateral resolution of the system is preserved over an axial range of 7.586mm with a magnification error of 2.55 %. The proposed work witnesses the advantages of providing fast axial scanning, extended axial scanning range, autofocusing, invariant magnification, constant FOV, and constant resolution along with a compact optical setup.The objective of this study was to assess protective efficacy of vaccination using CPD-attenuated chimeric PRRSV and Toll like receptor (TLR) agonists (HSP70 c-terminal domain and HSPX) as adjuvants through different inoculation routes. In this study, a chimeric PRRSV composed of two field isolates was synthesized and attenuated by CPD in NSP1 as described in the previous study. The infection of the CPD-attenuated chimeric PRRSV to pigs of 3 weeks-old showed no clinical signs without pathological lesions in necropsy, while it induced improved cross immunity between its parent strains. The TLR agonists were expressed in E. coli and purified to be used. In challenge experiment, pigs of 3 weeks-old were vaccinated using the CPD-attenuated chimeric virus with the prepared TLR agonists through intramuscular or intradermal route, following heterologous challenge after 4 weeks of vaccination. In results, intramuscular or intradermal inoculation of the CPD-attenuated chimeric virus demonstrated excellent protective efficacy against heterologous challenges. Importantly, intradermal inoculation with the TLR agonists enhanced protective effects as shown in the significantly increased level of PRRSV-specific IFN-γ-SCs and cytokines in sera, and the significant reduction of pathological lesion and viral load in lung. This study suggested that the intradermal inoculation of CPD-attenuated chimeric PRRSV plus TLR agonists should be more effective for protection of pigs against diverse PRRS field viruses.Cholesterol 25-hydroxylase (CH25 H), as a host restriction factor, has been reported to take a broad-spectrum antiviral effect. However, the role of CH25H in Senecavirus A (SVA) infection remains unknown. In this study, we first demonstrate that overexpression of CH25H inhibits SVA replication. Consistently, knockdown or knockout of the endogens CH25H promotes SVA infection. Further, the anti-SVA effect of 25-hydroxycholesterol (25HC), which is the product of CH25H, operates via inhibition of viral attachment and replication. On the other hand, the CH25H mutant (CH25H-M) lacking hydroxylase activity still restricts SVA infection, which can selectively interact and degrade SVA 3A protein via the ubiquitin-proteasome manner. Altogether, these results suggest that CH25H has an antiviral function in SVA infection and provides an alternative manner to control SVA.Related to ABI3/VP1 (RAV) transcription factors play important roles in regulating plant growth and stress tolerance, which have been studied in many plant species, but have remained largely unidentified in legumes. To functionally characterize RAV in legumes, MtRAV3 from legume model plant Medicago truncatula was isolated and its function was investigated by overexpressing MtRAV3 in M. truncatula. Expression analysis demonstrated that MtRAV3 was markedly induced by NaCl and polyethylene glycol (PEG). MtRAV3 overexpression enhanced tolerance of transgenic M. truncatula to mannitol, drought and salt stresses, and induced the expression of adversity-related genes, including MtWRKY76, MtMYB61, cold-acclimation specific protein 31 (MtCAS31), alternative oxidase 1 (MtAOX1) and ethylene response factor 1 (MtERF1). There were lower relative electrolyte leakage and higher chlorophyll content of leaves in the MtRAV3 overexpression plants than in wild type plants under both salt and drought stress. MtRAV3 overexpression M. truncatula were featured by some phenotypes of dwarfing, late flowering, more branches, smaller flower and leaf organs. Further investigations showed that the expression levels of DWARF14 (MtD14), CAROTENOID CLEAVAGE DIOXYGENASES 7 (MtCCD7) and GA3-oxidase1 (MtGA3ox1), which related to dwarf and branch phenotype, were obviously reduced, as well as MtGA3ox1' (MTR_1g011580), GA20-oxidase1 (MtGA20ox1), MtGA20ox1' (MTR_1g102070) and GA20-oxidase2 (MtGA20ox2) involved in gibberellins (GAs) pathway. Overall, our results revealed that MtRAV3 exerted an important role in adversity response and plant growth, was a multifunctional gene in M. truncatula, which provided reference for genetic improvement of alfalfa (Medicago sativa).In this study, the effects of three halotolerant rhizobacterial isolates AL, HR, and SB, which are able to grow at a salinity level of 1600 mM NaCl, with multiple plant growth promoting (PGP) traits on some seed and forage quality attributes, and vegetative, reproductive, biochemical and physiological characteristics of wheat plant irrigated with saline water (0, 40, 80, and 160 mM NaCl) were investigated. The ability of halotolerant bacterial isolates to produce PGP traits was affected by salinity levels, depending upon the bacterial isolates. Salinity stress significantly affected the yield, quality, and growth of wheat by modifying the morpho-physiological and biochemical traits of the exposed plants. However, all three bacterial isolates, especially isolate AL, significantly improved the biochemical (an increase in K+/Na+ ratio by 55%, plant P content by 50%, and plant Ca content by 31%), morphological (an increase in stem dry weight by 52%, root dry weight by 44%, spike dry weight by 34%, and grain dry weight by 43%), and physiological (an increase in leaf proline content by 50% and total phenol in leaf by 42%) attributes of wheat and aided the plant to tolerate salinity stress in contrast to un-inoculated plant.
