In this study, we generated a modified SOD1 characterized by an altered capacity to complex Zn. The study investigates the metal-binding dynamics of the enzyme, estimating the stability of a SOD1 protein lacking the appropriate Zn site complexation. Our mutant SOD1 possesses a double amino acid mutation (T135S and K136E) that interferes with the correct Zn site complexation. We found that the protein mutations produce unstable Zn coordination and lower enzymatic activity even when complexed with Cu. Analysis with circular dichroism (CD) spectra on metal titration showed a considerable difference between the two Zn entries in the native dimeric enzyme, and Cu presents a simultaneous entrance in the protein. Otherwise, the mutant T135S,K136E-SOD1 exhibited Zn and Cu complexation instability, being a useful in vitro model to study the SOD1 behavior in AD patients.The reduction of speckle noise by physically changing the pupil of the imaging system, as first envisioned in optical holography, is experimentally applied to a digital holographic microscope (DHM). Furthermore, the swelling ratio and the equilibrium water content of NZCF were compared to the commercial fertilizer and their effect on plant growth was observed. Slow-release studies were carried out for both NZCF and the commercial fertilizer. Through analyzing the records of spike times of grid cells and local entorhinal EEG obtained by Hafting et al., in the spatial navigation experiment, we find that grid cells are, to a large proportion, a kind of broadband-shift neurons, and the positive correlation between grid cell activity and power of low theta and gamma bands was observed. These results have well verified, promoted, and connected many scattered research conclusions, such as the broadband shift phenomenon of hippocampal neurons, the influence of low theta activity on the firing pattern of grid cells, and the positive correlation between single-cell activity and gamma-band activity. Results revealed that fructose has a very significant positive effect on the volumetric DHA content. Meanwhile, its involvement in affecting DHA biosynthetic capacity, though significant, is not very profound. It was also found that when cultivated in fructose medium, SW1 had a less steep log phase compared to that of glucose medium. However, after 48h of cultivation, biomass and lipid accumulation in fructose medium outweighed the other. Volumetric DHA content in fructose medium at 96h was 11% higher than that of glucose medium. link3 Overall, fructose was found to be a more suitable substrate for biomass, lipid and DHA accumulation in SW1 compared to the conventional source, glucose.Brasenia schreberi J.
