Mutations in collagen XII have been recently described to cause modifications in the connective tissue which phenotypic demonstration resembles that of collagen VI related myopathies. We describe a Caucasian 14-year old girl who complained to her doctor about not being able to practice schools' sports due to recurrent clavicle dislocation when trying to throw a ball. In addition, when questioned, she also reported a notion of lower strength in the upper limbs when compared to same age peers. Based on the clinical exam performed, a joint hypermobility with a slight reduction of the muscular strength of the upper limbs diagnosis was noted. Targeted sequencing identified a heterozygous missense mutation in COL12A1 - c.8336G > A (p. Arg2779His). Algorithms developed to predict the effect of the changes on the protein structure and function do not agree on the potential impact of this modification. This case shows the importance to consider collagen XII-related disorders when in presence of patients with an overlapping phenotype with both muscle and connective tissue abnormalities, once mutations in collagen VI have been excluded.
Anxiety is characterized by psychological, physiological, and cognitive complaints. Current treatments have significant limitations, and often overlook any potential benefits to common cognitive symptoms, notably attention and executive function issues. The current study aimed to investigate the use of transcranial direct current stimulation (tDCS) on both common anxiety symptoms and executive function abilities in a college aged sample.

As this is one of the first large scale anxiety studies utilizing tDCS, participants were given a single session of tDCS (anodal, cathodal, or sham) for 20min at 2mA over the left dorsolateral prefrontal cortex (lDLPFC). Participants also completed a series of self-reported anxiety measures and measures of executive functioning (Rey-O Copy and Wisconsin Card Sorting Task).

While there were no differences in anxiety between tDCS groups, a trend was noted demonstrating better performance on Rey-O Copy for the cathodal group. Anxiety increased pre to post for all groups.

Overall, results suggest that while anodal stimulation of the lDLPFC may benefit cognitive abilities for this population, targeting psychological symptoms of anxiety likely requires stimulation over other cortex, possibly right DLPFC. Further, the use of tDCS, whether active or sham, may be distressing to patients.

Zika virus has been associated with congenital anomalies and most ZIKV outbreaks have occurred in endemic areas of DENV. The present study investigated the effects of prior DENV serotype 1 (DENV1) immunity in immunocompetent female Swiss mice on gestational ZIKV infection in offspring. Physical/reflex development, locomotor activity, anxiety, visual acuity, and brain-derived neurotrophic factor (BDNF) levels were evaluated in offspring during infancy and adolescence. Anti-DENV1 and anti-ZIKV antibodies were detected in sera of the progenitors, whereas no ZIKV genomes were detected in the offspring brain. Pups from dams with only DENV1 immunity presented alterations of physical/reflex development. Pups from all infected dams exhibited time-related impairments in locomotor activity and anxiolytic-like behavior. Offspring from DENV/ZIKV-infected dams exhibited impairments in visual acuity during infancy but not during adolescence, which was consistent with morphometric analysis of the optic nerve. Pups from DENV1-, ZIKV-, and DENV/ZIKV-infected dams exhibited a decrease in BDNF levels during infancy and an increase during adolescence in distinct brain regions. In summary, we found no influence of prior DENV1 immunity on gestational ZIKV infection in offspring, with the exception of alterations of early visual parameters, and an increase in BDNF levels in the hippocampus during adolescence.To compare the effects of high- (HF-EMF) and low-frequency electromagnetic fields (LF-EMF) on the proliferation and differentiation of neural stem cells (NSCs). NSCs were obtained from SD rat hippocampus and cultured in suspension and adherent differentiation media. NSCs were exposed to LF-EMF (5 m T, 50 Hz, 30 min daily), HF-EMF (maximum magnetic induction 2.5 T, 40 % MO, 50 Hz, 10 min daily) and no electromagnetic field. At 3 d, cell viability and quantity of NSCs in suspension were detected by CCK-8 assay and cell counting plate. Immunofluorescence staining and qRT-PCR were performed to detect the percentage of Tuj-1 and GFAP-positive NSCs and the expression of Tuj-1 and GFAP mRNA. The P3 NSCs were positive with Nestin and induced NSCs expressed Tuj-1, GFAP and oligodendrocyte markers (MBP). CCK-8 assay and cell counting showed that the OD value and quantity of LF-EMF group were significantly higher than those in other two groups (both P 0.05). Both 50 Hz LF-EMF and HF-EMF can promote the proliferation of NSCs in vitro and LF-EMF can accelerate NSCs to differentiate into neurons.
