The analysis of immune cell signaling is critical for the understanding of the biology and pathology of the immune system, and thus a mandatory step for the development of efficient biomarkers and targeted therapies. Phosflow, which has progressively replaced the traditional western blot approach, relies on flow cytometry to analyze various signaling pathways at a single-cell level. This technique however suffers a lack of sensitivity largely due to the low signal/noise ratio that characterizes cell signaling analysis. In this study, we describe a new technique, which combines the use of biofunctionalized nanospheres (i.e., synthetic particulate antigens, SPAg) to stimulate the immune cells in suspension and imaging flow cytometry to identify homogenously-stimulated cells and quantify the activity of the chosen signaling pathway in selected subcellular regions of interest. Using BCR signaling as model, we demonstrate that SIBERIAN (SPAg-assIsted suB-cEllulaR sIgnaling ANalysis) allows assessing immune cell signaling with unprecedented sensitivity and specificity.
This clinical investigation examined the effects of brushing with a test toothpaste containing natural ingredients, that is, clove (Syzgium Aromaticum), aloe vera (Aloe Barbadensis), amla (Emblica Officinalis), neem (Azadirachta Indica), tulsi (Ocimum Basillicum) and honey (from Apis Mellifera) along with zinc salts and fluoride in comparison with a dentifrice formulated with fluoride alone on dental plaque and gingivitis over a 6-month period.

One hundred and eighty (180) subjects with mean plaque index scores greater than 1.5 (Turesky Modified Quigley Hein Plaque Index) and gingival index scores >1.0 (Loe and Silness gingival index) in good general health and who met other study criteria were enrolled in the clinical study. Subjects were evaluated for plaque and gingival index scores and were randomly assigned to perform twice-daily oral hygiene with either the test or the control toothpaste for the next 6months. One hundred and fifty (150) subjects [test toothpaste (n=72) and control toothpaste (n=7control toothpaste as showed by ANCOVA (p<0.001). In comparison with the control, the test demonstrated reductions in 23.5%, 25.6%, and 73.3% for dental plaque, gingival index and bleeding index outcomes, respectively, at the final visit. Reductions in the frequencies of sites with higher clinical scores were noted over the study with more sites registering improvements in the test group than in the control group.

A multitude of stabilizers exists, each of which may be hard to extract, be difficult if not impossible to separate from other stabilizers or necessitate very selected and time-consuming intermediate stages for separation. Certain polymer matrices even pose additional challenges, such as polyolefins being only soluble at elevated temperatures. One of the most well-established approaches for the extraction of stabilizers is Soxhlet extraction. However, even this highly successful approach shows only limited success with regard to the extraction of the ever more relevant oligomeric stabilizers or the extraction of multiple stabilizers in a one-shot approach. Moreover, performing Soxhlet extractions often necessitates ≥24 h. For these reasons, alternative approaches for the extraction of stabilizers from polymers are highly sought after. An approach with enormous potential is solid-phase extraction, which allows the selective retention and enrichment of stabilizers. Herein, the very first application of high-temperature solid-phase extraction for the extraction of stabilizers from polyolefin matrices is described; as with other extraction techniques, the identification and quantification of the stabilizers is then allowed. At temperatures of 140-160°C, it was possible to adsorb common polyolefin stabilizers selectively on a silica solid phase from their polyolefin matrix. To predict high-temperature solid-phase extraction test conditions, first LC tests are necessary, offering an elegant approach for the separation of polyolefins from oligomeric stabilizers, which was not achievable until now.Interspecific differences in sensitivity of the Antarctic moss Sanionia uncinata from King George Island (KGI) and James Ross Island (JRI) to photoinhibitory treatment were studied in laboratory conditions using chlorophyll fluorescence techniques. Slow (Kautsky) and fast (OJIP) kinetics were used for the measurements. Samples were exposed to a short-term (60 min) photoinhibitory treatment (PIT, 2000 μmol·m-2 ·s-1 PAR). The photoinhibitory treatment (PIT) led to photoinhibition which was indicated by the decrease in FV /FM and ΦPSII in KGI but not in JRI samples. However, this decrease was small and full recovery was reached 90 min after PIT termination. Non-photochemical quenching (NPQ) was activated during the PIT, and rapidly relaxed during recovery. Early stages of photoinhibition showed a drop in FV /FM and ΦPSII to minimum values within the first 10 s of the PIT, with their subsequent increase apparent within fast (0-5 min PIT) and slow (5-50 min PIT) phases of adjustment. The PIT caused a decrease in the performance index (Pi_Abs), photosynthetic electron transport per reaction centre (RC) (ET0 /RC). The PIT induced an increase in thermal dissipation per RC (DI0 /RC), effectivity of thermal dissipation (Phi_D0 ), absorption per RC (ABS/RC) and trapping rate per RC (TR0 /RC). In conclusion, PIT led to only slight photoinhibition followed by fast recovery in S. uncinata from KGI and JRI, since FV /FM and ΦPSII returned to pre-photoinhibitory conditions. Therefore, S. uncinata might be considered resistant to photoinhibition even in the wet state. The KGI samples showed higher resistance to photoinhibition than the JRI samples.
Pronounced asymmetric changes in ocular globe size during eye development have been observed in a number of species ranging from humans to lizards. In contrast, largely symmetric changes in globe size have been described for other species like rodents. We propose that asymmetric changes in the three-dimensional structure of the developing eye correlate with the types of retinal remodeling needed to produce areas of high photoreceptor density. To test this idea, we systematically examined three-dimensional aspects of globe size as a function of eye development in the bifoveated brown anole, Anolis sagrei.

During embryonic development, the anole eye undergoes dynamic changes in ocular shape. Initially spherical, the eye elongates in the presumptive foveal regions of the retina and then proceeds through a period of retraction that returns the eye to its spherical shape. During this period of retraction, pit formation and photoreceptor cell packing are observed. We found a similar pattern of elongation and retraction associated with the single fovea of the veiled chameleon, Chamaeleo calyptratus.

These results, together with those reported for other foveated species, support the idea that areas of high photoreceptor packing occur in regions where the ocular globe asymmetrically elongates and retracts during development.

Emergency department (ED) overcrowding is a problem for the delivery of adequate and timely emergency care. To improve patient flow and the admission process, the quick prediction of a patient's need for admission is crucial. We aimed to investigate the variables associated with hospitalisation after an ED visit, with a particular focus on the variables related to medication.

This prospective study was conducted from 2011 to 2018 in subacute medical ED of a French University Hospital. Specialised EDs (paediatric, gynaecologic, head and neck and psychiatric) and the outpatient unit of the ED were not included. Participation in this study was proposed to all adult patients who underwent a medication history interview with a pharmacist. Pharmacists conducted structured interviews for the completion of the medication history and the detection of adverse drug events (ADE). Relations between patient characteristics and hospitalisation were analysed using logistic regression.

Anti-tuberculosis drugs remain as an important cause of drug-induced liver injury (DILI) worldwide. Adverse drug reactions reduce the effectiveness of treatment. We aimed to determine the incidence and risk factors associated with anti-tuberculosis DILI (ATDILI).

Using established criteria and causality assessment methods, risk factors for ATDILI were identified in a contemporary cohort and validated in another cohort prospectively. Independent determinants of ATDILI were identified using Cox regression analysis.

In the derivation cohort (n=3155), 170 (5.4%) developed ATDILI of which 27 (15.9%) developed jaundice; 9(5.3%) developed acute liver failure (ALF) and 3 died. Among HBsAg positive patients, 11/27 (40.7%) of ATDILI developed after 3months of starting treatment. In addition, of 218 (6.9%) who developed raised alanine transferase (ALT) levels ≥3 times upper limit normal, 193 (88.5%) resolved and 25 (11.4%) progressed to DILI. Age (HR=1.014, 95% CI 1.005-1.023), baseline ALT (HR=1.014, 95% CI 1.003DILI are low. Age, baseline ALT, haemoglobin and HBsAg positivity are risk factors for the development of DILI and these inform monitoring and management of these patients.
The efficacy and safety of a fluid-filled intragastric balloon (IGB) for weight loss in cirrhotic patients on the liver-transplant waitlist is unknown.

We enrolled stable compensated waitlisted patients with BMI >35 for IGB placement endoscopically for a maximum of 6 months.

8 patients (7 male), 56 ± 4.6 years with MELDNa 14.1 ± 3.4 experienced weight reduction (146 ± 22.2kg vs.127 ± 21.6kg, p=0.005 with IGB in place and 130±24.6kg, p=0.014 at 6months), with TWBL12.2% ± 8.8% with IGB in place and 10.9± 8.9% ITT at 6 months. Body fat decreased from 48.6% ± 5.8% to 40.6% ± 6.4% (p=0.001) and lean mass increased from 51.3% ± 6% to 59.4% ± 6.4% (p=0.001). No change in MELDNa occurred (p=0.77). Early balloon retrieval was due to accommodative symptoms (n=2) and liver decompensation (n=1). Mallory-Weiss tears (n=3) but no portal hypertensive bleeding occurred. Liver decompensation and/or HCC developed in 3 patients. 4 with liver transplantation ±sleeve gastrectomy maintained overall weight loss. TP0427736 inhibitor 2/4 without transplant experienced weight regain.
