A regulatory ecosystem that engages with a broad range of stakeholders will mean that expertise in the ever-expanding field of regulatory science filters into teaching and research in a symbiotic way. In this way capacity development interventions will be a collaborative approach between the learning context (academic and training institutions) and the performance context (regulatory agencies and industry), which will ultimately best serve the patients. Monitoring and evaluation of capacity development interventions will be essential to show value of investments and ultimately guide continued funding and sustainability. This paper reviews the skills and human capacity gaps, reports on regulatory assessment pathways used in Ghana, South Africa and Zimbabwe and outlines a staged tactical approach for Africa that builds on previous efforts to strengthen African regulatory ecosystems.
Elevated epidermal interleukin (IL)-36 expression distinguishes psoriasis from eczematous dermatitis, but other psoriasiform dermatitides (PDs) have not been thoroughly investigated for IL-36 expression. In this study, we assess the IL-36 staining pattern (IL36-SP) in psoriasis variants and other PDs including lichen simplex chronicus (LSC), prurigo nodularis (PN), lichen planus (LP), tinea, pityriasis rubra pilaris (PRP), mycosis fungoides (MF), pemphigus foliaceus (PF), acute generalized exanthematous pustulosis (AGEP), impetigo (IMP), and syphilis (SY).
IL-36 immunostaining was performed on 307 cases of psoriasis and various PDs. IL36-SP in the upper epidermis was graded on a scale of 0-4.
High IL36-SP occurred in all variants of psoriasis, as well as in AGEP, PRP, PN, tinea, IMP, and LP (P > 0.05). SY, PF, LSC, and MF showed a lower IL36-SP (P≤ 0.05) compared with psoriasis.

We manipulated born-free White South Africans' experience of accessibility of memories of past ingroup wrongdoings and their current experiences of status loss. The parasite solely depends on this pathway for the synthesis of nucleotides due to the absence of the de novo pathway. This study intends to identify putative inhibitors towards Trypanosoma cruzi HGPRT (TcHGPRT). Initial virtual screening was performed with substructures of phosphoribosyl pyrophosphate (PRPP), an original substrate of HGPRT. Twenty compounds that had greater binding energy than the substrate was treated as hits and was further screened and narrowed down through induced fit docking which resulted in top five compounds which was distinguished into two groups based on the ligand occupancy within the PRPP binding site of TcHGPRT. Group-I compounds (PubChem CID 130316561 and 134978234) are analogous to PRPP structure with greater occupancy, were preferred over Group-II compounds which had lesser occupancy than the substrate. To evaluate the effect of nutritional supplementation on height, weight and body composition in short and lean male preadolescents.
A randomised, double-blinded, placebo-controlled trial of nutritional supplementation of short and lean prepubertal 10-14.5-year-old boys. Primary outcomes included Δheight-SDS and Δweight-SDS. Secondary outcomes included changes in body composition and BMI-SDS.
Of 160 boys enrolled, 126 (80%) completed 6months' intervention. Baseline age, height-SDS, weight-SDS, BMI-SDS, body composition and dietary intake were similar in the formula and placebo groups. 'Good' formula consumers (intake of ≥50% of the recommended dose, n=30) gained significantly more in weight-SDS, BMI-SDS, fat-free-mass and muscle mass (p<0.05) than did 'poor' consumers (n=35) and the placebo group (n=61). Only in the formula group, positive dose-response correlations were found between consumption of the formula and changes in the outcome parameters examined, including Δheight-SDS (r=0.301, p=0.015). Boys aged >11.4years who were 'good' formula consumers maintained their Δheight-SDS, while Δheight-SDS declined in 'poor' consumers and the placebo group of the same age (p=0.033).

KOS 953 Herein, a chromium-based metal-organic framework, namely, TYUT-96Cr, is reported, which has an unprecedented N2 capture capacity of 37.46 cm3 cm-3 and N2 /O2 (595, v/v) selectivity up to 26.95 (298 K and 1 bar), thus setting new benchmarks for all reported metal-organic frameworks and commercially used ones (Li-LSX and 13X). Breakthrough experiments reveal that N2 can be directly extracted from various N2 /O2 (7921, 5050, 595, and 199, v/v) mixtures by this material, affording a record-high O2 -production scale with 99.99% purity. Density functional theory calculations and in situ infrared spectroscopy studies demonstrate that the high-density open Cr (III) sites in TYUT-96Cr can behave as effective Lewis acidic sites, thus resulting in a strong affinity toward N2 . The high N2 adsorption selectivity, exceptional separation performance, and ultrahigh structural stability render this porous material with great potential for this important industrial application.Though it is well established that species composition affects ecosystem function, the way in which species combine to control overall ecosystem functioning is still debated. In experimental mesocosms, we planted three functionally distinct dry-heath species in varying proportions and measured multiple ecosystem properties related to nutrient cycling and carbon storage (hereafter functions). Overall ecosystem functioning was described as the main axes of variation in ecosystem functioning (functional space) and the proportion of ecosystem functions at high levels; for example, fast carbon and nutrient cycling (cluster-based multifunctionality). The first functional space axis, related to nitrogen availability, was driven by plant species abundance, particularly that of legumes, which strongly affected many individual functions. The second, related to total plant biomass and woodiness, was mostly driven by the abundance of dwarf shrubs. Similarly, cluster-based multifunctionality was related to the initial abundance of all species, but particularly the legume. Interactions between species also affected ecosystem multifunctionality, but these effects were smaller in magnitude. These results indicate that species interactions could play a secondary role to species abundance and identity in driving the overall ecosystem functioning of heathlands, but also that axes of variation in functional space are clearly linked to plant functional composition.With more than 200 million cases and 400,000 related deaths, malaria remains one of the deadliest infectious diseases of 2021. Unfortunately, despite the availability of efficient treatments, we have observed an increase in people infected with malaria since 2015 (from 211 million in 2015 to 229 million in 2019). This trend could partially be due to the development of resistance to all the current drugs. Therefore, there is an urgent need for new alternatives. We have, thus, selected common natural scaffolds, polyhydroxybenzoic acids, and synthesized a library of derivatives to better understand the structure-activity relationships explaining their antiplasmodial effect. Only gallic acid derivatives showed a noticeable potential for further developments. Indeed, they showed a selective inhibitory effect on Plasmodium (IC50 ~20 µM, SI > 5) often associated with interesting water solubility. Moreover, this has confirmed the critical importance of free phenolic functions (pyrogallol moiety) for the antimalarial effect. Frozen plasma (FP) is thawed prior to transfusion and stored for ≤5 days at 1-6°C. The effect of temperature excursions on the quality and safety of thawed plasma during 5-day storage was determined.
Four plasma units were pooled, split and stored at ≤-18°C for ≤90 days. Test units T30 and T60 were exposed to 20-24°C (room temperature [RT]) for 30 or 60 min, respectively, on days 0 and 2 of storage. Negative and positive control units remained refrigerated or at RT for 5 days, respectively. On Day 5, test units were exposed once to RT for 5h. Quality assays included stability of coagulation factors FV, FVII, FVIII, fibrinogen and prothrombin time. Bacterial growth was performed in units inoculated with ~1CFU/ml or ~100 CFU/ml of Serratia liquefaciens, Pseudomonas putida, Pseudomonas aeruginosa or Staphylococcus epidermidis on Day 0.
Testing results of all quality parameters were comparable between T30 and T60 units (p < 0.05). Serratia liquefaciens proliferated in cold-stored plasma, while P. putida showed variable viability.
