Fluorescence and specific UV absorbance at 254 nm (SUVA 254) data showed that fractions of DOC with higher SUVA 254 values (terrestrial-like fractions) were better removed by secondary IEX than those with lower SUVA 254 values (aquatic/microbial-like fractions). Scanning electron microscopy showed that biofilms on non-regenerated resins covered 5-15% of the resin surface and are composed of numerous species of bacteria with varying functions, with some protozoa present.Effects of calcium ions and polyaluminum chloride (PACl) on membrane fouling in coagulation-ultrafiltration (UF) process were investigated in this study. Filtration tests demonstrated three interesting filtration behaviors 1) high specific filtration resistance (SFR) of alginate solution with low CaCl2 or PACl addition (e. g. 3.51×1015 m·kg -1 under the condition of 1.5 mM CaCl2 addition); 2) unimodal pattern of alginate SFR with PACl or CaCl2 addition alone; 3) synergistic effects between CaCl2 and PACl on alginate SFR. It was found that, the foulant morphological changes driven by the thermodynamic mechanisms based on Flory-Huggins lattice theory take the critical roles in these filtration behaviors. Density functional theory (DFT) calculations showed that initial coordination of Ca2+ and Al3+ ions with alginates tended to form tetrahedron geometry and geometry of coordinating three terminal carboxyl groups, respectively, which facilitated to elongate the alginate chains (without clustering the flocs) and form more stable gel, increasing SFR. Improving Ca2+ and Al3+ dosages triggered transition to other geometries for clustering polymeric network and flocculation, reducing SFR. Due to the higher binding affinity of Ca2+ over Al3+, Ca2+ and Al3+ sequentially take roles of enlarging polymeric network and clustering the coordination compounds, and then facilitate to form large size flocs and reduce SFR, causing the synergistic effects between CaCl2 and PACl additions. The proposed thermodynamic mechanisms satisfactorily explained these interesting fouling behaviors, allowing to further optimize coagulation-UF process.Human cathepsin B (CatB) is an important biological target in cancer therapy. In this work, we performed a knowledge-based design approach and the synthesis of a new set of 19 peptide-like nitrile-based cathepsin inhibitors. Reported compounds were assayed against a panel of human cysteine proteases CatB, CatL, CatK, and CatS. Three compounds (7h, 7i, and 7j) displayed nanomolar inhibition of CatB and selectivity over CatK and CatL. The selectivity was achieved by using the combination of a para biphenyl ring at P3, halogenated phenylalanine in P2 and Thr-O-Bz group at P1. Likewise, compounds 7i and 7j showed selective CatB inhibition among the panel of enzymes studied. We have also described a successful example of bioisosteric replacement of the amide bond for a sulfonamide one [7e → 6b], where we observed an increase in affinity and selectivity for CatB while lowering the compound lipophilicity (ilogP). Our knowledge-based design approach and the respective structure-activity relationships provide insights into the specific ligand-target interactions for therapeutically relevant cathepsins.ATP-binding cassette (ABC) transporters are membrane-bound proteins involved in exporting various xenobiotic compounds from living cells. Bivalve mollusks can accumulate large amounts of paralytic shellfish toxins (PSTs) from marine dinoflagellates. For aquatic invertebrates, the importance of ABC proteins in multi-xenobiotic resistance has been demonstrated, however, the systematic identification of ABC transporters is very limited. In this study, 64 and 67 ABC genes containing all eight described subfamilies (A to H) were identified in Yesso scallop (Patinopecten yessoensis) and Zhikong scallop (Chlamys farreri), respectively, with massive gene expansion being observed in the ABCC and ABCG subfamilies. The kidney harbored more specifically expressed ABC genes than other organs/tissues, most of which belonged to ABCB, ABCC, and ABCG subfamilies. After feeding the scallops with PST-producing dinoflagellates, the expression of scallop ABC genes in the kidney was regulated in toxin- and species-dependent manners. In total, 20 and 24 ABC genes in Zhikong scallop (CfABCs) were induced after exposure to Alexandrium minutum and A. catenella, with the up-regulated members from both ABCC and ABCG subfamilies mainly showing acute and chronic induction by A. minutum and A. catenella, respectively, while the up-regulated CfABCBs mainly showing chronic induction by both dinoflagellates. In Yesso scallop, only eight ABC genes (PyABCs) were regulated after A. selleck chemicals llc catenella exposure, and all the five up-regulated PyABCs were acutely induced. Our findings imply the functional diversity of scallop ABC genes in coping with PST accumulation, which may contribute to the lineage-specific adaptation of scallops for dealing with algal toxins challenge.Sex differences in brain structure in children with disruptive behavior disorders (DBD) remain poorly understood. This study examined sex differences in gray matter volume in children with DBD in a priori regions-of-interest implicated in the pathophysiology of disruptive behavior. We then conducted a whole-brain analysis of cortical thickness to examine sex differences in regions not included in our hypothesis. Exploratory analyses investigated unique associations between structure, and dimensional measures of severity of disruptive behavior and callous-unemotional traits. This cross-sectional study included 88 children with DBD (30 females) aged 8-16 years and 50 healthy controls (20 females). Structural MRI data were analyzed using surface-based morphometry to test for interactions between sex and group. Multiple-regression analyses tested for sex-specific associations between structure, callous-unemotional traits, and disruptive behavior severity. Boys with DBD showed reduced gray matter volume in the left ventromedial prefrontal cortex (vmPFC) and reduced cortical thickness in the supramarginal gyrus, but not girls compared to respective controls. Dimensional analyses revealed associations between sex, callous-unemotional traits, and disruptive behavior for amygdala and vmPFC volume, and ventrolateral prefrontal cortex cortical thickness. Sex-specific differences in prefrontal structures involved in emotion regulation may support identification of neural biomarkers of disruptive behavior to inform target-based treatments.
