Fibrosis is the pathophysiological hallmark of progressive chronic kidney disease (CKD). The kidney is a highly metabolically active organ, and it has been suggested that disruption in its metabolism leads to renal fibrosis. We developed a longitudinal mouse model of acute kidney injury leading to CKD and an in vitro model of epithelial to mesenchymal transition to study changes in metabolism, inflammation, and fibrosis. Using transcriptomics, metabolic modeling, and serum metabolomics, we observed sustained fatty acid metabolic dysfunction in the mouse model from early to late stages of CKD. Increased fatty acid biosynthesis and downregulation of catabolic pathways for triglycerides and diacylglycerides were associated with a marked increase in these lipids in the serum. We therefore suggest that the kidney may be the source of the abnormal lipid profile seen in patients with CKD, which may provide insights into the association between CKD and cardiovascular disease.Reaching the climate targets set in the Paris Agreement on climate change requires decarbonizing all parts of the global economy. The electrification of industry processes-and more specifically, electrosynthesis (ES)-is an important decarbonization mechanism. To tap into this mechanism's potential and accelerate the decarbonization of these processes, I argue that public policy needs to perform two tasks. First, energy policy needs to enable the provision of CO2 emissions-free baseload electricity. Second, innovation policy needs to accelerate cost reductions for ES. Here, I discuss why this is the case, what the challenges are, how policy makers can address them, and how political ambition can be increased.Human mitochondrial NAD(P)+-dependent malic enzyme (ME2) is well recognized to associate with cancer cell metabolism, and the single nucleotide variants (SNVs) of ME2 may play a role in enzyme regulation. Here we reported that the SNVs of ME2 occurring in the allosteric sites lead to inactivation or overactivation of ME2. Two ME2-SNVs, ME2_R67Q and ME2-R484W, that demonstrated inactivating or overactivating enzyme activities of ME2, respectively, have different impact toward the cells. The cells with overactivating SNV enzyme, ME2_R484W, grow more rapidly and are more resistant to cellular senescence than the cells with wild-type or inactivating SNV enzyme, ME2_R67Q. Crystal structures of these two ME2-SNVs reveal that ME2_R67Q was an inactivating "dead form," and ME2_R484W was an overactivating "closed form" of the enzyme. The resolved ME2-SNV structures provide a molecular basis to explain the abnormal kinetic properties of these SNV enzymes.
Contraception plays a pivotal role in most women's lives, from teenage years to the menopause. Contraception and sexual wellbeing are closely related. Ideally, women should be able to access contraception and discuss issues concerning their sexual life during a contraceptive counselling session. Previously, only doctors conducted contraceptive consultations. Increasingly, other healthcare workers are providing contraceptive care. The aim of this study is to explore women's experiences and opinions related to contraceptive counselling.

An electronic questionnaire was distributed in 2017-2018. The texts of 308 women's written responses to open-ended questions were analyzed using systematic text-condensation.

The analysis resulted in four themes 1) Women-centered care, 2) Side-effects of hormonal contraceptives, 3) Non-hormonal methods and male involvement, and 4) Counsellors' professional background. Women wished for a consultation that would lead to the best choice of contraception for them, taking into this area need to be made more widely known.Stem cells are currently being used in many clinical trials for regenerative purposes. Here, we examine the interaction between sym-2, the gene encoding an ortholog of hnRNPF and hnRNPH, and hrpa-1, the ortholog of of the gene encoding hnRNPA2, which when mutated causes multisystem proteinopathy. We find that after 22 hours, but not 4 hours, of paraquat-induced oxidative stress, sym-2(mn617) has a mild glutamatergic neurodegeneration phenotype. Interestingly, this defect is rescued by expression of chimeric WT hrpa-1, but not mutant. Thus, we identify a curious genetic interaction between sym-2 and hrpa-1.[This corrects the article DOI 10.1021/acsabm.9b00343.].Sequences of phonologically similar words are more difficult to remember than phonologically distinct sequences. This study investigated whether this difficulty arises in the acoustic similarity of auditory stimuli or in the corresponding phonological labels in memory. Participants reconstructed sequences of words which were degraded with a vocoder. We manipulated the phonological similarity of response options across two groups. One group was trained to map stimulus words onto phonologically similar response labels which matched the recorded word; the other group was trained to map words onto a set of plausible responses which were mismatched from the original recordings but were selected to have less phonological overlap. Participants trained on the matched responses were able to learn responses with less training and recall sequences more accurately than participants trained on the mismatched responses, even though the mismatched responses were more phonologically distinct from one another and participants were unaware of the mismatch. The relative difficulty of recalling items in the correct position was the same across both sets of response labels. Mismatched responses impaired recall accuracy across all positions except the final item in each list. These results are consistent with the idea that increased difficulty of mapping acoustic stimuli onto phonological forms impairs serial recall. Increased mapping difficulty could impair retention of memoranda and impede consolidation into phonological forms, which would impair recall in adverse listening conditions.This study evaluated the effects of 2 supplement dry matter (DM) amounts, with or without calcium salts of fatty acids (CSFA), on growth performance, supplement and water intake behavior of grazing beef bulls. On day 0, 32 Nellore bulls were ranked by initial body weight (BW; 318 ± 11.2 kg), and then, randomly assigned to treatments (n = 8 bulls/treatment), in a 2 × 2 factorial design, which consisted of energy-based supplement DM amount of 0.3 (SP03) or 1.0 (SP1) % of BW with (+) or without (-) CSFA fortification (90 to 100 g/bull daily). Despite negative swab on day 10, he developed ACC on day 17. With antibiotics and supportive care, patient showed full recovery without any surgical intervention. The empirical results show that INARPQX(1) process is an important process to model over-dispersed time series of counts and can be used to predict the number of earthquakes with a magnitude greater than four.In recent years, therapy with immune modulating antibodies, termed immune checkpoint blockade (ICB), has revolutionized the treatment of advanced metastatic melanoma, yielding long-lasting clinical responses in a subgroup of patients. But despite this remarkable progress, resistance to therapy represents a major clinical challenge. ICB efficacy is critically dependent on cytotoxic CD8+ T cells targeting tumor cells in an HLA class I (HLA-I) antigen-dependent manner. Transcriptional suppression of the HLA-I antigen processing and presentation machinery (HLA-I APM) in melanoma cells leads to HLA-I-low/-negative tumor cell phenotypes escaping CD8+ T cell recognition and contributing to ICB resistance. In general, HLA-I-low/-negative tumor cells can be re-sensitized to T cells by interferons (IFN), augmenting HLA-I APM expression. However, this mechanism fails when melanoma cells acquire resistance to IFN, which recently turned out as a key resistance mechanism in ICB, besides HLA-I APM suppression. Seeking for a strategy to overcome these barriers, we identified a novel mechanism that restores HLA-I antigen presentation in tumor cells independent of IFN (Such et al. (2020) J Clin Invest, doi 10.1172/JCI131572). We demonstrated that tumor cell-intrinsic activation of the cytosolic innate immunoreceptor RIG-I by its synthetic ligand 3pRNA overcomes transcriptional HLA-I APM suppression in patient-derived IFN-resistant melanoma cells. De novo HLA-I APM expression is IRF1/IRF3-dependent and re-sensitizes melanoma cells to autologous cytotoxic CD8+ T cells. Notably, synthetic RIG-I ligands and ICB synergize in T cell activation, suggesting combinational therapy could be an efficient strategy to improve patient outcomes in melanoma.
