The self-perceptions of adolescents and young adults (AYAs) after cancer treatment are not well understood. As part of a two-arm, mixed-methods pilot randomized controlled trial (RCT), this qualitative sub-study explored AYAs' self-perceptions after cancer treatment and investigated how physical activity (PA) may contribute to their self-perceptions.

Data were collected from 16 AYAs who completed cancer treatment and who participated in a two-arm, mixed-methods pilot RCT. Recruited AYAs were randomized to a 12-week PA intervention or a wait-list control group, and semi-structured interviews were conducted at baseline (pre-randomization) and 12 weeks later (post-intervention or post-waiting period) to elicit discussions on self-perceptions and PA. Data were analyzed thematically using inductive and deductive approaches. The exercise and self-esteem model (EXSEM) was the theoretical lens for the deductive analysis.

Data were organized into four unified main themes (1) I came out on top and am (re)discovering myself, (2) Comparison to my past self and others induces negative feelings, (3) My changed body brings me down, but it does not rule my life, and (4) My previous experience with PA informs my expectations for my future PA, and two themes contingent on group allocation (5) Participating in a PA program made me feel better about myself, and (6) I did not notice any changes while waiting for the PA program, but I am anticipating support.

AYAs' self-perceptions are amenable to change, positively and negatively valenced, and influenced by PA. Although the EXSEM captured intrapersonal factors related to AYAs' self-perceptions after cancer treatment, interpersonal and contextual factors beyond the EXSEM also shaped their self-perceptions.

In this context, multiple risk factors play an important role for the diagnostic and therapeutic approaches as well as for the interdisciplinary multimodal pain therapy developed for this purpose. The Global Year of the International Association for the Study of Pain (IASP) 2020 addressed the prevention of (chronic) pain, a welcome opportunity to provide a short review of the evidence for and clinical experiences with timely diagnostic and therapeutic options and to summarize the current framework conditions and scientific recommendations for Germany. Head and neck squamous cell carcinoma (HNSCC) are a highly aggressive tumor with an extremely poor prognosis. Thus, we aimed to develop and validate a robust prognostic signature that can estimate the prognosis for HNSCC.

Data on gene expressions and clinical were downloaded from TCGA and GEO database. To develop the best prognosis signature, a LASSO Cox Regression model was employed. Survival differences were evaluated by log-rank test, while multivariate analysis was performed by a Cox proportional hazards model.

A 17-HRGPs composed of 24 unique genes was constructed, which was significantly related to OS. In the TCGA and GEO datasets, patients in the high hypoxia risk group have a poor prognosis (TCGA P < 0.001, GEO P < 0.05). After adjusting for other clinicopathological parameters, the 17-HRGP signature was independent prognostic factors in patients with HNSCC (P < 0.05). Head and neck squamous cell carcinoma (HNSCC) is a heterogeneous disease characterized by different molecular subtypes with different prognosis and response to treatment. Therefore, the aim of this study was to construct reliable gene signatures based on immune checkpoint-related genes to distinguish between subgroups of patients with different risks.

We obtained the HNSCC data from The Cancer Genome Atlas (TCGA) database and Gene Expression Omnibus (GEO) as a training set and the external validation set, respectively. First, differentially expressed immune checkpoint-related genes in tumor tissues and normal tissues were determined, and the potential functions of differential genes were explored through GO function annotation and KEGG pathway enrichment analysis. Using univariate Cox regression analysis, 20 immune checkpoint-related genes in HNSCC patients were significantly associated with overall survival (OS). Subsequently, seven genes were selected by multivariate Cox regression analysis to create a respectively; and in GSE41613 were 0.748, 0.719, and 0.727, respectively. The calibration chart curve showed that the nomogram has strong clinical performance in the prognosis prediction of HNSCC patients.

We questioned that whether LCN2 might also associate exogenous iron chelator as does in inherent way and the association may influence their respective function. To address this issue, we investigated the effect of LCN2 on action of DpdtC (2,2'-dipyridine ketone hydrazone dithiocarbamte), an iron chelator in proliferation and metastasis-related gene expression. The results showed that exogenous LCN2 and DpdtC could inhibit growth of HepG2 cells, while the combination treatment enhanced their inhibitory effect both in proliferation and colony formation. This encouraged us to investigate the effect of the interaction on metastasis-related gene expression. The results revealed that both LCN2 and DpdtC impaired the wound healing of HepG2, but the inhibitory effect of DpdtC was significantly enhanced upon association with LCN2. Undergoing epithelium-mesenchymal transition (EMT) is a crucial step for cancer metastasis, LCN2 and DpdtC had opposite effects on EMT markers, the binding of DpdtC to LCN2 significantly weakened the regulation of it (or its iron chelate) on EMT markers. To insight into the interaction between LCN2 and DpdtC-iron, fluorescence titration and molecular docking were performed to obtain the association constant (~ 104 M-1) and thermodynamic parameters (ΔG = - 26.10 kJ/mol).
