Long-term analyses included 3072 risankizumab-treated patients (exposure 7927.2 PY); median (range, excluding 4 outliers) treatment duration was 2.9 years (2 days-5.9 years). Exposure-adjusted adverse event (AE) rates did not increase with long-term treatment (318.0 vs 170.9 events/100 PY for short- and long-term analyses). With long-term risankizumab treatment, rates of serious AEs were 7.8/100 PY, serious infections, 1.2/100 PY, non-melanoma skin cancer (NMSC), 0.7/100 PY, malignant tumors excluding NMSC, 0.5/100 PY, and adjudicated major adverse cardiovascular events, 0.3/100 PY, with no important identified risks. Limitations include that study inclusion/exclusion criteria varied and three studies enrolled ≤50 patients.

In the last decades, the amount of emergency department (ED) transfers of nursing home (NH) residents has disproportionally increased in western countries. The main role of emergency medical dispatcher (EMD) among this population is to refer residents to EDs in the most appropriate way. The aim of this study was to assess risk factors of inappropriate transfers from NH to ED after EMD request.

This research was a secondary analysis of a prospective observational multicenter study carried out in 17 EDs entitled FINE aimed to assess potentially inappropriate transfer prevalence among this population. Inappropriate transfers were determined in the FINE study threw a standardized approach by a unique expert team.

Overall, 572/1037 (55.2%) of residents were transferred to the ED after an EMD's decision. Among them, 92/572 (16.1%) transfers was defined as inappropriate. The average age was 87.3years old (SD = 0.3). The main reason for ED transfer were falls (217/572, 37.9%). In multivariate analysis, the presence of a Special Care Unit in NH was significantly associated with a high rate of inappropriate transfer (OR 1.78; 95 CI [1.07-2.93]; p = 0.02) whereas a medical examination by a general practitioner before the transfer (OR 0.55; 95 CI [0.33-0.83]; p = 0.02) and a prompt access to psychiatric advice (OR 0.54; 95 CI [0.33-0.84]; p = 0.007) were associated with a low rate of potentially inappropriate transfer.

We enrolled and sampled NH residents and community-dwelling healthcare workers who received the BNT162b2 mRNA vaccine, to assess local or systemic reactogenicity and antibody levels (immunogenicity).

NH residents reported reactions at a much lower frequency and lesser severity than the community-dwelling healthcare workers. These reactions were mild and transient with all subjects experiencing more local than systemic reactions. Based on our reactogenicity and immunogenicity data, we developed a linear regression model predicting log-transformed anti-spike, anti-receptor-binding domain (RBD), and neutralizing titers, with a dichotomous variable indicating the presence or absence of reported reactions which revealed a statistically significant effect, with estimated shifts in log-transformed titers ranging from 0.32 to 0.37 (all p < 0.01) indicating greater immunogenicity in subjects with one or more reported reactions of varying severity.

With a significantly lower incidence of post-vaccination reactions among NH residents as reported in this study, the BNT162b2 mRNA vaccine appears to be well-tolerated among this vulnerable population. If validated in larger populations, absence of reactogenicity could help guide clinicians in prioritizing vaccine boosters.

However, the pathogenesis and infection mechanism mediated by T3SS1 are not entirely clarified. Similar to previous studies on other T3SS-positive bacteria, the T3SS1 needle is a major extracellular component in V. parahaemolyticus. We recently showed that the needle gene-deletion mutant (ΔvscF) exhibited markedly decreased cytotoxicity and effector translocation during interaction with HeLa cells. To further elucidate the pathogenesis of T3SS1 during host cell infection, bacterial RNA was extracted from wild-type POR-1 and ΔvscF mutants under infected condition for comparative RNA sequencing analysis in HeLa cell. The results showed that 120 differentially expressed genes (DEGs) were identified in the ΔvscF-infected group. These encoded proteins of DEGs, such as VP2088, VP2089, and VP2091, were annotated as ABC transporter system, whereas VP0757, VP1123, and VP1289 may be new transcriptional regulators. In addition, the downregulation of T3SS1 had a positive influence on the expression of T3SS2. This study aimed to investigate the expression level of lncRNA myocardial infarction associated transcript (MIAT) in serum of pregnant women with hypertensive disorders in pregnancy (HDP) and its clinical significance.

A total of 135 pregnant women with HDP were selected, including 69 pregnant women with gestational hypertension(GH) and 66 pregnant women with preeclampsia (PE). Sixty-eight normal pregnant women were selected as healthy control group (HC). The expression level of serum MIAT of all subjects was detected by quantitative real-time polymerase chain reaction (qRT-PCR), and the diagnostic value of MIAT for GH was evaluated by constructing receiver operating characteristic (ROC) curve. Pearson correlation coefficient was used to analyze the correlation between MIAT and patients' clinical indicators. Logistics regression analysis evaluated the influencing factors of GH development into PE.

The level of MIAT in GH group was significantly higher than that in HC group, while MIAT level in PE group was more significantly upregulated than that in GH group and HC group. ROC curve showed that MIAT had the ability to distinguish between GH patients and healthy controls. Pearson correlation coefficient suggested that HOTAIR expression was positively correlated with systolic blood pressure (SBP) and diastolic blood pressure (DPB). Logistic regression analysis showed that MIAT was an independent influencing factor for the development of GH to PE.
