Unisexual vertebrates typically form through hybridization events between sexual species in which reproductive mode transitions occur in the hybrid offspring. This evolutionary history is thought to have important consequences for the ecology of unisexual lineages and their interactions with congeners in natural communities. However, these consequences have proven challenging to study owing to uncertainty about patterns of population genetic diversity in unisexual lineages. Of particular interest is resolving the contribution of historical hybridization events versus post formational mutation to patterns of genetic diversity in nature. Here we use restriction site associated DNA genotyping to evaluate genetic diversity and demographic history in Aspidoscelis laredoensis, a diploid unisexual lizard species from the vicinity of the Rio Grande River in southern Texas and northern Mexico. The sexual progenitor species from which one or more lineages are derived also occur in the Rio Grande Valley region, although patterns of distribution across individual sites are quite variable. Results from population genetic and phylogenetic analyses resolved the major axes of genetic variation in this species and highlight how these match predictions based on historical patterns of hybridization. We also found discordance between results of demographic modelling using different statistical approaches with the genomic data. We discuss these insights within the context of the ecological and evolutionary mechanisms that generate and maintain lineage diversity in unisexual species. As one of the most dynamic, intriguing, and geographically well investigated groups of whiptail lizards, these species hold substantial promise for future studies on the constraints of diversification in unisexual vertebrates.Grassland restoration is gaining momentum worldwide to tackle the loss of biodiversity and associated ecosystem services. Restoration methods and their effects on ecological community reassembly have been extensively studied across various grassland types, while the importance of post-restoration management has so far received less attention. Grassland management is an important surrogate for natural disturbances, which most ancient grasslands have co-evolved with. Thus, without the reintroduction of management-related disturbance, restoration targets are unlikely to be achieved in restored grasslands. In this study, we aimed to explore how 20 years of management by mowing once a year or light cattle grazing, affects restoration success in Palearctic meadow-steppe grasslands restored by either sowing native grasses ('sown sites'), applying Medicago sativa as a nurse plant ('Medicago sites'), or allowing spontaneous succession ('spontaneous sites'). We found that following mowing, sown sites maintained long-laembly comparable to the choice of restoration method. Restoration planners may, therefore, need to put more emphasis on future management than on the initial restoration method. However, our findings also imply that if local constraints, such as potentially high invasive propagule pressure, necessitate the application of restoration methods that could also hinder the establishment of target species, the long-term recovery of the grassland can still be ensured by wisely chosen post-restoration management.We investigated whether highly available organic residues in Brazil can be used as substrates for the production of the oyster mushroom Pleurotus ostreatus, instead of the conventional cultivation using the eucalyptus sawdust substrate. We assessed the mushroom yield on 13 substrates, of which 12 were formulated with different concentrations of organic residues and one with pure eucalyptus sawdust, and verified whether the raw material used in the substrate formula and the concentration of such alternative residues influenced their biological efficiencies. The clinical community has no objective biomarker to inform the diagnosis and subtyping of ADHD. This study aimed to explore the potential diagnostic biomarkers of ADHD among surface values, volumetric metrics, and radiomic features that were extracted from structural MRI images. Public data of New York University and Peking University were downloaded from the ADHD-200 consortium. MRI T1-weighted images were pre-processed using CAT12. We calculated surface values based on the Desikan-Killiany atlas. The volumetric metrics (mean grey matter volume and mean white matter volume) and radiomic features within each AAL brain area were calculated using DPABI and IBEX, respectively. The differences among three groups of participants were tested using ANOVA or Kruskal-Wallis test depending on the normality of the data. We selected discriminative features and classified typically developing controls (TDCs) and ADHD patients as well as two ADHD subtypes using least absolute shrinkage and selection operator and support vector machine algorithms. Our results showed that the radiomics-based model outperformed the others in discriminating ADHD from TDC as well as classifying ADHD subtypes (area under curve [AUC] 0.78 and 0.94 in training test; 0.79 and 0.85 in testing set). Combining grey matter volumes, surface values, and clinical factors with radiomic features can improve the performance for classifying ADHD patients and TDCs with training and testing AUCs of 0.82 and 0.83, respectively. This study demonstrates that MRI T1-weighted features, especially radiomic features, are potential diagnostic biomarkers of ADHD.The global decline of marine foundation species (kelp forests, mangroves, salt marshes, and seagrasses) has contributed to the degradation of the coastal zone and threatens the loss of critical ecosystem services and functions. Restoration of marine foundation species has had variable success, especially for seagrasses, where a majority of restoration efforts have failed. While most seagrass restorations track structural attributes over time, rarely do restorations assess the suite of ecological functions that may be affected by restoration. Here we report on the results of two small-scale experimental seagrass restoration efforts in a central California estuary where we transplanted 117 0.25 m2 plots (2,340 shoots) of the seagrass species Zostera marina. We quantified restoration success relative to persistent reference beds, and in comparison to unrestored, unvegetated areas. Within three years, our restored plots expanded ~8500%, from a total initial area of 29 m2 to 2513 m2 . The restored beds rapidly began to resemble the reference beds in 1) seagrass structural attributes (canopy height, shoot density, biomass), 2) ecological functions (macrofaunal species richness and abundance, epifaunal species richness, nursery function), and 3) biogeochemical functions (modulation of water quality). We also developed a multifunctionality index to assess cumulative functional performance, which revealed restored plots are intermediate between reference and unvegetated habitats, illustrating how rapidly multiple functions recovered over a short time period. Our comprehensive study is one of few published studies to quantify how seagrass restoration can enhance both biological and biogeochemical functions. Our study serves as a model for quantifying ecosystem services associated with the restoration of a foundation species and demonstrates the potential for rapid functional recovery that can be achieved through targeted restoration of fast-growing foundation species under suitable conditions.Plant-pollinator interactions represent a crucial ecosystem function threatened by anthropogenic landscape alterations. Disturbances that reduce plant diversity are associated with floral resource and pollinator declines. Establishing wildflower plantings is a major conservation strategy targeting pollinators, the success of which depends on long-term persistence of seeded floral communities. However, most pollinator-oriented seeding projects are monitored for a few years, making it difficult to evaluate the longevity of such interventions. Selecting plant species to provide pollinators diverse arrays of floral resources throughout their activity season is often limited by budgetary constraints and other conservation priorities. To evaluate the long-term persistence of prairie vegetation seeded to support pollinators, we sowed wildflower seed mixes into plots on a degraded reclaimed strip-mine landscape in central Ohio, USA. We examined how pollinator habitat quality, measured as floral abundance and diversity abundance patterns of seeded and adventive species blooms resulted in a relatively constant floral abundance across the growing season. Seeded plant communities followed classic successional patterns in which annual species quickly established and flowered but were replaced by perennial species after the first few summers. Long-term data on establishment and persistence of flower species can guide species selection for future-oriented pollinator habitat restorations.
Diabetic population have a twofold to threefold increased risk of developing liver cancer, and hyperglycaemia is a prime causative factor that propends the tumour cells to undergo aggressive metabolic growth. In this study, we aimed to examine the molecular mechanism by which lutein inhibits hyperglycaemia-induced human hepatocarcinoma (HepG2) cell proliferation.

The effect of lutein on high glucose-induced proliferation was measured using the WST-1 reagent. Its effect on intracellular reactive oxygen species (ROS) levels was measured by DCF assay. The effect on the expression of antioxidant enzymes, cell cycle regulatory proteins and intracellular protein kinases was analysed by western blotting. The modulatory effect of lutein on different phases of the cell cycle was analysed by flow cytometry.

The data showed that lutein at 5µM concentration significantly blocked glucose-promoted HepG2 cell proliferation. Suppression of high glucose-induced cell proliferation by lutein was not associated with apoptosis induction, but it was linked with inhibition of hyperglycaemia-mediated elevated ROS and upregulated expression of high glucose-mediated repressed heme oxygenase 1 (HO1). Furthermore, G2/M phase cell cycle arrest and associated phosphorylation of Cdk1 and P53 were found to be linked with suppressed hyperglycaemia-mediated cell proliferation by lutein. In addition, lutein inhibited hyperglycaemia-induced activation of P38 which relates to high glucose-induced ROS-mediated growth suppression and modulated the phosphorylation of Erk, JNK and Akt in hyperglycaemic HepG2 cells.
