The change in the macrophage development after deletion of CD98hc is associated with increased apoptotic gene expression. We sought to evaluate the effect of overweight/obesity on clinical outcomes of patients with vasospastic angina (VA) at 1-year follow-up. The VA-KOREA (Vasospastic Angina in Korea) registry was a cohort of 11 centers from 2010 to 2015. Using the body mass index (BMI) cut-off for Asians, 517 patients with definite VA were divided into either an overweight/obese (BMI ≥ 23 kg/m2) group (n = 378) or a normal weight (BMI 18.5-22.9 kg/m2) group (n = 139). The overweight/obese group showed a significantly lower rate of the primary endpoint composite (2.4% vs 7.9%, p = 0.004) and ACS (0.8% vs 4.3%, p = 0.007) than the normal weight group in the crude population. Similarly, in propensity-score matched analysis, the overweight/obese group showed a significantly lower rate of the primary endpoint composite (2.3% vs 8.4%, p = 0.006) and ACS (1.1% vs 4.6%, p = 0.035) than the normal weight group. However, there were no significant differences in CD and new-onset arrhythmia between the two groups in both the crude and propensity-score matched population. Independent predictors of the primary endpoint were overweight/obesity and dyslipidemia. In patients with VA, the overweight/obese group was associated with a favorable 1-year primary endpoint and the difference was mainly driven by the lower rate of ACS compared with the normal weight group.Arbuscular mycorrhizal fungi (AMF) and phosphate solubilizing bacteria (PSB) could interact synergistically because PSB solubilize sparingly available phosphorous compounds into orthophosphate that AMF can absorb and transport to the host plant. Little is known about the interactions between these two groups in terms of promoting Jerusalem artichoke, Helianthus tuberosus L., which is widely planted by farmers because of its high inulin content. This study aimed to isolate and characterize PSB and investigate the effects of co-inoculation of AMF and PSB on plant performance and inulin accumulation. Isolate UDJA102x89-9, identified as Klebsiella variicola (KV), showed phosphate-solubilizing ability and produced high amounts of several organic acids in vitro and of indole-3-acetic acid (IAA). The experiment combined KV and two AMF species (Glomus multisubtensum (GM) and Rhizophagus intraradices (RI)). Co-inoculation of KV with RI, in combination with rock phosphate, showed the largest increases in plant growth and tuber inulin content, compared both to an unfertilized and fertilized control. The high combined effect may have the potential for use by farmers in the future as a biofertilizer for inulin production by Helianthus tuberosus L.Tumor Mutational Burden (TMB) is a measure of the abundance of somatic mutations in a tumor, which has been shown to be an emerging biomarker for both anti-PD-(L)1 treatment and prognosis; however, multiple challenges still hinder the adoption of TMB as a biomarker. The key challenges are the inconsistency of tumor mutational burden measurement among assays and the lack of a meaningful threshold for TMB classification. Here we describe a new method, ecTMB (Estimation and Classification of TMB), which uses an explicit background mutation model to predict TMB robustly and to classify samples into biologically meaningful subtypes defined by tumor mutational burden.Designing non-natural antibody formats is a practical method for developing highly functional next-generation antibody drugs, particularly for improving the therapeutic efficacy of cancer treatments. One approach is constructing bispecific antibodies (bsAbs). We previously reported a functional humanized bispecific diabody (bsDb) that targeted epidermal growth factor receptor and CD3 (hEx3-Db). We enhanced its cytotoxicity by constructing an Fc fusion protein and rearranging order of the V domain. In this study, we created an additional functional bsAb, by integrating the molecular formats of bsAb and high-affinity mutants previously isolated by phage display in the form of Fv. Introducing the high-affinity mutations into bsDbs successfully increased their affinities and enhanced their cytotoxicity in vitro and in vivo. However, there were some limitations to affinity maturation of bsDb by integrating high-affinity Fv mutants, particularly in Fc-fused bsDb with intrinsic high affinity, because of their bivalency. The tetramers fractionated from the bsDb mutant exhibited the highest in vitro growth inhibition among the small bsAbs and was comparable to the in vivo anti-tumor effects of Fc-fused bsDbs. This molecule shows cost-efficient bacterial production and high therapeutic potential.
