Fructose-driven glycolysis is proposed as an alternative pathway capable of sustaining ATP production even under anoxic conditions. Here, we tested the hypothesis that facilitating fructose-driven metabolism exerts a protective effect against anoxic stress in Drosophila. Genetically modified flies with the human fructose transporter (GluT5) and ketohexokinase (KHK) genes downstream of upstream activating sequence (UAS) were constructed. The GAL4-UAS system was confirmed to (i) increase the expression of GluT5 and KHK in a tissue-specific and a time-dependent manner (i.e., whole flies [with Act5c-gene switch GAL4 driver], neurons [with elav-gene switch GAL4 driver]) and (ii) reduce mortality of flies when placed under anoxic stress. Taken together, these data suggest that increasing fructose metabolism may be a clinically relevant approach to minimize hypoxia-induced cellular damage.
Women may receive stroke care less often than men. We examined the contribution of clinical care on sex differences and health-related quality of life (HRQoL) after stroke.

We included first-ever strokes registered in the Australian Stroke Clinical Registry (2010-2014) with HRQoL assessed between 90 and 180days after onset (EQ-5D-3L instrument) that were linked to hospital administrative data (up to 2013). Study factors included sociodemographics, comorbidities, walking ability on admission (stroke severity proxy) and clinical care (e.g. stroke unit care). Responses to the EQ-5D-3L were transformed into a total utility value (-0.516 'worse than death' to 1 'best' health). Quantile regression models, adjusted for confounding factors, were used to determine median differences (MD) in utility scores by sex.

Approximately 60% (6852/11418) of stroke survivors had an EQ-5D-3L assessment (median 139days; 44% female). Compared with men, women were older (median age 77.1 years vs. men 71.2 years) and fewer could walk on admission (37.9% vs. men 46.1%, P<0.001). Women had lower utility values than men, and the difference was explained by age and stroke severity, but not clinical care [MD
-0.039, 95% confidence interval -0.056, -0.021]. Poorer HRQoL was observed in younger men (aged<65years), particularly those with more comorbidities, and in older women (aged≥75years).

The proband, a 2-year-old boy (Patient 1) with early onset atopic dermatitis-like eczema and recurrent bacterial infections, was suspected of autosomal dominant hyper immunoglobulin E syndrome on the basis of his symptoms and family history. His mother (Patient 2) also had skin eczema and recurrent bacterial infections, and his sister (Patient 3) had skin eczema. A novel STAT3 mutation (p.S476F) was detected in all three patients, but not in the father, who had no such symptoms. A significant decrease in peripheral blood Th17 subsets and IL-17 production was found in all the patients. Curiously, all three patients carrying the p.S476F mutation in STAT3 lacked connective tissue signs such as distinctive facial features, retention of primary teeth, and joint hyperextensibility.

A survey of veterinary ophthalmology clinicians was distributed over two international list serves servicing veterinary ophthalmologists, trainees, and individuals whose practice consisted primarily of ophthalmic patients. The survey was developed following analysis of historical and currently available medical options for control of intraocular pressure and for neuroprotection.

Responses from 199 veterinary ophthalmology clinicians were evaluated. While a large variety of topical anti-hypertensive drugs and protocols were used, the most commonly used medications were aqueous humor production suppressors such as dorzolamide 2.0% ophthalmic solution, timolol 0.5% ophthalmic solution, and a combination product containing both drugs. Latanoprost 0.005% ophthalmic solution was used infrequently for prophylaxis by comparison. This study aimed to assess calvarial bone conversion 4 months after being grafted in the edentulous maxillary bone.

In 13 patients (age65.3 ± 8.7 years) the atrophic maxilla was reconstructed with autologous calvarial bone. Biopsies were taken from fresh calvarial bone grafts and from the reconstructed maxillae after 4 months of healing. Micro-CT, histomorphometric, and histological analysis were performed. From three patients biopsies were obtained after 9, 11, or 45 months.

The micro-CT analysis revealed that in the maxilla the calvarial bone was well preserved even after 45 months. Histology showed progressive incorporation of grafted bone within a maxillary bone. Osteoid and osteocytes were present in all biopsies indicating new bone formation and vital bone. Histomorphometrically, the percentage of grafted bone volume over total volume decreased from 79.8% (IQR78.7-83.3) in fresh calvarial grafts to 59.3% (IQR44.8-64.6) in healed grafts. The biopsies were taken after 9, 11, and 45 months showed similar values.

We articulate challenges and benefits of these partnerships for developmental science research, identify relevant research frameworks that may inform these partnerships, and provide an example of an ongoing research-practice partnership.Global vegetated coastal habitats (VCHs) represent a large sink for organic carbon (OC) stored within their soils. The regional patterns and causes of spatial variation, however, remain uncertain. The sparsity and regional bias of studies on soil OC stocks from Chinese VCHs have limited the reliable estimation of their capacity as regional and global OC sinks. Here, we use field and published data from 262 sampled soil cores and 181 surface soils to report estimates of soil OC stocks, burial rates and losses of VCHs in China. We find that Chinese mangrove, salt marsh and seagrass habitats have relatively low OC stocks, storing 6.3 ± 0.6, 7.5 ± 0.6, and 1.6 ± 0.6 Tg C (±95% confidence interval) in the top meter of the soil profile with burial rates of 44 ± 17, 159 ± 57, and 6 ± 45 Gg C/year, respectively. The variability in the soil OC stocks is linked to biogeographic factors but is mostly impacted by sedimentary processes and anthropic activities. All habitats have experienced significant losses, resulting in estimated emissions of 94.2-395.4 Tg CO2 e (carbon dioxide equivalent) over the past 70 years. Reversing this trend through conservation and restoration measures has, therefore, great potential in contributing to the mitigation of climate change while providing additional benefits. This assessment, on a national scale from highly sedimentary environments under intensive anthropogenic pressures, provides important insights into blue carbon sink mechanism and sequestration capacities, thus contributing to the synchronous progression of global blue carbon management.
