Although chemotherapy treatment is a standard for TNBC, it lacks a specific target. Therefore, new therapeutic strategies are required to be investigated. In this study, a combined doxorubicin (DOX) and small interfering RNA (siRNA) therapy is proposed as therapeutic strategy for targeting TGFβ1 gene. Hs578T cell line is used as in vitro model for TNBC, wherein TGFβ1siRNA therapy is employed to enhance therapeutic effects. Cell proliferation rate is measured using an MTT test, and morphological alterations are assed using microscopically approached, while gene expression is determined by qRT-PCR analysis. The combined treatment of TGFβ1siRNA and DOX reduced levels of cell proliferation and mitochondrial activity and promoted the alteration of cell morphology (dark-field microscopy). DOX treatment caused downregulation of six genes and upregulation of another six genes. The combined effects of DOX and TGFβ1siRNA resulted in upregulation of 13 genes and downregulation of four genes. Silencing of TGFβ1 resulted in activation of cell death mechanisms in Hs578T cells, to potentiate the effects of DOX, but not in an additive manner, due to the activation of genes involved in resistance to therapy (ABCB1 and IL-6).We explored the association of fecal bacterial species and somatic telomere changes in patients with chronic disease. The results showed that the length of the combined range of telomere and the methylated subtelomere was correlated with the increase of bacteria species and the numerical superiority of certain strains in feces, the increase of streptococci in men and women, and the increase of E. coli specifically in women. These results suggest that the aging status reflected by telomere length and/or demethylation of neighboring regions correlate with intestinal conditions which influences the proportion of the intestinal microbial population. Shortened telomere length and subtelomeric demethylation status are thought to represent the degree of aging and the accelerating stage of aging velocity, respectively. Hence, the observed biased microbial status is considered to be associated with advanced stage or acceleration phase of biological aging.Prevalence of obesity becomes an important health issue worldwide, but the management of obesity remains unsatisfied. This study aimed to explore the mechanism of long non-coding RNA TUG/miR-204/SIRT1 axis, which was involved in the pathogenesis of obesity. Obesity mouse model was induced by high-fat diet and treated with taurine upregulated gene1 (TUG1) virus via tail intravenous injection. Then, body weight, serum glucose, insulin tolerance, testicular fat weight were detected, as well as the expression of TUG1, microRNA-204 (miR-204), sirtuin1 (SIRT1), and inflammation and fatty accumulation associated proteins and cytokines. Regulatory relationship between TUG1/SIRT1 and miR-204 was confirmed by dual-luciferase reporter activity assay. A high-glucose-induced 3T3-L1 cell model was also constructed to explore the regulatory mechanism of TUG/miR-204/SIRT1 axis in the pathogenesis of obesity at cell level after altering the expression of TUG1, miR-204, and SIRT1. Overexpression of TUG1 could significantly attenuate the weight, serum glucose, glucose, insulin tolerance, fatty accumulation, and inflammation in obesity mice, as well as the elevation of miR-204, but increase the expression of SIRT1, phosphorylated AKT (p-AKT), glucose transporter4 (GLUT4), and peroxisome proliferator activated receptorγ (PPARγ). Both TUG1 and SIRT1 were targets of miR-204 and could be negatively regulated by miR-204. Overexpression of TUG1 could suppress the inflammation in adipocytes via downregulating miR-204 and promote GLUT4/PPARγ/AKT pathway high-glucose-induced inflammation in 3T3-L1 cells. miR-204 inhibitors could also suppress high-glucose-induced inflammation in 3T3-L1 cells via promoting SIRT1/ GLUT4/PPARγ/AKT pathway. LncRNA TUG1 could negatively regulate miR-204 to alleviate inflammation and insulin tolerance via promoting SIRT1/GLUT4/PPARγ/AKT pathway.New York City (NYC) has experienced large reductions in violent crime over the last two decades, but gun-related violence continues to pose a threat to public safety. Despite strong gun laws, high-risk individuals in NYC neighborhoods are unfortunately still able to access and misuse firearms. This research analyzes NYC's underground gun market by closely examining the flow of guns into the two boroughs where gun violence and crime gun recoveries are most prevalent the Bronx and Brooklyn. A mixed methods approach is utilized that consists of an assessment of firearms trace data and in-depth interviews with individuals considered to be at high risk for involvement in gun violence. Findings suggest that guns recovered in the Bronx and Brooklyn were significantly more likely to originate in states with less restrictive gun laws and more likely to have changed ownership in unregulated transactions relative to guns recovered elsewhere in NYC. Interviews revealed three primary avenues for illegal guns reaching Bronx and Brooklyn neighborhoods high-volume gun brokers, middlemen, and individuals who make episodic low-level acquisitions from straw purchasers in other states. We aim to fill in the research gaps with a study focusing on urban health. We develop and apply analytical approaches to address two challenges. First, domain expertise on drivers of neighborhood-level stroke outcomes is limited. Second, commonly used linear regression methods may provide incomplete and biased conclusions. Highly flexible machine learning identifies true drivers of neighborhood cardiovascular health outcomes from wide-ranging information in an agnostic and reproducible way. In this paper, we aimed to investigate the effect of Shatavarin IV (SIV), a steroidal saponin isolated from Asparagus racemosus Willd. on dietary restriction (DR) induced longevity in Caenorhabditis elegans. SIV significantly increased the lifespan to 18% which is independent of antimicrobial activity and reduced the aging by-product, lipofuscin along with increased locomotion, and chemotaxis behavior in wild type worms. The longevity effect has been dependent on eat-2, which was further validated via reduced pharyngeal pumping rate that established the effect similar to DR induced longevity. Moreover, like eat-2 mutant worms, SIV reduces the total progeny number of wild type worm along with a significant alleviation of stored fat, which reconfirms the involvement of eat-2 mediated longevity. Further, it was also observed that DR induced longevity mechanism by SIV requires mTOR which works in PHA-4/FOXA dependent manner. link3 In addition to this, the role of autophagy mechanism concerning SIV mediated DR was confirmed via bec-1, unc-51, and lgg-1. The longevity effect achieved by SIV was also dependent on SKN-1/NRF-2 and partially dependent on DAF-16/FOXO. Furthermore, the DR-induced longevity by SIV was found to be independent of hsf-1 exhibiting non-significant alteration in the mRNA expression of downstream target genes hsp-16.2 and hsp-70. Altogether, this study provides first-hand information on the pro-longevity effect of SIV in worms that have been mediated by the DR-regulating gene induced autophagy.In this study, we report for the first time a novel type of sorbent that can be used for mercury adsorption from the air-based off-gasses-vermiculite impregnated with alkali polysulfides and thiosulfates. In contrast to other sorbents, vermiculite exhibits superior thermal stability in air and low adsorption capacity for organic vapors. This allows for a more favorable design of the soil remediation unit-direct coupling of thermal desorber with catalytic oxidizer using air as a carrier gas. In the bench-scale test at 180 °C, the sulfur/vermiculite sorbent exhibited significantly higher efficiency for the adsorption of mercury vapor from the off-gasses than the commercial sulfur/activated carbon sorbent at its highest operating temperature (120 °C). The average mercury concentration in the adsorber off-gas decreased from 1.634 mg/m3 for the sulfur/activated carbon to 0.008 mg/m3 achieved with impregnated vermiculite. The total concentration of organic compounds in the soil after thermal desorption was below the detection limit of the employed analytical method.
