The potential binding sites of miR-148a-3p in the 3'-UTR of QKI were predicted by the TargetScan and verified by the dual luciferase report assay. The co-localization of miR-148a-3p and QKI through in situ hybridization was observed in the rumen tissues but not in the intestinal tracts. Moreover, the expression of miR-148a-3p in the epithelium was significantly higher than that in the other layers of the rumen, suggesting that miR-148a-3p is involved in the development of the rumen epithelial cells by targeting QKI. Subsequently, miR-148a-3p inhibitor was found to induce the proliferation of GES-1 cells. Taken together, our study identified DEMs involved in the development of the rumen and provides insights into the regulation mechanism of rumen development in goats.Carbon fiber reinforced polycarbonate (CF/PC) composites have attracted attention for their excellent performances. However, their performances are greatly affected by environmental factors. In this work, the composites were exposed to hydrothermal aging to investigate the effects of a hot and humid environment. The mechanical properties of CF/PC composites with different aging times (0, 7, 14, 21, 28, 35, and 42 days) were analyzed. It was demonstrated that the storage modulus of CF/PC composites with hot water aged for seven days has the highest value in this sampling period and frequency. Through the solid particle erosion experiment, it was found that the hydrothermal aging causes the deviation of the maximum erosion angle of composites, indicating the composites underwent ductile-brittle transformation. Furthermore, the crack and cavity resulting from the absorption of water was observed via the scanning electron microscope (SEM). This suggested that the hydrothermal aging leads to the plasticization and degradation of CF/PC composites, resulting in a reduction of corrosion resistance.Philadelphia-negative chronic myeloproliferative neoplasms (MPNs) have recently been revealed to be related to chronic inflammation, oxidative stress, and the accumulation of reactive oxygen species. It has been proposed that MPNs represent a human inflammation model for tumor advancement, in which long-lasting inflammation serves as the driving element from early tumor stage (over polycythemia vera) to the later myelofibrotic cancer stage. It has been theorized that the starting event for acquired stem cell alteration may occur after a chronic inflammation stimulus with consequent myelopoietic drive, producing a genetic stem cell insult. When this occurs, the clone itself constantly produces inflammatory components in the bone marrow; these elements further cause clonal expansion. In BCR-ABL1-negative MPNs, the driver mutations include JAK 2, MPL, and CALR. Transcriptomic studies of hematopoietic stem cells from subjects with driver mutations have demonstrated the upregulation of inflammation-related genes capable of provoking the development of an inflammatory state. The possibility of acting on the inflammatory state as a therapeutic approach in MPNs appears promising, in which an intervention operating on the pathways that control the synthesis of cytokines and oxidative stress could be effective in reducing the possibility of leukemic progression and onset of complications.Control of the crop pest African cotton leafworm, Spodoptera littoralis (Boisduval), by chemical insecticides has led to serious resistance problems. Ajuga plants contain phytoecdysteroids (arthropod steroid hormone analogs regulating metamorphosis) and clerodanes (diterpenoids exhibiting antifeedant activity). We analyzed these compounds in leaf extracts of the Israeli Ajuga iva L. by liquid chromatography time-of-flight mass spectrometry (LC-TOF-MS) and thin-layer chromatography (TLC), and their efficiency at reducing S.littoralis fitness. First and third instars of S. littoralis were fed castor bean leaves (Ricinus communis) smeared with an aqueous suspension of dried methanolic crude extract of A. iva phytoecdysteroids and clerodanes. Mortality, larval weight gain, relative growth rate and survival were compared to feeding on control leaves. We used '4',6-diamidino-2-phenylindole (DAPI, a fluorescent stain) and phalloidin staining to localize A. iva crude leaf extract activity in the insect gut. Ajuga iva crude leaf extract (50, 100 and 250 µg/µL) significantly increased mortality of first-instar S. littoralis (36%, 70%, and 87%, respectively) compared to controls (6%). HS94 Third-instar larval weight gain decreased significantly (by 52%, 44% and 30%, respectively), as did relative growth rate (-0.05 g/g per day compared to the relevant controls), ultimately resulting in few survivors. Crude leaf extract (250 µg/µL) reduced gut size, with relocation of nuclei and abnormal actin-filament organization. Ajug iva extract has potential for alternative, environmentally safe insect-pest control.This study presents the results of experimental research and numerical calculations regarding models of a typical torsion box fragment, which is a common thin-walled load-bearing structure used in aviation technology. A fragment of this structure corresponding to the spar wall was made using 3D printing. The examined system was subjected to twisting and underwent post-critical deformation. The aim of this study was to evaluate the protective effects of HT in an in vitro model of lipopolysaccharide (LPS)-induced inflammation in bovine uterine endometrial cells. Our results showed that HT had a significant protective effect in LPS-induced inflammation and oxidative stress. HT was also able to increase the capacity of endogenous antioxidant systems through the up-regulation of the NRF2 pathway. Furthermore, HT restored the tight junction protein expressions. In conclusion, our results showed the protective effects of HT in LPS-stimulated BEND cells. Therefore, the results of this study suggest an important protective role of HT in the treatment and prevention of uterine pathologies in dairy cows.This study explores the energy partition patterns of modern growing pigs at 25 kg and 65 kg raised at gradient-ambient temperatures. It also investigates the underlying changes in plasma under such conditions, based on the integrative analysis of indirect calorimetry and non-target metabolomics profiling. Thirty-six barrows with initial BW of 26.4 ± 1.9 kg and 24 barrows with initial BW of 64.2 ± 3.1 kg were successively allotted to six respiration chambers with ambient temperatures set as 18 °C, 21 °C, 23 °C, 27 °C, 30 °C, and 32 °C, and four respiration chambers with ambient temperatures set as 18 °C, 23 °C, 27 °C, and 32 °C, respectively. Each pig was kept in an individual metabolic crate and consumed feed ad libitum, then transferred into the respiration chamber after a 7-day adaptation period for 5-day indirect calorimetry assay and 1-day fasting. As the ambient temperature increased from 18 °C to 32 °C, the voluntary feed intake, metabolizable energy intake, nitrogen intake, and retention, total heat production, and energy retention as a protein of growing pigs at 25 kg and 65 kg all linearly decreased (p less then 0.05), with greater coefficients of variation for pigs at 65 kg when temperatures changed from 18 °C to 32 °C. The cortisol and thyroid hormone levels in the plasma of 25 kg pigs linearly decreased as the ambient temperature increased from 18 °C to 32 °C (p less then 0.05), and 13 compounds were identified through metabolomics analysis, including up-regulated metabolites involved in fatty acid metabolism, such as adrenic acid and down-regulated metabolites involved in amino acid metabolism, such as spermidine at 32 °C. These results suggested that modern growing pigs at heavier bodyweight were more sensitive to high temperatures on energy intake and partition. Most of the identified metabolites altered at high ambient temperatures are associated with suppressed fatty acid oxidation and elevated lipogenesis and protein degradation.A cross-sectional survey was conducted to determine the seroprevalence of Leptospira in a cohort of horses and to evaluate potential risk factors for Leptospira seropositivity in horses in New Zealand. The convenience sample included 499 Thoroughbred racing and breeding horses from 25 commercial properties in North Island, New Zealand. A questionnaire was used to collect demographic data on horses and property-level information on grazing and management practices, pest (rodent) management, access to natural waterways, other livestock on the property, and possible contact with wildlife. The microscopic agglutination test was used to test sera for serovars Ballum, Copenhageni, Hardjo (bovis), Pomona, and Tarassovi. Logistic regression was used to investigate the risk factors for Leptospira seropositivity to at least one serovar and for each serovar individually. A total of 124 (25%, 95% confidence interval (CI) 21-29%) horses had positive titres to any one of the five serovars. The seroprevalence of Ballum, Copenhageni, Hardjo (bovis), Pomona, and Tarassovi was 5% (95% CI 3-7%), 9% (95% CI 7-12%), 6% (95% CI 4-8%), 6% (95% CI 4-8%), and 6% (95% CI 4-8%), respectively. Broodmares, compared to racehorses and alternately grazing horses with sheep, increased the odds of exposure to any one serovar, whilst grazing the same time as sheep and alternately grazing horses with cattle increased the odds of exposure to Ballum and Hardjo (bovis), respectively. Historical exposure to Leptospira in racing and breeding horses was identified, and risk factors were consistent with pasture-based exposure.The World Health Organization (WHO) recommends exclusive breastfeeding for six months and continuation of breastfeeding for up to two years. link3 Formal child care has an important role in supporting breastfeeding, as many Australian infants commence care before two years of age. Currently, little is known about support or barriers in child care contexts. The present qualitative instrumental case study explores practices which protect, promote and support breastfeeding at a child care centre located in the Australian Capital Territory's outer suburbs. Extending from a previously published collective case study, a cultural-institutional focus of analysis was used to explore the roles of proximity, flexibility and communication in supporting breastfeeding within a child care centre located close to an infant's home. Interviews with centre staff and mothers, triangulated with observations of the centre environment and policy documents provide insight into the environment. Affirming the roles of flexibility in routine and staff rostering and two-way communication, findings suggest longer-term benefits may be derived from selecting a child care centre close to an infant's home, provided mothers can overcome barriers to breastmilk expression in the workplace.
