Moreover, metabolic engineering approaches are briefly highlighted to develop cell factories to make the lignocellulose biorefinery platforms appealing.The environmental and dietary pesticide exposures can cause thyroid hormones (THs) disorders, which are associated with the high incidence of thyroid diseases worldwide. The structures of diphenyl ether pesticides and their metabolites are very similar to the structure of THs. Based on this, in silico molecular simulation approaches were used to predict, screen, evaluate and identify the binding interactions of 98 diphenyl ether structure pesticides and their metabolites (DEPMs) with 10 THs related proteins in the study. The research results indicated that these DEPMs such as fluoroglycofen (FOG), rafoxanide, diclofop, ethoxyfen and difenopenten were considered to have the greater potentials to interfere with the related proteins of THs biosynthesis, blood transport, receptor binding and metabolism. And FOG can interact with thyroid hormone receptor beta (TRβ) to form non-bond interactions. Furthermore, the results of molecular dynamics simulations showed that there were strong and stable interactions between FOG and TRβ. These results suggested that the herbicide FOG was likely to disturb THs nuclear receptor. And benzene rings and hydrophobic groups might be the characteristic chemical functional groups for DEPMs to disrupt TRβ. The relevant results of this study can be used to provide references for environmental toxicology evaluation, food safety risk assessment, and formulation and revision of pesticides and their metabolites residue limits in agricultural products and food.Microplastics have become a global environmental problem due to the ubiquitous existence. check details The impacts of microplastics on heavy metals behaviors in aquatic environment are widely investigated, however, the impacts of microplastics on bioaccumulation of heavy metals in vegetables in terrestrial environment are seldom investigated. Herein, batch experiments were carried out, the microplastics (0.001%, 0.01%, 0.1%) and heavy metal (50, 100 mg/kg Cu2+ or 25, 50 mg/kg Pb2+) were single or combined spiked into soil to cultivate rapes (Brassica napus L.) in greenhouse. Copper and lead contents of rapes in MP0.1+Cu100 and MP0.1+Pb50 treatments reached 38.9 mg/kg and 9.4 mg/kg, which were significantly (p less then 0.05) higher than those of Cu100 (35.3 mg/kg) and Pb50 (8.7 mg/kg) treatments, respectively. Results showed that microplastics in soil would facilitate heavy metals entering rape plants. In addition, contents of total chlorophyll, soluble sugar, vitamin C, malondialdehyde contents, activities of superoxide dismutase and guaiacol peroxidase, as well as related gene expression were analyzed to investigate the toxic effects of these pollutants (microplastics, Cu, and Pb) to rape plants. Malondialdehyde contents of rapes in MP0.1+Cu50, MP0.1+Cu100, MP0.1+Pb25, and MP0.1+Pb50 treatments reached 0.102 mmol/mg Protein, 0.123 mmol/mg Protein, 0.101 mmol/mg Protein, and 0.119 mmol/mg Protein, which were 1.42, 1.37, 1.46, and 1.45 times of those in Cu50, Cu100, Pb25, and Pb50 treatments, respectively. The changes of malondialdehyde content, activities of superoxide dismutase and guaiacol peroxidase, as well as contents of sugar and vitamin C indicated that microplastics in soil would bring severer damage and deteriorate quality of rape plants. The data in this study indicated that microplastics would increase the bioaccumulation of heavy metals in vegetables and damage to vegetables.The demonstration in model organisms that cellular senescence drives aging and age-related disease has led to widespread efforts to identify compounds able to selectively kill senescent cells, termed senolytics. Approaches used to identify senolytics include bioinformatic analysis of senescent cell anti-apoptotic pathways (SCAPs) for drug development and screening of drugs libraries on different senescent cell types in culture. Alternatively, cytotoxic compounds can be made specific to senescent cells through a prodrug strategy such as linking the compound to a galactose moiety where toxicity is activated by lysosomal β-galactosidase. Here we tested the role of PLCγ2 in a mouse model of epidermolysis bullosa acquisita triggered by antibodies against type VII collagen (C7), a component of the dermo-epidermal junction. PLCγ2-deficient (Plcg2-/-) mice and bone marrow chimeras with a Plcg2-/- hematopoietic system were completely protected from signs of anti-C7-induced skin disease including skin erosions, dermal-epidermal separation and inflammation, despite normal circulating levels and skin deposition of anti-C7 antibodies. PLCγ2 was required for the tissue infiltration of neutrophils, eosinophils and monocytes/macrophages, as well as for the accumulation of proinflammatory mediators (including IL-1β, MIP-2 and LTB4) and reactive oxygen species. Mechanistic experiments revealed a role for PLCγ2 in the release of proinflammatory mediators and reactive oxygen species but not in the intrinsic migratory capacity of leukocytes. Precision and reproducibility of sample preparation is an essential step. Here, we present a simple and efficient protocol for saliva 1H NMR metabolic profiling. This procedure has been specifically designed and optimized for the identification and quantification of low concentration metabolites (as low as 1.1 μM) and is suitable for all the saliva subtypes.Efflux proteins are the transport proteins expressed in the plasma membrane, which are involved in the movement of unwanted toxic substances through specific efflux pumps. Several studies based on computational approaches have been proposed to predict transport proteins and thereby to understand the mechanism of the movement of ions across cell membranes. However, few methods were developed to identify efflux proteins. This paper presents an approach based on the contextualized word embeddings from Bidirectional Encoder Representations from Transformers (BERT) with the Support Vector Machine (SVM) classifier. BERT is the most effective pre-trained language model that performs exceptionally well on several Natural Language Processing (NLP) tasks. Therefore, the contextualized representations from BERT were implemented to incorporate multiple interpretations of identical amino acids in the sequence. A dataset of efflux proteins with annotations was first established. The feature vectors were extracted by transferring protein data through the hidden layers of the pre-trained model. Our proposed method was trained on complete training datasets to identify efflux proteins and achieved the accuracies of 94.15% and 87.13% in the independent tests on membrane and transport datasets, respectively. This study opens a research avenue for the implementation of contextualized word embeddings in Bioinformatics and Computational Biology.This study examined how impairments in sensorimotor abilities of individuals with Parkinson's Disease (PD) can be related to the use and understanding of co-speech hand gestures involving literal and figurative actions. We tested individuals with PD (n = 18, 12 males, Mage = 56.5, SDage = 8.16, PD duration since onset M = 5.36 years, SD = 3.51, Hoehn and Yahr ScaleMH&Y = 2.09, SDH&Y = 0.50) and age- and education-matched neurotypical controls (n = 18, 14 males, Mage = 56.61, SDage = 8.88) with two experimental tasks. In the gesture production task, participants retold the narratives presented to them in a written format. In the gesture comprehension task, participants were asked to match a gesture with a novel verb in literal and figurative sentence contexts. Results showed that patients with PD gestured significantly less than the neurotypical controls. No group differences were found for the type of gesture use. Individuals with PD performed worse than controls on matching gestures with novel verbs, particularly for figurative meanings. Individuals' severity in the disease negatively correlated with their performance for these figurative novel verb-gesture matches. The performances in the two tasks did not correlate. These findings suggest that problems in sensorimotor abilities resulting from PD can influence overall gesture production and gesture comprehension, providing further evidence on the relations between PD and the impaired use of multimodal language.Molecular data have transformed approaches to trematode taxonomy by providing objective evidence for the delineation of species. However, although the data are objective, the interpretation of these data regarding species boundaries is subjective, especially when different markers conflict. Conserved markers can lead to an underestimation of richness and those used for finer species delineation have the capacity to inflate species recognition, perhaps unrealistically. Here we examine molecular and morphological evidence for species recognition in an especially confusing system, the lepocreadiid genus Preptetos (Pritchard, 1960) in acanthuriform fishes of the tropical Indo-west Pacific. We consider species boundaries within this genus based on combined data (ITS2 and 28S rDNA; cox1 mtDNA and morphometrics) for substantial new collections. Delineation of species using only morphological data suggest fewer species than analysis of the sequence data; the latter suggests the presence of potential cryptic species arecognise, as species, populations with unvarying morphology and similar host relationships but which may have a complex population structure over their range. In our view, this paradigm has the capacity to render tractable the interpretation of the species status of the huge trematode fauna of the tropical Indo-west Pacific.
