HER2 is an orphan receptor that is activated ligand-independently by homodimerization. In addition, HER2 is able to heterodimerize with EGFR, HER3, and HER4. Heterodimerization has been proposed as a mechanism of resistance to therapy for HER2 overexpressing breast cancer. Here, a method is presented for the simultaneous detection of individual EGFR and HER2 receptors in the plasma membrane of breast cancer cells via specific labeling with quantum dot nanoparticles (QDs). Correlative fluorescence microscopy and liquid phase electron microscopy were used to analyze the plasma membrane expression levels of both receptors in individual intact cells. Fluorescent single-cell analysis of SKBR3 breast cancer cells dual-labeled for EGFR and HER2 revealed a heterogeneous expression for receptors within both the cell population as well as within individual cells. Subsequent electron microscopy of individual cells allowed the determination of individual receptors label distributions. QD-labeled EGFR was observed with a surface density of (0.5-5) × 101 QDs/µm2, whereas labeled HER2 expression was higher ranging from (2-10) × 102 QDs/µm2. Although most SKBR3 cells expressed low levels of EGFR, an enrichment was observed at large plasma membrane protrusions, and amongst a newly discovered cellular subpopulation termed EGFR-enriched cells.In this paper, we propose a fast and simple approach for the fabrication of the electrocatalytically active ruthenium-containing microstructures using a laser-induced metal deposition technique. The results of scanning electron microscopy and electrical impedance spectroscopy (EIS) demonstrate that the fabricated ruthenium-based microelectrode had a highly developed surface composed of 10 μm pores and 10 nm zigzag cracks. The fabricated material exhibited excellent electrochemical properties toward non-enzymatic dopamine sensing, including high sensitivity (858.5 and 509.1 μA mM-1 cm-2), a low detection limit (0.13 and 0.15 μM), as well as good selectivity and stability.In this study, we investigated the relationship between sociodemographic, clinical, anthropometric, and lifestyle characteristics and the type of statin prescribed for primary prevention of cardiovascular disease (CVD). We conducted an observational study in workers who began statin treatment. Statin therapy was categorized as "high-intensity" or "low-moderate-intensity". Workers were classified according to the alignment of their statin therapy with the recommended management practices. Logistic regression models were used to evaluate the association between the different variables studied and the probability of being prescribed high-intensity statins. The only variables associated with a higher probability of being treated with high-intensity statins were increased physical activity (>40 versus less then 20 METs (metabolic equivalent of task) h/wk; odds ratio (OR), 1.65; 95%CI, 1.08-2.50) and, in diabetics, higher low-density lipoprotein cholesterol (LDL-C) levels (≥155 mg/dL versus less then 155 mg/dL; OR, 4.96; 95%CI, 1.29-19.10). The model that best predicted treatment intensity included LDL-C, diabetes, hypertension, smoking, and age (area under the Receiver Operating Characteristic curve (AUC), 0.620; 95%CI, 0.574-0.666). The prescribing and type of statin used in primary CVD prevention did not correspond with the indications in current guidelines. The probability of receiving high-intensity statins was higher in diabetics with high LDL-C levels and in more physically active individuals. These findings underscore the great variability and uncertainty in the prescribing of statins.In 2018, 208,619 new cases of leprosy were reported to the World Health Organization (WHO). Of these, 30,957 occurred in the Americas region and 28,660 (92.6% of the total in the Americas) were reported in Brazil. This study aimed to show the reality of the profile of a population in an endemic leprosy area in northern Brazil in relation to age, gender, and bacillus Calmette-Guérin (BCG) vaccination status through the collection of data in the field with the evaluation of the study individuals, who were recruited by spontaneous demand. A total of 405 individuals participated in the study, with 100 multibacillary, 57 paucibacillary, and 248 healthy contacts. A relationship was observed between the occurrence of the disease, as well as the multibacillary form with the largest age group. The male gender was associated with leprosy per se, with the multibacillary form and was the largest representative of the group that was not vaccinated once. BCG vaccination was effective both in protecting against leprosy per se and in the multibacillary form. These results are limited by sample size, may not be conclusive, and will need further confirmation in a larger cohort.Early intervention of gestational diabetes mellitus (GDM) is effective in reducing pregnancy disorders. Fetal growth, measured by routine ultrasound scan a few weeks earlier before GDM diagnosis, might be useful to identify women at high risk of GDM. In the study, generalized estimating equations were applied to examine the associations between ultrasonic indicators of abnormal fetal growth at 22-24 weeks and the risk of subsequent GDM diagnosis. Of 44,179 deliveries, 8324 (18.8%) were diagnosed with GDM between 24 and 28 weeks. At 22-24 weeks, fetal head circumference (HC) less then 10th, fetal femur length (FL) less then 10th, and estimated fetal weight (EFW) less then 10th percentile were associated with 13% to 17% increased risks of maternal GDM diagnosis. Small fetal size appeared to be especially predictive of GDM among women who were parous. Fetal growth in the highest decile of abdominal circumference (AC), HC, FL and EFW was not associated with risk of subsequent GDM. The observed mean difference in fetal size across gestation by GDM was small; there was less than 1 mm difference for AC, HC, and FL, and less than 5 g for EFW before 24 weeks. Despite similar mean fetal growth among women who were and were not later diagnosed with GDM, mothers with fetuses in the lowest decile of HC, FL and EFW at 22-24 weeks tended to have higher risk of GDM.Carbofuran (CBF) is an efficient and broad-spectrum insecticide. As testing indicators for water quality and agricultural products, CBF and its metabolite 3-hydroxy-carbofuran (3-OH-CBF) are regulated by many countries. The detection of CBF and 3-OH-CBF is of great importance for the environment and human health. However, an immunosensor detection method for the simultaneous analysis of CBF and 3-OH-CBF remains unavailable. Herein, we report a waveguide-based fluorescent immunosensor for detecting CBF and 3-OH-CBF, synchronously. The immunosensor is based on a broad-spectrum monoclonal antibody with high binding affinity against CBF and 3-OH-CBF. The linear detection ranges for CBF and 3-OH-CBF are 0.29-2.69 and 0.12-4.59 μg/L, with limits of detection of 0.13 μg/L for CBF and 0.06 μg/L for 3-OH-CBF, respectively. This paper presents the application of an established reference piloting methodology and the consequent development of a coherent, robust workflow. Key challenges include ensuring methodological soundness and data validity while protecting partners' intellectual property. link3 The authors draw on their experiences in a 34-partner project aimed at assessing the impact of advanced automated driving functions, across 10 European countries. In the first step of the workflow, we captured the quantitative requirements of each RQ in terms of the relevant data needed from the tests. Most of the data come from vehicular sensors, but subjective data from questionnaires are processed as well. Next, we set up a data management process involving several partners (vehicle manufacturers, research institutions, suppliers and developers), with different perspectives and requirements. Finally, we deployed the system so that it is fully integrated within the project big data toolchain and usable by all the partners. Based on our experience, we highlight the importance of the reference methodology to theoretically inform and coherently manage all the steps of the project and the need for effective and efficient tools, in order to support the everyday work of all the involved research teams, from vehicle manufacturers to data analysts.Different fractions of hemp shives are used in the mixtures of the hemp-lime composite. The market offers shives of different granulation. It depends on the type of industrial hemp and on the capabilities of decortication machines. The aim of the research presented in the article is to check differences in the mechanical and hygro-thermal properties of composites with different shives fractions. The research part of the paper presents the preparation method and investigation on hemp-lime composites. Apparent density, total porosity, thermal conductivity, capillary uptake, vapor permeability, specific heat, mass absorptivity, flexural and compressive strength were examined. The results confirm that the shives fraction influences the individual properties of the composites. Hemp-lime composites with fine shives are characterized by higher water absorption, thermal conductivity, mechanical strength, vapor permeability as well as lower capillary-lifting capacity and specific heat than composites with thick shives.
