An in vitro synovial MSC migration model had previously been developed for screening but had problems in practical application. We herein developed a novel in vitro model, explored cytokines for synovial MSC mobilization with this model, and verified whether MSCs in synovial fluid increase following intra-articular injection of the cytokine.

Human synovial MSCs embedded in a mixture of Matrigel and type 1 collagen hydrogel were placed on a culture insert and then put in medium containing migration factor. Of the six cytokines, we identified the one that mobilizes the highest number of MSCs. PDGF-BB or PBS was injected into rat knees, and 48h later, synovial fluid was collected and cultured. The cells were examined for MSC properties.

PDGF-BB was the most effective for synovial MSC mobilization among six cytokines. The effect of PDGF-BB was inhibited by a PRGFR inhibitor. Injection of PDGF-BB into rat knees increased colony-forming cells in the synovial fluid. These cells had surface epitopes and multipotency comparable to MSCs and a higher capacity for proliferation, colony formation, and chondrogenesis.

Basing on the investigation of the groundwater-lake system in the mining area, data of hydrological, meteorological and remote sensing image, the mathematical model of groundwater level-lake area response mechanism in the mining area was constructed by using the principle of water balance. And the influence of mining area development on the lake area of Yimin basin had been predicted and analyzed. The results show that in the past 35 years of coal mining, the number of lake groups in Yimin basin has changed from 5 before mining (1982) to 2 (2018), and the total area of lakes has reduced from 6.94 km2 before mining to 1.43 km2 with an area reduction rate of 79%. The prairie lake-groundwater coupling model was constructed based on the principle of water balance, and the goodness-of-fit reached more than 0.83 after testing. Thus, the water quality in this river concerns the health of local inhabitants. The present study reveals seasonal variations of various physicochemical and heavy metals parameters and microbial load of water at 15 sites from Qena to Sohag cities, Egypt. The water is fresh with TDS ≤ 270 and 410 mg L-1 in summer and winter, respectively. Fe, Mn, Cd, Cr, Cu, Ni, and Zn concentrations were within drinking water specification in both seasons except Cr and Cd in summer. Viable numbers of total coliform, fecal coliform, and fecal streptococci were recorded in both seasons with fecal streptococci's disappearing in winter. The concentrations of salts and ions in winter were higher than summer due to decreased water quantity and flow rate in this season. Management of vertebral and discovertebral lesions depends on highly specific modalities like biopsy and histopathology. A systematic literature search was conducted in the common databases using MESH terms, searching for predictive factors of curve progression in adolescent idiopathic scoliosis("adolescent idiopathic scoliosis" OR "ais" OR "idiopathic scoliosis") AND ("predictive factors" OR "progression" OR "curve progression" OR "prediction" OR "prognosis").The identified and analysed factors of each study were rated to design a top five scale of the most relevant factors.

A cohort of 24 UC patients (14 responders and 10 refractory to therapy) and 26 control subjects was prospectively enrolled to undergo virus-specific serology (by ELISA assay) and assessment of both CD4+ and CD8+ virus-specific T-cell response (by interferon-γ enzyme-linked immunospotanalysis). In parallel, mucosal viral load was determined by quantitative real-time PCR and the values were correlated with both clinical and endoscopic indexes of activity. For statistics, the t-test, Mann-Withney test, Fisher's exact test and Spearman rank correlation test were applied; p  less then  0.05 was considered significant. EBV-specific CD4+ and CD8+ T-cell responses were significantly lower in UC patients compared to controls (p  less then  0.0001 and p = 0.0006, respectively), whereas no difference was found for HCMV-specific T-cell response. When dividing the UC group according to response to therapy, both responders and refractory UC patients showed a deficient EBV-specific CD4+ T-cell response with respect to controls (p  less then  0.04 and p = 0.0003, respectively). link3 Moreover, both EBV and HCMV mucosal loads were significantly higher in refractory UC than in responders and controls (p = 0.007 and 0.003; and p = 0.02 and 0.001, respectively), and correlated with activity indexes. Steroid therapy seemed the main risk factor for triggering EBV colitis. Finally, no cases of IgM positivity were found in the study population. An impaired EBV-specific immunity was clearly evident in UC patients, mostly in those refractory to therapy. The ELISPOT assay may serve as new tool for quantifying and monitoring virus-specific T-cell immunity in UC.
