This study defined the pH threshold for dark cutting beef of Asian consumers using a web-based survey and examined their preferences for beef appearance. Ninety individual images of beef steaks with different pH values were randomly selected to be scored by 4322 respondents. Gender, age and beef eating habits of respondents were found to impact on color preferences. With respondent's visual score of 4 set as the acceptance threshold, linear regression analysis between visual scores and beef instrumental colorimetrics, showed that the acceptance thresholds of instrumental color values were L* ≥ 31.4, a* ≥ 16.4, b* ≥ 6.5, chroma ≥17.4 and hue ≥22.5. These corresponded to pH threshold values of 6.11, 6.20, 6.28, 6.25 and 6.29, respectively. This pH benchmark (6.11 to 6.29) for dark cutters is higher than pH thresholds applied in most countries, indicating that Asian consumers prefer darker meat than expected, however, the likely tenderness of beef with pH's at this level merits consideration.Within the insensitization by electronarcosis and the bleeding processes performed at the pig's slaughterhouses, there are some factors that hinder the achievement of an adequate slaughter of these animals, being this a critical phase in which animal welfare must be guaranteed; the objective of this study was to evaluate and compare the efficiency of insensitization by electronarcosis and two types of bleeding direction (horizontal and vertical). Dependent variables were measured as indicators of animal welfare (absence of the corneal reflex, absence of reflex of sensitivity to painful stimuli, attempts to reinstatement or posture recovery and vocalization), after the stunning and bleeding process, in four slaughterhouses of national category in "Eje Cafetero", Colombia. The methodological approach included the binomial distribution, descriptive statistics, hypothesis testing and statistical significance. The results show that the efficiency of the insensitization procedures and type of bleeding direction depends on multiple aspects, including the tranquility of the animals during their handling, the correct position of the insensitization clamps, the amperage used and the time between insensitization and bleeding. Oxidative stress and redox homeostasis have been shown to be involved at each stage of a malignant melanocyte transformation, called melanomagenesis, as well as during drug resistance. Although challenging, targeting redox homeostasis in melanoma progression remains to be a promising therapeutic approach, especially valid during melanoma drug resistance.Tissue engineering technology requires porous scaffolds, based on biomaterials, which have to mimic as closely as possible the morphological and anisotropic mechanical properties of the native tissue to substitute. Anisotropic fibrous scaffolds fabricated by template-assisted electrospinning are investigated in this study. Fibers of electrospun Polycaprolactone (PCL) were successfully arranged spatially into honeycomb structures by using well-shaped 3D micro-architected metal collectors. Fibrous scaffolds present 2 × 4 mm2 wide elementary patterns with low and high fiber density areas. Distinct regions of the honeycomb patterns were analyzed through SEM images revealing different fiber diameters with specific fiber orientation depending on the regions of interest. Tensile test experiments were carried out with an optical observation of the local deformation at the pattern scale, allowing the determination and analysis, at small and large deformation, of the axial and transverse local strains. The honeycomb patterned mats showed significantly different mechanical properties along the two orthogonal directions probing an anisotropic ratio of 4.2. Stress relaxation test was performed on scaffolds at 15% of strain. This measurement pointed out the low contribution of the viscosity of about 20% in the mechanical response of the scaffold. An orthotropic linear elastic model was consequently proposed to characterize the anisotropic behavior of the produced patterned membranes. This new versatile method to produce architected porous materials, adjustable to several polymers and structures, will provide appealing benefits for soft regenerative medicine application and the development of custom-made scaffolds.In computational neuroscience, as well as in machine learning, neuromorphic devices promise an accelerated and scalable alternative to neural network simulations. Their neural connectivity and synaptic capacity depend on their specific design choices, but is always intrinsically limited. Here, we present a strategy to achieve structural plasticity that optimizes resource allocation under these constraints by constantly rewiring the pre- and postsynaptic partners while keeping the neuronal fan-in constant and the connectome sparse. Patients with cancer are at increased risk of complicated severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) infection, but it is still unclear if the risk of mortality is influenced by cancer type or ongoing anti-cancer treatments. An interesting debate concerning the potential relationship between androgen deprivation therapy (ADT) and SARS-CoV-2 infection has recently been opened in the case of prostate cancer (PC), and the aim of this multi-centre cohort study was to investigate the incidence and outcomes of SARS-CoV-2 infection in patients with metastatic castration-resistant prostrate cancer (mCRPC).

We retrospectively reviewed the clinical records of patients with mCRPC who developed SARS-CoV-2 infection, and recorded their baseline clinical characteristics, their history of PC and SARS-CoV-2 infection, and their oncological status and treatment at the time of infection. The primary study end point was the death rate and the possible impact of the patients' PC-related history and treatments on mortality.

Thirty-four of the 1433 patients with mCRPC attending the participating centres (2.3%) developed SARS-CoV-2 infection, 22 (64.7%) of whom were hospitalised. Most of the patients were symptomatic, the most frequent symptoms being fever (70.6%), dyspnoea (61.8%), cough (52.9%)and fatigue (38.2%). After a median follow-up of 21 days (interquartile range 13-41), 13 patients had died (38.2%), 17 recovered (50.0%)and four (11.7%) were still infected. The number of treatments previously administered for mCRPC had a significant impact on mortality (p=0.004).

The protease inhibitor ritonavir (RTV) is a clinical-stage inhibitor of the human immunodeficiency virus. In a drug repositioning approach, we here exhibit the additional potential of RTV to augment current treatment of glioblastoma, the most aggressive primary brain tumour of adulthood.

We explored the antitumour activity of RTV and mechanisms of action in a broad spectrum of short-term expanded clinical cell samples from primary and recurrent glioblastoma and in a cohort of conventional cell lines and non-tumour human neural controls invitro. To validate RTV efficacy in monotherapeutic and in combinatorial settings, we used patient-derived xenograft models in a series of invivo studies.

RTV monotherapy induced a selective antineoplastic response and demonstrated cytostatic and anti-migratory activity at clinical plasma peak levels. Additional exposure to temozolomide or irradiation further enhanced the effects synergistically, fostered by mechanisms of autophagy and increased endoplasmic reticulum stress. In xenograft models, we consequently observed increasing overall survival under the combinatorial effect of RTV and temozolomide.

Our data establish RTV as a valuable repositioning candidate for further exploration as an adjunct therapeutic in the clinical care of glioblastoma.

This study took a person-centered approach to identify profiles of lifetime substance use and discern the psychosocial differences among them. PF-06700841 concentration To survey a spectrum of drug use severity, 1106 adults (43.4% women) were recruited from forensic and community samples. Participants reported on the frequency of lifetime substance use across multiple drug categories (sedatives, stimulants, marijuana, heroin, hallucinogens, misuse of prescription drugs) and alcohol use. Latent profile analysis was used to identify distinct profiles of substance use that were then compared on potential risk and maintenance factors for substance use. Four profiles of lifetime substance use emerged that diverged on severity of use and degree of mono vs. polysubstance use (Recreational Marijuana Use, Heavy Multidrug Intoxication, Heavy Marijuana Use, and Heavy Opioid and Polysubstance Use). The profiles differed on affective motivations for substance use (e.g., using to cope vs. using to seek a thrill), age of use onset, drug-related functional impairment, and experiences of childhood maltreatment. Cognitive functioning did not differentiate the heavy substance use profiles. Results provide compelling initial evidence that lifetime patterns of use can be used to identify groups of substance users with distinct risk and maintenance factors. Results highlight affective motivations for substance use and maltreatment history as potential treatment targets and underscore the importance of studying polysubstance use in the context of the opioid epidemic.
