Idiopathic inflammatory myopathies (IIM) is a group of heterogeneous autoimmune systemic diseases, which not only involve skeletal muscle but also myocardium. Cardiac involvement in IIM, which eventually develops into heart failure, is difficult to identify by conventional examinations at early stage. The aim of this study was to investigate if multi-parametric cardiac magnetic resonance (CMR) imaging can screen for early cardiac involvement in IIM, compared with clinical score (Myositis Disease Activity Assessment Tool, MDAAT). Forty-nine patients of IIM, and 25 healthy control subjects with comparable age-range and sex-ratio were enrolled in this study. All subjects underwent CMR examination, and multi-slice short-axis and 4-chamber cine MRI were acquired to evaluate biventricular global circumferential strain (GCS) and global longitudinal strain (GLS). Native T1 and T2 mapping were performed, and post-contrast T1 mapping and LGE were acquired after administration of contrast. A CMR score was developed from native T1 mean and T2 mean for the identification of cardiac involvement in the IIM cohort. Using contingency tables MDAAT and CMR were compared and statistically analyzed using McNemar test. McNemar's test revealed no significant difference between CMR score and MDAAT (p = 0.454). CMR score had potential to screen for early cardiac involvement in IIM patients, compared to MDAAT.Assessment of enamel subsurface lesion remineralisation is essential for the evaluation of novel remineralisation technologies. The gold standard to assess subsurface mineral gain of enamel lesions is transverse microradiography (TMR). However, some studies have utilised surface microhardness (SMH) to evaluate efficacy of remineralisation agents. The aim of this study was to assess remineralisation of enamel subsurface lesions using TMR and SMH after in vitro treatment with calcium-containing technologies, and to test correlation between the TMR and SMH measurements. The parameters obtained from the TMR and SMH analyses of enamel subsurface remineralisation were not significantly correlated. Furthermore, the enamel subsurface remineralisation as measured by TMR was significantly correlated with the water-soluble calcium concentration of the remineralisation products. Scanning electron microscopy revealed surface precipitates formed by specific remineralisation treatments obfuscated accurate assessment of remineralisation by SMH. It was concluded that TMR is a more appropriate method for analysis of enamel subsurface remineralisation, and that SMH values of remineralised enamel should be interpreted with caution. Using TMR the level of remineralisation (%R) by the different technologies was CPP-ACP/F (31.3 ± 1.4%); CPP-ACP (24.2 ± 1.4%); CaSO4/K2HPO4/F (21.3 ± 1.4%); f-TCP/F (20.9 ± 1.0%); Nano-HA/F (16.3 ± 0.3%); Nano-HA (15.3 ± 0.6%) and F alone control (15.4 ± 1.3%).Despite efforts towards bridging the education gap between tribal (Scheduled Tribe) and non-tribal (Non-Scheduled Tribe) children, contrasting poor-quality education questioned the tribal children's educational wellbeing in India. Early childhood circumstances render a remarkable impact on the educational wellbeing of children in later years. This study examined the influence of early childhood circumstances (child, household and community characteristics) during 2005 on the educational wellbeing inequality (among India's tribal and non-tribal children) during 2012 using the India Human Development Survey panel dataset of 8611 children. selleck compound The Educational wellbeing score was obtained from reading, mathematical and writing test scores using Principal Component Factor Analysis. We performed the Blinder-Oaxaca decomposition of the educational wellbeing inequality among India's tribal and non-tribal children. The ST children's average educational wellbeing score (-0.41) was much lower than the Non-ST children (0.04). Findings from the Blinder-Oaxaca decomposition show that the household economic condition in children's early ages contributed to 24% of educational wellbeing inequality among tribal and non-tribal children. Further, the education status of males and female adults and the sanitation condition of families considerably impacted educational wellbeing. The present study concludes that caste antagonism has not reduced with time. The missing focus on the minority groups resulted in a deteriorated educational wellbeing. One of the main problems that a brain-computer interface (BCI) face is that a training stage is required for acquiring training data to calibrate its classification model just before every use. Transfer learning is a promising method for addressing the problem. In this paper, we propose a Riemannian geometry-based transfer learning algorithm for code modulated visual evoked potential (c-VEP)-based BCIs, which can effectively reduce the calibration time without sacrificing the classification accuracy. The algorithm includes the main procedures of log-Euclidean data alignment (LEDA), super-trial construction, covariance matrix estimation, training accuracy-based subject selection (TSS) and minimum distance to mean classification. Among them, the LEDA reduces the difference in data distribution between subjects, whereas the TSS promotes the similarity between a target subject and the source subjects. The resulting performance of transfer learning is improved significantly. Sixteen subjects participated in a c-VEP BCI experiment and the recorded data were used in offline analysis. Leave-one subject-out (LOSO) cross-validation was used to evaluate the proposed algorithm on the data set. The results showed that the algorithm achieved much higher classification accuracy than the subject-specific (baseline) algorithm with the same number of training trials. Equivalently, the algorithm reduces the training time of the BCI at the same performance level and thus facilitates its application in real world.Finite element analysis is a powerful computational technique for augmenting biomedical research, prosthetics design, and preoperative surgical assessment. However, the validity of biomechanical data obtained from finite element analysis is dependent on the quality of the preceding data processing. Until now, little information was available about the effect of the segmentation process on finite element models and biomechanical data. The current investigation applied 4 segmentation approaches to 129 femur specimens, yielding a total of 516 finite element models. Biomechanical data including average displacement, pressure, stress, and strain were collected from experimental groups based on the different segmentation approaches. The results indicate that only a 5.0% variation in the segmentation process leads to statistically significant differences in all 4 biomechanical measurements. These results suggest that it is crucial for consistent segmentation procedures to be applied to all specimens within a study. This methodological advancement will help to ensure that finite element data will be more accurate and that research conclusions will have greater validity.Understanding how different online communities engage with COVID-19 misinformation is critical for public health response. For example, misinformation confined to a small, isolated community of users poses a different public health risk than misinformation being consumed by a large population spanning many diverse communities. Here we take a longitudinal approach that leverages tools from network science to study COVID-19 misinformation on Twitter. Our approach provides a means to examine the breadth of misinformation engagement using modest data needs and computational resources. We identify a subset of accounts from different Twitter communities discussing COVID-19, and follow these 'sentinel nodes' longitudinally from July 2020 to January 2021. We characterize sentinel nodes in terms of a linked domain preference score, and use a standardized similarity score to examine alignment of tweets within and between communities. We find that media preference is strongly correlated with the amount of misinformation propagated by sentinel nodes. Engagement with sensationalist misinformation topics is largely confined to a cluster of sentinel nodes that includes influential conspiracy theorist accounts. By contrast, misinformation relating to COVID-19 severity generated widespread engagement across multiple communities. Our findings indicate that misinformation downplaying COVID-19 severity is of particular concern for public health response. We conclude that the sentinel node approach can be an effective way to assess breadth and depth of online misinformation penetration.Previous research has shown that both the daily experiences and personal traits of adolescents are linked to aggression. Our aim was to further investigate the relationship between leisure experience, self-esteem, and aggression according to the general aggression model. In addition, within frustration-aggression theory, we proposed that leisure experience and aggression have a negative correlation. Furthermore, based on broaden-and-build theory, we explored the mediating role of self-esteem between leisure experience and aggression. The participants included 660 Chinese teenagers with an average age of 14.3. Among them, male students accounted for 310 (49.4%) and female students accounted for 318 (50.6%). The results showed that leisure experience was positively correlated with self-esteem and negatively correlated with aggression, while self-esteem was also negatively correlated with aggression. Additionally, self-esteem fully mediated the relationship between leisure experience and aggression. Our study could enrich research on leisure and provide a basis for protective factors of aggression in adolescents.Stem cell therapy has great potential for replacing beta-cell loss in diabetic patients. However, a key obstacle to cell therapy's success is to preserve viability and function of the engrafted cells. While several strategies have been developed to improve engrafted beta-cell survival, tools to evaluate the efficacy within the body by imaging are limited. Traditional labeling tools, such as GFP-like fluorescent proteins, have limited penetration depths in vivo due to tissue scattering and absorption. To circumvent this limitation, a near-infrared fluorescent mutant version of the DrBphP bacteriophytochrome, iRFP720, has been developed for in vivo imaging and stem/progenitor cell tracking. Here, we present the generation and characterization of an iRFP720 expressing human induced pluripotent stem cell (iPSC) line, which can be used for real-time imaging in various biological applications. To generate the transgenic cells, the CRISPR/Cas9 technology was applied. A puromycin resistance gene was inserted into the AAVS1 locus, driven by the endogenous PPP1R12C promoter, along with the CAG-iRFP720 reporter cassette, which was flanked by insulator elements. Proper integration of the transgene into the targeted genomic region was assessed by comprehensive genetic analysis, verifying precise genome editing. Stable expression of iRFP720 in the cells was confirmed and imaged by their near-infrared fluorescence. We demonstrated that the reporter iPSCs exhibit normal stem cell characteristics and can be efficiently differentiated towards the pancreatic lineage. As the genetically modified reporter cells show retained pluripotency and multilineage differentiation potential, they hold great potential as a cellular model in a variety of biological and pharmacological applications.
