Some Natural Resistance-Associated Macrophage Proteins (NRAMPs) have been reported to play critical roles in Mn uptake and reutilization in low Mn conditions. NRAMP6 was demonstrated to regulate cadmium tolerance and iron utilization in Arabidopsis. Nevertheless, it is unclear whether NRAMP6 plays a role in Mn nutrition. Here, we report that NRAMP6 cooperates with NRAMP1 in Mn utilization. Mutation of NRAMP6 in nramp1 but not in a wild-type background reduces root growth and Mn translocation from the roots to shoots under Mn deficient conditions. Grafting experiments revealed that NRAMP6 expression in both the roots and shoots is required for root growth and Mn translocation under Mn deficiency. We also showed that NRAMP1 could replace NRAMP6 to sustain root growth under Mn deficiency, but not vice versa. Mn deficiency does not affect the transcript level of NRAMP6, but is able to increase and decrease the protein accumulation of NRAMP6 in roots and shoots, respectively. Furthermore, NRAMP6 can be localized to both the plasma membrane and endomembranes including the endoplasmic reticulum, and Mn deficiency enhances the localization of NRAMP6 to the plasma membrane in Arabidopsis plants. NRAMP6 could rescue the defective growth of the yeast mutant Δsmf2, which is deficient in endomembrane Mn transport. Our results reveal the important role of NRAMP6 in Mn nutrition and in the long-distance signaling between the roots and shoots under Mn deficient conditions.Pathological cardiac hypertrophy is the most important risk factor for developing chronic heart failure. Therefore, the discovery of novel agents for treating pathological cardiac hypertrophy remains urgent. In the present study, we examined the therapeutic effect and mechanism of periplocymarin (PM)-mediated protection against pathological cardiac hypertrophy using angiotensinII (AngII)-stimulated cardiac hypertrophy in H9c2 cells and transverse aortic constriction (TAC)-induced cardiac hypertrophy in mice. In vitro, PM treatment significantly reduced the surface area of H9c2 cells and expressions of hypertrophy-related proteins. Meanwhile, PM markedly down-regulated AngII-induced translocation of p-STAT3 into the nuclei and enhanced the phosphorylation levels of JAK2 and STAT3 proteins. The STAT3 specific inhibitor S3I-201 or siRNA-mediated depleted expression could alleviate AngII-induced cardiac hypertrophy in H9c2 cells following PM treatment; however, PM failed to reduce the expressions of hypertrophy-related proteins and phosphorylated STAT3 in STAT3-overexpressing cells, indicating that PM protected against AngII-induced cardiac hypertrophy by modulating STAT3 signalling. In vivo, PM reversed TAC-induced cardiac hypertrophy, as determined by down-regulating ratios of heart weight to body weight (HW/BW), heart weight to tibial length (HW/TL) and expressions of hypertrophy-related proteins accompanied by the inhibition of the JAK2/STAT3 pathway. These results revealed that PM could effectively protect the cardiac structure and function in experimental models of pathological cardiac hypertrophy by inhibiting the JAK2/STAT3 signalling pathway. PM is expected to be a potential lead compound of the novel agents for treating pathological cardiac hypertrophy.
Limited understanding of the fate of pesticides in apple orchards may lead to recurring pests or pose risks to food safety. In this study, through a field experiment conducted in an apple orchard, a dynamic plant uptake model, coupled with a soil water model, was developed to simulate measured pesticide concentrations in soil and different plant compartments.

Results showed that the overall model could adequately describe the data set of four pesticides in the apple orchard. An estimated 15%-24.7% of applied pesticides were deposited on leaves and 0.37%-0.58% on fruits. Decreasing pesticide concentrations in fruits were observed after pesticide application, with 9.6%-64.8% of this decrease explained by biodegradation, 29.8%-75.8% by fruit growth dilution and 11.3%-47.6% by wash-off. Furthermore, a first estimation of dietary risks indicated that ingestion of the apples may not represent an acute or chronic risk to human health.

Two imperatives in these settings are to minimize restrictive practices such as seclusion and restraint and to provide recovery-oriented care. Safewards is a model and a set of ten interventions aiming to improve safety by understanding the relationship between conflict and containment as a means of reducing restrictive practices. To date, the research into Safewards has largely focused on its impact on measures of restrictive practices with limited exploration of consumer perspectives. There is a need to review the current knowledge and understanding around Safewards and its impact on consumer safety. This paper describes a mixed-methods integrative literature review of Safewards within inpatient and forensic mental health units. The aim of this review was to synthesize the current knowledge and understanding about Safewards in terms of its implementation, acceptability, effectiveness and how it meets the needs of rature. This review also revealed the limitations of fidelity measures and the importance of involving staff in the implementation. A major gap in the literature to date is the lack of consumer perspectives on the Safewards model, with only two papers to date focusing on the consumers point of view. This is an important area that requires more research to align the Safewards model with the consumer experience and improved recovery orientation.Adzuki bean (Vigna angularis), also called red bean, is a legume of Fabaceae (Leguminosae) family. Suggestions on how to better utilize the adzuki bean are also provided to facilitate its development as a functional grain. Adzuki bean and its components can be further developed into value-added and nutritionally enhanced products.Methylglyoxal (MG) is a toxic by-product of the glycolysis pathway in most living organisms and was previously shown to inhibit seed germination. MG is detoxified by glyoxalase I and II family proteins in plants. MG is abundantly produced during early embryogenesis in Arabidopsis seeds. However, the mechanism that alleviates the toxic effect of MG in maturing seeds is poorly understood. In this study, by T-DNA mutant population screening, we found that mutations in a glyoxalase I gene (named GERMINATION-IMPAIRED GLYOXALASE 1, GIG1) led to significantly impaired germination compared with wild-type seeds. Transformation of full-length GIG1 cDNA under the constitutively active cauliflower mosaic virus 35S promoter in the gig1 background completely recovered the seed germination phenotype. Quantitative reverse transcription polymerase chain reaction (qRT-PCR) analyses revealed that GIG1 is uniquely expressed in seeds and is upregulated by abscisic acid (ABA) and downregulated by gibberellic acid (GA) during seed germination. An ABA signaling component, ABI3, directly activated GIG1 in maturing seeds. In addition, PHYTOCHROME INTERACTING FACTOR 1 (PIF1) also plays cooperatively with ABI3 in the regulation of GIG1 expression in the early stage of imbibed seeds. While synthetic bioinspired fibers have shown impressive tensile properties, the fiber formation process remains poorly understood. Moreover, these systems are highly complex and their formation is environmentally and economically costly. Controlled fiber formation under ambient conditions from polyacrylamide solutions with properties comparable to natural fibers such as wool and coir is demonstrated. Photopolymerization and subsequent microscale fibrillation of different acrylamides in water/ethanol mixtures yield a simple and energy-efficient route to fiber formation. This strategy reduces required processing energy by two-to-three orders of magnitude. Through extensive experimental elucidation, insight into precise fiber forming conditions of polymeric solutions is achieved. Ethanol is utilized as a chain transfer agent to control the molecular weight of the polyacrylamides, and the entanglement regimes of the solutions are determined through rheological characterization showing fiber formation above the entanglement concentration. Unique from previously reported hydrogel microfibers, it is shown that fibers with good mechanical properties can be obtained without the need for composites or crosslinkers. The reported approach offers a platform for fiber formation under ambient conditions with molecular-level understanding of their assembly.Insect taxa vary in their phenology across space creating dissimilar patterns of species abundance over time. The role of human disturbances and invasive species in these patterns of temporal dissimilarity (phenological differences) across space, however, remain largely unexplored. To dissect these patterns, we evaluated four common pests and one newly invasive species (Contarinia nasturtii; Swede midge) at 220 time points across 2 years on organic and conventional farms. We first summarized across time and evaluated differences in pest abundance between farm management (organic and conventional). We then used generalized additive models to describe temporal patterns of abundance, disentangling phenological differences across management systems. Last, we conducted a temporal beta diversity analysis to identify which species and management practices contribute most to dissimilarity. We found that aggregating pest abundance across time and species masked differences in pest phenology across management systems and concealed variation in pest abundance that was strongly driven by an invasive species, respectively.
