To develop an "off-the-shelf" allogeneic γδ T cell product, the cell manufacturing process must be optimized to include cryopreservation. It is known that cryopreservation can dramatically reduce viability of primary cells and other cell types after thawing, although the exact effects of cryopreservation on γδ T cell health and functionality have not yet been characterized. Our aim was to characterize the effects of a freeze/thaw cycle on γδ T cells and to develop an optimized protocol for cryopreservation. γδ T cells were expanded under serum-free conditions, using a good manufacturing practice (GMP) compliant protocol developed by our lab. We observed that cryopreservation reduced cell survival and increased the percentage of apoptotic cells, two measures that could not be improved through the use of 5 GMP compliant freezing media. The choice of thawing medium, specifically human albumin (HSA), improved γδ T cell viability and in addition, chromatin condensation prior to freezing increased cell viability after thawing, which could not be further improved with the use of a general caspase inhibitor. Finally, we found that cryopreserved cells had depolarized mitochondrial membranes and reduced cytotoxicity when tested against a range of leukemia cell lines. One of the ways to study the relationship between lipid content and cryotolerance of cells is to study the effect of lipidome modification on laboratory mice. The objective of this research was to study how in vitro culture of mouse embryos with linoleic acid (LA) will affect lipid phase transition (LPT) during cooling and subsequent embryo development after cryopreservation. Embryos obtained in vivo at the 2-cell stage were cultured with 200 μM LA for 46 h up to the morula/blastocyst stage. Thereafter, one portion of embryos was slowly frozen to reveal the effect of LA on survival after cryopreservation, another portion was used to characterize the lipid composition and to determine the temperature of the LPT onset. Confocal fluorescence microscopy of Nile Red stained embryos showed a significant increase in lipid content of the LA treated group compared to the controls. Raman measurements showed that the onset of LPT in LA treated embryos is lower than in untreated ones -5 °C vs +2 °C. However, these changes in the LPT onset did not affect the survival rates of embryos after cryopreservation. Although the sperm proteome seems to affect cell cryoresistance, studies aiming at identifying proteins involved in sperm freezing-tolerance are scarce. The aims of this study were to investigate differences of sperm freezability and proteome between epididymal and ejaculated sperm in three mountain ungulates Iberian ibex, Mouflon and Chamois. Sperm samples were cryopreserved in straws by slow freezing. Tandem mass tag-labeled peptides from sperm samples were analyzed by high performance liquid chromatography coupled to a mass spectrometer in three technical replicates. The statistical analysis was done using the moderated t-test of the R package limma. Differences of freezability between both types of sperm were associated with differences of the proteome. Overall, epididymal sperm showed higher freezability than ejaculated sperm. Between 1490 and 1883 proteins were quantified in each species and type of sperm sample. Cross species comparisons revealed a total of 76 proteins that were more abundant in epididymal than in ejaculated sperm in the three species of study whereas 3 proteins were more abundant in ejaculated than epididymal sperm in the three species of study (adjusted P 0.5). Many of the proteins that were associated with higher cryoresistance are involved in stress response and redox homeostasis. In conclusion, marked changes of sperm proteome were detected between epididymal and ejaculated sperm. The subset analysis of this multicenter, randomized, single-blind, phase IIb trial identified significant improvement in 36-month disease-free survival (DFS) between NPS (n = 55) and placebo (n = 44) in TNBC (HR 0.25, p = 0.01) and those who express HLA-A24 (HR 0.41, p = 0.05). The TNBC cohort demonstrated improved 36-month DFS in those with HER2 1+ expression (HR 0.17, p = 0.01), HLA-A24 positivity (HR 0.08, p less then 0.01), or in those who received neoadjuvant chemotherapy (HR 0.21, p less then 0.01). NPS vaccination with trastuzumab was associated with improved 36-month DFS among patients with TNBC. The present study reports first results of the BfR MEAL Study regarding methylmercury in fish, seafood and mushrooms. In total, 34 MEAL foods were purchased nationally or regionally according to a defined sampling plan, prepared in a representative way for German households, pooled into 49 samples, homogenized and subjected to ICP-MS analysis. For women with breast cancer receiving photon 3-dimensional conformal radiation therapy, bolus was used routinely for chest walls but was omitted for breast reconstructions by about half of radiation oncologists from 2007 to 2011. Eligible patients had newly diagnosed invasive breast cancers without skin involvement (pT1-4a, any-N, M0) treated with adjuvant or neoadjuvant radiation therapy. For the bolus and no-bolus groups, we compared the cumulative incidence of local recurrence (LR) and locoregional recurrence (LRR) with distant recurrence and death as competing risks and breast cancer mortality (BCM). Multivariable analysis of LR and BCM included stage, subtype, lymphovascular invasion, grade, margin status, beam energy, bolus use, hormone therapy, chemotherapy, and reconsy, radiation therapy, and modern systemic therapy, the cumulative incidence of LR was low, with or without bolus. Because bolus use increases toxicity and does not reduce LR or BCM, it should no longer be used routinely for patients without skin involvement who receive systemic therapy.In recent decades, environmental concerns and increasing consumer demand for healthy and nutritious food products with prolonged shelf life have made the food packaging industry pay more attention to the preparation of multifunctional biodegradable packaging films based on biopolymers containing active components such as antioxidant and antimicrobial agents. In this study, bio-nanocomposite films were fabricated from gelatin (G) and cellulose nanofibers (CNFs), and different concentrations of zinc oxide (ZnO) and/or Selenium (Se) nanoparticles (NPs) by the casting method. The mechanical, barrier, optical, and structural (FTIR, XRD, and SEM) properties of the films were investigated along with their antibacterial and antioxidant features. The incorporation of ZnO and Se NPs improved the physicomechanical and water resistance of G/CNF films. In this regard, the maximum tensile strength value was obtained for the G/CNF containing 5% w/w ZnO NPs (G/CNF/ZnO3) and G/CNF containing 0.1% w/w Se NPs (G/CNF/Se2) films (~2.20-fold and ~2.13-fold higher than the G/CNF film, respectively). Also, G/CNF with 3% w/w ZnO NPs (G/CNF/ZnO2) film had the lowest water vapor permeability and water solubility among all films. Results of the disc diffusion assay showed a stronger antibacterial effect of ZnO NPs compared with Se NPs. The bacterial susceptibility to the antibacterial films was as follows Listeria monocytogenes > Escherichia coli > Staphylococcus aureus > Pseudomonas fluorescens. The G/CNF films incorporated with Se nanoparticles possessed the higher property of scavenging free radicals in comparison films containing ZnO nanoparticles. Also, the combination of Se NPs and ZnO NPs enhanced the antioxidant effect of the films. In conclusion, gelatin-based edible films containing CNFs, ZnO NPs, and Se NPs can be used in the development of active food packaging products.Compression-decompression surface pressure (π)-specific molecular area (A) isotherm cycle of human serum albumin (HSA) monolayer is performed on water surface at four different subphase pH conditions, i.e., below and above the isoelectric point (pI ≈ 4.7) of HSA molecule. For all pH conditions, the decompression curve nearly follows the compression curve, however, at pH ≈ 5.0, hysteresis is observed at higher surface pressure. Out-of-plane structures and in-plane morphologies obtained from the X-ray reflectivity and AFM studies show that only the film thickness variation takes place with the change in surface pressure, which is also evidenced from the BAM images. With increase in surface pressure, the oblate-shaped HSA molecules start tilting making an angle with the water surface and as the monolayer is decompressed the molecules regain their initial untilted monomolecular configuration. Depending upon the subphase pH and local surface charge of the specific protein molecule, electrostatic repulsive interaction dominates over the van der Waals attraction and as a result decompression curve follows the compression curve as the molecules repel each other, however, closer to the isoelectric point as strength of the interactions reverses, a hysteresis is obtained at higher surface pressure and accordingly monolayer behaviour modifies on the water surface.
