Neurodegenerative diseases (NDDs) such as Alzheimer's disease (AD), Parkinson's disease (PD), frontotemporal dementia (FTD), and Lewy body dementia (LBD) are posing a larger population burden of dementia worldwide. Neurodegenerative dementia is one of the main challenges in public health with its main characteristics being permanent loss of memory, impairment in cognition, and impaired daily functions. The published literature about genetic studies of these disorders suggests genetic underpinning in the pathogenesis of neurodegenerative dementia. In the process of underlining the pathogenesis of NDD, growing evidence has related genetic variations in the triggering receptor expressed on myeloid cells 2 (TREM2). This review paper aims to provide a detailed information regarding the association of TREM2 and NDDs leading to dementia. A central consideration is AD that accounts for almost 50%-70% of all late-life dementias alone or in combination with other neurological disorders. Other prevalent neurodegenerative conditions that lead to dementia are also discussed. Such studies are important as they can give a comprehensive knowledge of TREM2's role in various NDDs, in order to maximize the potential for developing new therapeutic approaches.
Schizophrenia is considered one of the most disabling and severe human diseases worldwide. The etiology of schizophrenia is thought to be multifactorial and evidence suggests that DNA methylation can play an important role in underlying pivotal neurobiological alterations of this disorder. Some studies have demonstrated the effects of dietary supplementation as an alternative approach to the prevention of schizophrenia, including folic acid. However, no study has ever investigated the role of such supplementation in altering the DNA methylation system in the context of schizophrenia.
The present study aims to investigate the effects of maternal folic acid supplementation at different doses on nuclear methyltransferase activity of adult rat offspring subjected to an animal model schizophrenia induced by ketamine.
Adult female Wistar rats, (60days old) received folic acid-deficient diet, control diet, or control diet plus folic acid supplementation (at 5, 10, or 50mg/kg) during pregnancy and lactation. Afhizophrenia pathophysiology, while FA supplementation could present a protective effect since it demonstrated persistent effects in epigenetic parameters in adult offspring.
Meltdose tacrolimus (Envarsus) is marketed as a formulation with a more consistent exposure. Due to the narrow therapeutic window, therapeutic drug monitoring is essential to maintain adequate exposure. The primary objective of this study was to develop a population pharmacokinetic (PK) model of Envarsus among liver transplant patients and select a limited sampling strategy (LSS) for AUC estimation. The secondary objective was to investigate potential covariates including CYP3A/IL genotype suitable for initial dose optimization when converting to Envarsus.
Adult liver transplant patients were converted from prolonged release tacrolimus (Advagraf) to Envarsus and blood samples were obtained using whole blood and dried blood spot sampling. Subsequently the population PK parameters were estimated using nonlinear-mixed effect modelling. Demographic factors, and recipient and donor CYP3A4, CYP3A5, IL-6, -10 and -18 genotype were tested as potential covariates to explain interindividual variability.
Fifty-five patients were included. A 2-compartment model with delayed absorption was the most suitable to describe population PK parameters. The population PK parameters were as follows clearance, 3.27 L/h; intercompartmental clearance, 9.6 L/h; volume of distribution of compartments 1 and 2, 95 and 500 L, respectively. No covariates were found to significantly decrease interindividual variability. The best 3-point LSS was t = 0,4,8 with a median bias of 1.8% (-12.5-12.5).
The LSS can be used to adequately predict the AUC. No clinically relevant covariates known to influence the PK of Envarsus, including CYP3A status, were identified and therefore do not seem useful for initial dose optimization.

These data demonstrate consistent patterns of victim selection based upon the age, gender, and YSO affiliation of each victim. We created two variables to explore whether the revoked individual was "likely pedophilic (LP)" or a "mixed offender (MO)" based upon their behaviors and patterns of victim selection. Compared with the non-LPs and non-MOs, LPs and MOs had more known victims, were more often involved in other YSOs, and were more likely to have had contact with law enforcement. We utilized a public health perspective to interpret these findings, suggesting that child sexual abuse in YSOs reflects a societal problem that is not a unique phenomenon specific to programs offering services to youth.Ideas of resistance have become common in media and political discourse in contemporary times where there is growing awareness of racial violence and xenophobia. Calls to "resist" and awakenings to public life prompt questions about the kind of citizenship being cultivated, the social meanings individuals reproduce and create through participation in "resistance," and the changing sense of their positions and agency as they act in the world. Here I examine the citizen-subject that comes into being through "resistance" to racial injustice, drawing on the case of Faith in Action (formerly PICO) and its development of a theological organizing framework, the Theology of Resistance. This study analyzes the discourses and content of two public data sources-The Prophetic Resistance Podcast series and news media about prophetic resistance within the organizing network. These sources offer a means to examine the negotiated nature of political selves that are created through processes that socialize and subjugate as well as through processes wherein subjects produce and sometimes transform social positions. Findings show that centering a racial analytic and prioritizing racial justice outcomes, shifts that were made within FIA, results in the cultivation of a political subject that is reflexive about internal and external subjugating forces, relational as it discards the armor of racial hierarchy and exclusion, and constructive as it creates conditions or contexts for new political subjects through prophetic action. This study contributes to the conceptual development of organizing as a mechanism to generate social change; specifically, it offers the lens of political subjectivity as a meaningful analytic to enrich understandings of this mechanism.
To clinically evaluate the use of a titanium-reinforced PTFE mesh for vertical bone augmentation (VBA) of deficient alveolar ridges.
This case series documented consecutive patients treated for VBA with a newly developed PTFE mesh. VBA was performed in anterior and posterior, maxillary and mandibular arches using anorganic bovine bone combined with autogenous graft in a 11 ratio. Healing time from initial surgery to re-opening was recorded. Baseline vertical deficiency, absolute bone gain (gross height gained), and relative gain (percentage of defect fill with respect to the baseline deficiency) were registered.
Fifty-seven patients (65 defects) were included in the analysis. The mean baseline vertical deficiency was 5.5±2.6mm. The mean absolute bone gain was 5.2±2.4mm. A relative gain of 96.5±13.9% was achieved. Overall, 89.2% of cases showed complete regeneration, which occurred in all sites with baseline deficiencies of <5mm, in 95.6% of sites with 5-8mm deficiencies, and in 89.4% of sites with >8mm deficiencies. Each 1-mm addition to the baseline height deficiency increased the likelihood of incomplete bone regeneration by 2.5 times. Defect location had a statistically significant but a limited clinical impact on the bone height gained (<0.5mm). Complications were observed in three cases (3%).
