The phosphor has been synthesized using solid-state reaction method. The formation and surface morphology of the phosphor have been studied using X-ray diffraction (XRD) and field emission scanning electron microscopy (FE-SEM) respectively. The optical band gap of the phosphor is determined using UV-Visible spectroscopy. Different TL properties like TL glow curve, dose response, fading etc. have been investigated in detail. The TL glow curve of CaSiO3Yb phosphor consists of two prominent glow peaks. Under gamma irradiation, the main dosimetric peak is centered at 141 °C and other peak is centered at 265 °C. The dose response behaviour has been studied in the range 10 Gy-1000 Gy. However, C ion beam irradiated phosphor also consists similar TL glow curve ste use in gamma dosimetry.Xylocarpin H is a natural limonoid that has the potential to be a viable therapeutic candidate whether used alone or in combination with other medications. However, the pharmacokinetic properties of xylocarpin H are still not well understood and require further investigation. Sensitive analysis of xylocarpin H using liquid chromatography tandem triple-quadrupole mass spectrometry is challenging due to its structural feature. In the present research, a sensitive UHPLC-QTOF-MS/MS method was developed and validated to determine xylocarpin H in rat plasma. After simple methanol protein precipitation pretreatment with 50 mM esterase inhibitor bis(4-nitrophenyl)-phosphate (BNPP), xylocarpin H and Schisandrin (internal standard) were separated on a Synergi 4 u fusion-RP C18 column (50 mm × 3.0 mm, 4 µm) using isocratic elution with a mobile phase consisting of formic acid (0.1%) and acetonitrile, which were detected only within 3 min. The QTOF-MS/MS detection, which is a high-resolution mode, can accurately quantify analytes (ppm less then 5). The peak area of xylocarpin H was determined by adding the single peak areas of m/z 483.2013, m/z 465.1930 and m/z 345.1333 as the superimposed multiple product ions (SMPI) mode for data processing with optimized the high-resolution extraction widths of fragment ions at ± 0.05 Da. The S/N of the SMPI mode was found to be approximately two to seven times greater than the independent product ion mode. The linearity, precision, accuracy, and stability were completely validated for the method. Finally, the validated method was employed for the pharmacokinetic study of xylocarpin H in rats. The study indicated that the optimized high-resolution extraction widths of fragment ions and SMPI mode for data processing significantly improve the quantitative sensitivity of QTOF-MS/MS. In addition, esterase inhibitor BNPP was added in the process of sample treatment to inhibit the degradation of xylocarpin H in rat blood, which further improved the sensitivity of the quantitative method.We developed a novel approach to analyze multiple DNA targets based on microdroplet PCR coupled with denaturing gradient gel electrophoresis (MPDG) to achieve high-throughput detection of biological samples. The target DNAs were preamplified using specific primers. Subsequently, the preamplified products were separated into individual microreactors for parallel amplification with high efficiency, avoiding the interference of different primers and templates, and preventing inconsistent amplification efficiency and non-specific amplification. The final products were analyzed using denaturing gradient gel electrophoresis (DGGE). Using genetically modified (GM) maize as samples, the MPDG method could be used for the simultaneous detection of three DNA targets with an absolute limit of detection of 0.5% (w/w), with no cross reaction with other non-GM samples. The simulated sample assay of MPDG suggested that the method is suitable for practical application. The MPDG approach, with high sensitivity and specificity, could play a crucial role in the field of nucleic acid detection.Chromosomal instability (CIN) is a hallmark of the most aggressive malignancies. Here, we review the development of CIN, and examine CIN in the context of cancer evolution, tumor genomic evolution, and therapeutic resistance. We also discuss the role of whole-genome duplications, breakage-fusion-bridge cycles, ecDNA or double minutes in gene amplification promoting tumor evolution.
Seizures of frontal or temporal lobe origin can associate with vocalizations in humans. Our objective was to assess whether rats emit specific seizure-related patterns of ultrasonic vocalizations (USVs) during seizures and epileptiform activity.

Adult male Sprague-Dawley rats were treated with a single administration of pentylenetetrazol (PTZ, 50mg/kg, i.p.) and monitored with simultaneous USV and video-electroencephalogram recordings for up to 15min. USVs were detected using a deep learning algorithm (DeepSqueak-Screener) and manually annotated into the 15 previously described subcategories. The number, frequency, duration, sonographic structure, and temporal relationship of the USVs to seizures and epileptiform activity were assessed.

A total of 2147 USVs were recorded in 12 rats that expressed a total of 22 PTZ-induced seizures. Of the USVs, 77% were in the 50-kHz range (i.e., appetitive state) and 23% in the 22-kHz ( i.e., aversive state) range. More than a third (37%) of the USVs could be classifie vocalizations are a seizure-related behavioral feature in rats and recording USVs provides a novel noninvasive tool for detecting experimental seizures. Further studies are needed to explore USV occurrence during spontaneous seizures and their potential for screening novel anti-seizure drugs.4-Aminopyridine (4-AP) induces ictal-like epileptiform discharges in a variety of brain regions. These events are associated with enhanced inhibitory and excitatory synaptic neurotransmission. The relative contribution of specific subclasses of GABAergic interneurons (INs) to epileptiform activity in the 4-AP model is not well characterized. We have used genetically encoded channelrhodopsin (ChR) and Archaerhodopsin (Arch) expression in parvalbumin (PV), somatostatin (SST) and vasoactive intestinal polypeptide (VIP) INs to investigate the role of interneuron subclasses in 4-AP-induced epileptiform discharges. Whole-cell patch-clamp recordings were obtained from L5 pyramidal cells (PYRs) in somatosensory cortex of 30-to-70-day old mice. In the presence of 100 µM 4-AP, photostimulation of ChR in PV and SST, but not VIP INs, evoked epileptiform discharges similar to spontaneous and electrically evoked events. Light activation of Arch in PV INs was more effective in reducing epileptiform activity compared to SST and VIP INs. Epileptiform discharges were evoked at offset of Arch induced hyperpolarizations in PV and SST interneurons but not VIP INs. PV and SST INs could both initiate and inhibit 4-AP-induced epileptiform activity in L5 PYRs. VIP INs did not contribute significantly to eliciting or inhibiting epileptiform discharges. These results suggest that subclasses of INs contribute differently to the initiation and modulation of epileptiform discharges in cortical networks.
To describe the clinical features of childhood epilepsy in Qatar.

A retrospective cross-sectional chart review analysis was conducted at the only tertiary pediatric hospital in Qatar in 1422 patients with epilepsy followed between November 2016 and October 2019.

55% (781) were males and 70% were non-Qatari. Age of epilepsy onset was in the neonatal period in 9% (114/1207 patients). In the non-neonatal cohort, mean age of onset was 4yrs 9mos (±1.4mos). Focal epilepsy was the predominant epilepsy type in 45% (594/1314 patients) versus generalized epilepsy in 37% and combined focal/generalized epilepsy in 12%. Etiology was unknown in most children (782/1363, 57%) whereas structural and genetic causes represented 23% and 11% of cases respectively. No differences in epilepsy type and etiology were found between different ethnic groups. Children with genetic or structural epilepsies had an earlier epilepsy onset compared to those with unknown etiologies. At the last follow up, only 36% of patients were seizurnt of co-morbidities and medically-intractable epilepsy.
This large multi-ethnic population-based study confirms that the prevalence, incidence and clinical features of epilepsy in Qatar is in accordance with other epidemiologic studies and highlights risk factors for the development of co-morbidities and medically-intractable epilepsy.
To understand quality of life (QoL) perceptions of people with epilepsy (PWE) through knowledge, attitude, and behavior (KAB); PWE; their families (PWEf); and the general population (GPop).

This descriptive study was conducted in Jakarta and its surrounding cities from January to December 2019. PWE were recruited from outpatient clinics. PWEf were caregivers who lived with PWE. GPop were age matched, randomly selected, and interviewed for public events. The perception of QoL was scaled from 1 to 5 (1=very poor to 5=very good). KAB was obtained from open- and closed-ended questionnaires, scaled from 1 to 5 (1=strongly disagree to 5=strongly agree). The differences in each group were analyzed using t-tests and analysis of variance.

We interviewed 371 participants, predominantly female and senior high school graduates. Unemployment and singlehood were higher in PWEs. QoL perception in PWE was similar to GPop (3.01 [0.75] vs. 3.07 [0.76], p=0.49), yet lower in PWEf (2.78 [0.76]; p<0.05). According to PWEB. Despite similar knowledge of PWE and PWEf, better perceptions came from PWE and GPop. The reluctance to form deeper bonds between GPop and PWE, along with PWEf's skepticism, could lead to low self-esteem, unemployment, and unmarried rates. Further studies are required to elaborate on these issues.Synaptic Vesicle Glycoprotein 2A (SV2A) has been proposed as a presynaptic marker in several neurological disorders. Not only is SV2A the target for the antiepileptic drug levetiracetam, but also considered a marker of mature pre-synapses. In this study, we aimed to assess the binding of [3H]UCB-J as a selective radioligand for SV2A to visualize and determine changes during different stages of epileptogenesis by in-vitro autoradiography in rat models of temporal lobe epilepsy. Two different kainic acid (KA) injection routes were used to model temporal lobe epilepsy in the rat; a systemic (10 mg/kg KA injected intraperitoneally) and a local model (1.875 mM KA injected intrahippocampally). Brain tissue was sampled at different time points after the initial status epilepticus and semi-quantitative [3H]UCB-J autoradiography was performed to determine temporal and spatial changes under the progression of epileptogenesis. A decrease in [3H]UCB-J binding was observed in many brain areas in the acute phases after both types of kainic acid administration.
