Thermoresponsive Fluorescence Changes Depending on Au@pNIPAM Nanoparticles.
The frequent use of insecticides to manage soybean aphids, Aphis glycines (Hemiptera Aphididae), in the United States has contributed to field-evolved resistance. Pyrethroid-resistant aphids have nonsynonymous mutations in the voltage-gated sodium channel (vgsc). We identified a leucine to phenylalanine mutation at position 1014 (L1014F) and a methionine to isoleucine mutation (M918I) of the A. glycines vgsc, both suspected of conferring knockdown resistance (kdr) to lambda-cyhalothrin. We developed molecular markers to identify these mutations in insecticide-resistant aphids. We determined that A. Selleck Thiamet G glycines which survived exposure to a diagnostic concentration of lambda-cyhalothrin and bifenthrin via glass-vial bioassays had these mutations, and showed significant changes in the resistance allele frequency between samples collected before and after field application of lambda-cyhalothrin. Thus, a strong association was revealed between aphids with L1014F and M918I vgsc mutations and survival following exposure to pyrethroids. Specifically, the highest survival was observed for aphids with the kdr (L1014F) and heterozygote super-kdr (L1014F + M918I) genotypes following laboratory bioassays and in-field application of lambda-cyhalothrin. These genetic markers could be used as a diagnostic tool for detecting insecticide-resistant A. glycines and monitoring the geographic distribution of pyrethroid resistance. We discuss how generating these types of data could improve our efforts to mitigate the effects of pyrethroid resistance on crop production.LKB1 loss of function is one key oncogenic event in lung cancer. Clinical data suggest that LKB1 loss of function is associated with patients' smoking status. The responsible ingredients and molecular mechanisms in tobacco for LKB1 loss of function, however, are not defined. In this study, we reported that NNAL, a major metabolite of a tobacco-specific carcinogen NNK, induces LKB1 phosphorylation and its loss of function via the β-AR/PKA signaling pathway in an isomer-dependent manner in human lung cancer cells. NNAL exposure also resulted in enhanced lung cancer cell migration and chemoresistance in an LKB1-dependent manner. A 120-day NNAL exposure in lung cancer cells, mimicking its chronic exposure among smokers, resulted in more prominent LKB1 phosphorylation, cell migration, and chemoresistance even in the absence of NNAL, indicating the long-lasting LKB1 loss of function although such an effect eventually disappeared after NNAL was removed for two months. These observations were confirmed in a lung cancer xenograft model. More importantly, human lung cancer tissues revealed elevated LKB1 phosphorylation in comparison to the paired normal lung tissues. These results suggest that LKB1 loss of function in human lung cancer could be extended to its phosphorylation, which may be mediated by NNAL from tobacco smoke in an isomer-dependent manner via the β-AR/PKA signaling pathway.NSCLC is the first cause of cancer-related deaths in China and threatens life expectancy of the people. Novel drugs and treatment strategies are urgently required. Capsaicin is noticed as a potential new drug for lots of tumors due to its anti-proliferative effect on cancer cells. Our study evaluated the roles of capsaicin in NSCLC cells (A549 and NCI-H23) and further explored its underlying mechanisms. Effect of capsaicin treatment on cell viability was determined by MTT assay and IC50 values for A549 and NCI-H23 cells were ascertained. The iron kit detected the total iron levels and the ferric divalent ions levels in A549 and NCI-H23 cells. GSH kit was used to detect the expression of GSH in A549 and NCI-H23 cells. Additionally, mRNA and protein levels of SLC7A11 and GPX4 were analyzed by real-time PCR and western blot analysis. Through MTT assay, we found that 200 μM capsaicin in cultured A549 cells for 48 h could reach the IC50 value, and the condition was 100 μM and 48 h for NCI-H23 cells. Capsaicin increased total iron levels and ferrous ion levels in A549 and NCI-H23 cells in contrast with the control group, whereas the levels of GSH was reduced in contrast with the control group. Besides, mRNA and protein levels of SLC7A11 and GPX4 were decreased significantly in A549 and NCI-H23 cells treated with capsaicin in contrast with the control group. Our study indicated that capsaicin inhibited the proliferation of A549 and NCI-H23 cells and induced ferroptosis by inactivating SLC7A11/GPX4 signaling. Capsaicin could be used as a potential anticancer agent in the treatment of NSCLC.Improvements in understanding the pathophysiology of the different benign liver nodules have refined their nosological classification. New criteria have been identified using imaging, histology and molecular analyses for a precise diagnosis of these tumours. Improvement in the classification of liver tumours provides a more accurate prediction of disease progression and has modified patient management. Haemangioma and focal nodular hyperplasia, the most common benign liver tumours that develop in the absence of chronic liver disease, are usually easy to diagnose on imaging and do not require specific treatment. This Review aims to explain the main pathogenic mechanisms of benign liver tumours and how this knowledge could influence clinical practice.Phosphorus (P) deficiency is the main hurdle in achieving sustainable crop production ps especially in calcareous soils. Using bio-fertilizers like phosphate solubilizing bacteria (PSB) could be a useful approach for sustainable P management as they improve P availability in soil via dissolution, desorption and mineralization reactions. In addition, application of organic amendments with PSB could further ameliorate soil conditions for sustainable management of immobilized nutrients in calcarious soils. Therefore, we performed pot experiment to study the role of PSB in nullifying antagonistic effects of liming (4.78, 10, 15 and 20%) on P availability from poultry manure (PM), farm yard manure (FYM), single super phosphate (SSP) and rock phosphate (RP) in alkaline soils. PSB inoculation improved wheat growth, P availability and stimulated soil acidification over control regardless of P sources and lime levels. Soil calcification adversely affected plant growth, P nutrition, induced soil salinity and alkalinity, however, PSB and manures application potentially nullified such harmful effects over mentioned traits. Individually, organic sources were superior than mineral sources however, the performance of mineral fertilizers with PSB was at par to sole application of manures. Furthermore, application of RP with PSB proved as effective as sole SSP. Therefore, using PSB as bio-fertilizer has huge potential for improving P availability in calcareous soils.Prohibitins are highly conserved eukaryotic proteins in mitochondria that function in various cellular processes. The roles of prohibitins in fungal virulence and their regulatory mechanisms are still unknown. Here, we identified the prohibitins ChPhb1 and ChPhb2 in a plant pathogenic fungus Colletotrichum higginsianum and investigated their roles in the virulence of this anthracnose fungus attacking crucifers. We demonstrate that ChPhb1 and ChPhb2 are required for the proper functioning of mitochondria, mitophagy and virulence. ChPhb1 and ChPhb2 interact with the autophagy-related protein ChATG24 in mitochondria, and ChATG24 shares similar functions with these proteins in mitophagy and virulence, suggesting that ChATG24 is involved in prohibitin-dependent mitophagy. ChPhb1 and ChPhb2 modulate the translocation of ChATG24 into mitochondria during mitophagy. The role of ChATG24 in mitophagy is further confirmed to be conserved in plant pathogenic fungi. Our study presents that prohibitins regulate fungal virulence by mediating ATG24-assisted mitophagy.
Late-Onset Neonatal Sepsis (LOS) is a rare condition, involving widespread infection, immune disruption, organ dysfunction, and often death. Because exposure to pathogens is not completely preventable, identifying susceptibility factors is critical to characterizing the pathophysiology and developing interventions. Prior studies demonstrated both genetics and infant sex influence susceptibility. Our study was designed to identify LOS associated genetic variants.

We performed an exploratory genome wide association study (GWAS) with 224 LOS cases and 273 controls from six European countries. LOS was defined as sepsis presenting from 3 to 90 days of age; diagnosis was established by clinical criteria consensus guidelines. We tested for association with both autosomal and X-chromosome variants in the total sample and in sex-stratified analyses.

In total, 71 SNPs associated with neonatal sepsis at p < 1 × 10
in at least one analysis. Most importantly, sex-stratified analyses revealed associations with multiple SNPs (28 in males and 16 in females), but no variants from single-sex analyses associated with sepsis in the other sex. Pathway analyses showed NOTCH signaling is over-represented among genes linked to these SNPS.

Our results indicate genetic susceptibility to LOS is sexually dimorphic and corroborate that NOTCH signaling plays a role in determining risk.

Camera-trap data has been analyzed through several methods to assess the temporal and spatio-temporal niche partitioning. However, different analytical approaches used to may evaluate niche partitioning detect different results. In this study, we evaluated the temporal or spatio-temporal partitioning among sympatric medium-sized carnivores, red foxes, raccoon dogs, and Japanese martens, based on three analytical methods-the temporal overlap, temporal co-occurrence, and time-to-encounter analysis-to evaluate. From May to October 2019 and 2020, we obtained the activity of the target species using camera-traps in northeastern Japan. We analyzed the data with the coefficient of temporal overlap, probabilistic co-occurrence analysis, checkerboard score, and multi-response permutation procedures. The results of the assessment of the niche partitioning differed depending on the analytical methods based on temporal and spatio-temporal partitioning.
