Human dicer is an enzyme that cleaves pre-miRNAs into miRNAs. Several models have been developed to predict human dicer cleavage sites, including PHDCleav and LBSizeCleav. Given an input sequence, these models can predict whether the sequence contains a cleavage site. However, these models only consider each sequence independently and lack interpretability. Therefore, it is necessary to develop an accurate and explainable predictor, which employs relations between different sequences, to enhance the understanding of the mechanism by which human dicer cleaves pre-miRNA.
In this study, we develop an accurate and explainable predictor for human dicer cleavage site - ReCGBM. We design relational features and class features as inputs to a lightGBM model. Computational experiments show that ReCGBM achieves the best performance compared to the existing methods. Further, we find that features in close proximity to the center of pre-miRNA are more important and make a significant contribution to the performance improvement of the developed method.
The results of this study show that ReCGBM is an interpretable and accurate predictor. Besides, the analyses of feature importance show that it might be of particular interest to consider more informative features close to the center of the pre-miRNA in future predictors.

Monochasma savatieri is a medicinal root hemiparasitic herb that extracts water and nutrients from the host plant via a haustorium. M. savatieri exhibits an enhanced growth after the establishment of parasite-host associations, but little is known about the molecular mechanism responsible. In this study, endogenous hormones, RNA sequencing and small RNA sequencing analysis were performed on M. savatieri before and after establishment of parasite-host associations.
When grown with the host, decreased contents of jasmonic acid (JA) and indole-3-acetic acid (IAA) and increased abscisic acid (ABA) content were observed in M. savatieri with the established parasitic relationship. When grown with the host, 46,424 differentially expressed genes (DEGs) and 162 differentially expressed miRNAs (DEmiRs) were identified in the comparison between M. savatieri with the established parasitic relationship and without the established parasitic relationship. Analysis of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) showed that these DEGs and targets of DEmiRs mostly participated in plant hormone signal transduction, starch and sucrose metabolism, carbohydrate metabolism, cell growth and death, and transport and catabolism. Furthermore, correlation analysis of mRNA and miRNA revealed that 10 miRNA-target pairs from novel_mir65, novel_mir40, novel_mir80, miR397-5p_1, novel_mir36, novel_mir25 and novel_mir17 may have important roles in regulating the parasitic development of M. savatieri.

Lesion mimics (LMs) are disease-like symptoms that occur randomly on plant green leaves in the absence of pathogens. A previous study showed that LMs are related to enhanced resistance to a broad spectrum of diverse pathogen races and programmed cell death (PCD). Stripe rust is a globally epidemic fungal disease that can substantially reduce the quality and yield of crops. The development of resistant cultivars is an economical and environmentally friendly way to enhance the adaptability and yield stability of crops instead of the use of fungicide applications.
In this study, a novel LM gene affording Pst resistance was identified and mapped with molecular markers developed for marker-assisted selection (MAS)-based wheat breeding. In this study, a novel LM gene named lm4, which is closely linked (8.06 cM) to SSR markers Xgwm210 and Xgwm455, was identified by using a Yanzhan 1/Neixiang 188 RIL population. The genetic distance of lm4 was then narrowed such that it was flanked by SSR markers with 0.51 cM and 0.77 cM intervals. Two SSR markers, lm4_01_cib and lm4_02_cib, were developed based on the content in the Chinese Spring genome database and wheat 660 K SNP results; these markers can be used to conduct MAS of LMs in wheat. The results also showed that lm4 significantly improved the resistance of stripe rust in wheat.
Therefore, lm4 is associated with stripe rust resistance, which may provide theoretical support for future crop disease-resistance breeding and for understanding the plant apoptosis mechanism.

The ablation targets of atrial fibrillation (AF) are adjacent to bronchi and pulmonary arteries (PAs). We used computed tomography (CT) to evaluate the anatomical correlation between left atrium (LA)-pulmonary vein (PV) and adjacent structures.
Data were collected from 126 consecutive patients using coronary artery CT angiography. MK-5348 supplier The LA roof was divided into three layers and nine points. The minimal spatial distances from the nine points and four PV orifices to the adjacent bronchi and PAs were measured. The distances from the PV orifices to the nearest contact points of the PVs, bronchi, and PAs were measured.
The anterior points of the LA roof were farther to the bronchi than the middle or posterior points. The distances from the nine points to the PAs were shorter than those to the bronchi (5.19 ± 3.33mm vs 8.62 ± 3.07mm; P < .001).
