Background About 5-10% of incidences of breast cancers have been reported as a result of germline mutations of BRCA genes. However, the mutational spectrum of BRCA1 and BRCA2 genes among breast cancer Saudi women patients is inadequate at present. Therefore, the present study aimed to report the specific germinal mutation of BRCA1 and BRCA2 in the entire coding regions, to investigate the prevalence rate of BRCA1 & BRCA2 mutations among Saudi women and the effect of these mutations, both benign and malignant tumors. Methodology A total of 270 tissue samples of benign and malignant breast tumors were collected from Saudi women patients, Riyadh, Saudi Arabia. Examination of BRCA1 and BRCA2 germline mutations was performed using heteroduplex DNA analysis (HDA) or single-stranded conformation analysis (SSCA). 177 breast cancer women with malignant tumors and 93 with benign tumors were enrolled in the study. A total of 62 out of 177 breast cancer patients carried a BRCA1 or BRCA2 mutation (54 BRCA1 and 8 BRCA2). The analysis was done using the Sanger sequence assay. Results Point and frameshift mutations through the entire coding area of the two genes indicated that all the mutations were germline alterations and of early-onset breast cancers. The mean ages of diagnosed breast cancer women for BRCA1 and BRCA2 mutation carriers were 36.3 (± 3.5) and 37.9 (± 3.7) years, whereas that of benign control was 35(± 2.5) years. Conclusion Point and frameshift mutations across the entire coding region of BRCA1 and BRCA2 are responsible for many breast cancers cases. Each year, hunters from 12 of the 27 European Union (EU) countries and the UK shoot over 6 million large game mammals, 12 million rabbits and hares and over 80 million birds. They support an international game meat market worth over 1.1 thousand million Euros. Animals shot with lead ammunition frequently contain lead fragments in the carcass which contaminate meals made from game meat with concentrations of lead substantially above the maximum allowable level (ML) set by European Commission Regulation EC1881/2006 for meat from domesticated animals. This poses a health risk to frequent consumers of wild-shot game meat, with children and pregnant women being particularly vulnerable. Total replacement of lead rifle and shotgun ammunition with available non-toxic alternatives is needed for all hunting in EU nations to prevent exposure of humans and wildlife to ammunition-derived lead and to allow the depletion of the long-term environmental legacy of lead from spent ammunition. We propose that EC1881/2006 is amended to incorporate an ML for game meats as a supplementary measure to the replacement of lead ammunition. This would harmonise food safety standards for lead in meats traded across and imported into the EU.Mitigation of eutrophication, intensified by excessive nutrient load discharge in wastewaters regulated by restrictive legal requirements, remains one of today's most important global problems. Despite implementation of the Water Framework Directive, the Urban Wastewater Directive and the HELCOM recommendations, the actual condition of surface water is still not satisfactory. In response to the above, the study presents an alternative approach for surface water protection against eutrophication based on the selection of appropriate nutrient removal technologies. An activated sludge model simulation was used to enable the identification of environmentally justified nutrient removal systems with lowest eutrophication potential of treated wastewater conditioned by bioavailable nutrient forms content. Based on the outcome of the study, the 3-stage Bardenpho system was identified as the most efficient for bioavailable phosphorus removal, while the Johannesburg system proved to have the highest efficiency for bioavailable nitrogen removal. The proposed eutrophication mitigation approach underlines the need for a reconsideration of current legal regulations which ignore nutrient bioavailability and key eutrophication limiting factors.Uncertain future payoffs and irreversible costs characterize investment in climate change adaptation and mitigation. Under these conditions, it is relevant to analyze investment decisions in a real options framework, as this approach takes into account the economic value associated with investment time flexibility. In this paper, we provide an overview of the literature adopting a real option approach to analyze investment in climate change adaptation and mitigation, and examine how the uncertain impacts of climate change on the condition of the human environment, risk preferences, and strategic interactions among decisions-makers have been modeled. We found that the complex nature of uncertainties associated with climate change is typically only partially taken into account and that the analysis is usually limited to decisions taken by individual risk neutral profit maximizers. Our findings call for further research to fill the identified gaps.Ethiopia aims to restore 15 million ha degraded forests and woodlands, but effects on the potentially contrasting goals of long-term carbon storage, biodiversity and sustainable livelihoods are unknown. To quantify the effects of grazing exclusion on vegetation and fire behaviour, we established six 30 × 30 m fenced exclosures with grazed controls, in a mesic wooded savanna. Experimental burns were done after 1.5 years. Tree seedlings were few but more common inside fences. Field layer cover and biomass increased inside fences, and grass species increased in numbers and cover. Fire intensity was higher inside fences, killing shrubs and saplings but not mature trees. Interviews confirmed that overgrazing has resulted in "cool fires", causing shrub encroachment. ONO-7475 molecular weight High-intensity fires occurred in the 1980s after a zoonotic disease killed most livestock. Short-term increase in carbon storage through fire and grazing exclusion may lead to loss of pasture, and in the long-term increased wildfire risk.Introduction Children may experience medical trauma when undergoing medical procedures even when procedures are minor. While virtual reality (VR) is effective for managing procedural pain and anxiety, few studies address how families feel about using VR. We explore pediatric patient and guardian views regarding the acceptability of using VR during procedures to mitigate medical trauma. Methods Semi-structured qualitative interviews with 18 patient-guardian dyads at a tertiary outpatient infusion center for inflammatory bowel disease (IBD) treatment. Interviews explored how VR may change the infusion experience, including benefits, risks, and recommendations for clinical integration. Interviews were recorded, transcribed, and analyzed in ATLAS.ti. Two coders used a 3-step coding approach to (1) identify themes; (2) develop a codebook and code transcripts using the constant comparative method; and (3) describe themes/patterns. Results Potential benefits of VR were distraction from infusion-related anxiety and pain and generating excitement for the appointment.
