In this study, magnetic activated carbon (MAC) was added as a stabilization and removal adsorbent to native pond sediment with known contamination of HMs and PAHs to reduce the tissue residues and dietary risks of HMs and PAHs in a model aquaculture species (Venerupis philippinarum) in the course of a 28-day bioaccumulation experiment. Meanwhile, passive sampling techniques based on diffusive gradient in thin films (DGT) and polydimethylsiloxane (PDMS) were applied to sense the bioavailable fraction of HMs and PAHs in sediment during the stabilization process. The results showed that 3% dosage of MAC to sediment achieved the most cost-effective stabilization for HMs and PAHs. A remarkable decrease was observed with the tissue residues of HMs and PAHs in V. selleck inhibitor philippinarum (28-47% for HMs and ~76% for ∑PAHs), which was quantitatively linked to the decline in their bioavailable concentrations in sediment pore water (31-46% for HMs and ~76% for ∑PAHs). Consequently, the target hazard quotients (THQs) posed by HMs and incremental lifetime cancer risks (ILCRs) by PAHs in V. philippinarum were reduced by 38% and 46%, respectively. Along with the magnetic recovery of ~70% MAC from the sediment, HMs (4.8-13%) and PAHs (2-60%) can be effectively removed. We further established a multi-domain equilibrium sorption model that was able to predict the optimal amendment of MAC for quantitative mitigation of bioavailable PAHs in sediment pore water within a certain range of MAC dosage. Future studies are warranted to explore the applicability domain of MAC for in situ remediation in aquaculture ponds to ensure the quality of farming organisms or to serve other purposes in aquatic systems.Consumption of mercury (Hg) contaminated rice can be a major environmental health issue but the toxicokinetics is not well known. Hg isotopes have been shown to be good tracers in studying Hg exposure and metabolic processes. We established a Hg mass balance and Hg isotope model in rats fed with Hg contaminated rice (THg 51.3 ng/g; MeHg 25 ng/g) for 90 days to investigate Hg toxicokinetics. Overall 80% of feeding THg was recovered in rat body and excrement, while the excrement accounted for 55% of total observed THg in rats. Feces were the main route of Hg elimination in rats, while urinary excretion was negligible. However, only 32% of utilized MeHg was recovered in rats, indicating significant demethylation of MeHg in rat body. Positive net fractionations of δ202Hg (relative to the feeding rice) were observed in hair and blood samples (1.21‰ and 1.25‰, respectively), which have similar trend with the results obtained in human hair study, exhibiting higher δ202Hg values (2‰- 3‰) than consumed fish and rice. Most importantly, we observed negative net fractionations in feces (-0.44‰), which confirmed the missed Hg with negative δ202Hg signal. We concluded that mass balance and Hg isotope are useful tools for quantifying toxicokinetics of Hg. Demethylation of MeHg in the intestine were the important detoxification process in rat body characterizing with negative net Hg fractionations in feces.Osteoarthritis (OA) is a debilitating disease with no approved disease-modifying therapies. Among the challenges for developing treatment is achieving targeted drug delivery to affected joints. This has contributed to the failure of several drug candidates for the treatment of OA. Over the past 20 years, significant advances have been made in antisense oligonucleotide (ASO) technology for achieving targeted delivery to tissues and cells both in vitro and in vivo. Since ASOs are able to bind specific gene regions and regulate protein translation, they are useful for correcting aberrant endogenous mechanisms associated with certain diseases. ASOs can be delivered locally through intra-articular injection, and can enter cells through natural cellular uptake mechanisms. Despite this, ASOs have yet to be successfully tested in clinical trials for the treatment of OA. Recent chemical modification to ASOs have further improved cellular uptake and reduced toxicity. Among these are locked nucleic acid (LNA)-based ASOs, which have shown promising results in clinical trials for diseases such as hepatitis and dyslipidemia. Recently, LNA-based ASOs have been tested both in vitro and in vivo for their therapeutic potential in OA, and some have shown promising joint-protective effects in preclinical OA animal models. In order to accelerate the testing of ASO therapies in a clinical trial setting for OA, further investigation into delivery mechanisms is required. In this review article, we discuss opportunities for viral-, particle-, biomaterial-, and chemical modification-based therapies, which are currently in preclinical testing. We also address potential roadblocks in the clinical translation of ASO-based therapies for the treatment of OA, such as the limitations associated with OA animal models and the challenges with drug toxicity. Taken together, we review what is known and what would be useful to accelerate translation of ASO-based therapies for the treatment of OA.Fractures are common injuries in children, but their underlying biological and environmental risk factors are not well known. Maternal alcohol consumption during pregnancy is a known risk factor for bone malformations and impaired growth, in connection with Fetal Alcohol Spectrum Disorders (FASD). There is evidence that even lower doses of alcohol than what is needed for FASD can cause changes in the developing bone. Birth weight and length may also associate to childhood fractures. The aim of this study was to find out whether there exist associations between maternal alcohol use during pregnancy, birth weight or length and fractures of the long bones in childhood. A prospective birth cohort was performed, including all women in Northern Finland with an expected date of delivery between July 1985 and June 1986, and their offspring (N = 9432). The National Hospital Discharge Register (NHDR) provided the information on inpatient treated fractures. The subjects who declined participation or were treated as outpatient were excluded.
