Gaucher disease (GD) is one of the most prevalent lysosomal disorders, with an estimated incidence of 1 in 40,000 live births worldwide. Skeletal involvement is one of the main features of GD, causing morbidity and impacting long-term quality of life in patients with type 1 GD.

To characterize bone marrow infiltration in patients with type 1 GD followed at the Gaucher Disease Referral Center of Porto Alegre, Brazil, and to assess whether the Bone Marrow Burden score (BMB) correlates with clinical or laboratory parameters. We also evaluated whether the BMB score is a suitable parameter for long-term follow-up of patients with type 1 GD.

All included patients underwent magnetic resonance imaging for BMB score calculation at baseline, 1year, and every other year thereafter or as clinically indicated from 2012 to 2018.

The BMB score tended to decrease during the first 5years of treatment, at a rate of -1.08 points per year; after the 5-year mark, BMB tended to remain stable.

Previous research suggests that imitation as a social process depends on the similarity between interaction partners. However, some of the experiments supporting this notion could not be replicated and all of the supporting experiments manipulated not only similarity between actor and observer, but also other features. Thus, the existing evidence leaves open whether similarity as such moderates automatic imitation. To directly test the similarity account, in four experiments, we manipulated participants' focus on similarities or differences while holding the stimulus material constant. In Experiment 1, we presented participants with a hand and let them either focus on similarities, differences, or neutral aspects between their own hand and the other person's hand. The results indicate that focusing on similarities increased perceived similarity between the own and the other person's hand. In Experiments 2 to 4, we tested the hypothesis that focusing on similarities, as compared with differences, increases automatic imitation. Experiment 2 tested the basic effect and found support for our prediction. Experiment 3 and 4 replicated this finding with higher-powered samples. Exploratory investigations further suggest that it is a focus on differences that decreases automatic imitation, and not a focus on similarities that increases automatic imitation. Theoretical implications and future directions are discussed.Nuclear simulation methods were applied to two systems that investigate radon transport within geological porous media a) a laboratory system built to test, under controlled climate conditions, the effect of temperature on radon transport, and b) a field monitoring system comprising gamma and alpha detectors in an abandoned water well. The use of Monte Carlo simulations of NaI and BGO scintillation detectors in continuous underground radon measurements by gamma counting, to estimate the photon flux in the detector volume, is presented. The advantages of shielding side-view NaI detectors were demonstrated for a laboratory system containing ground phosphate rock, including avoiding high counting rates and reducing the effective source volume in radon transport studies. The gross gamma counting procedure was shown to result in a lower uncertainty than spectrometric measurement, by at least a factor of two, despite it being a simpler and more suitable procedure for field measurements. The calculation of simulated source volumes for a BGO detector in a borehole and the measurements in the field support the assumption that the gamma signal comes primarily from radon flowing in the bedrock's air-filled pores. As a practical outcome of this study, positioning the detector a few cm off-center from the borehole's axis increased the gamma counting efficiency; however, measurements in groundwater taken too close to the iron casing had a lower detection efficiency. The conversion factor from the scintillator signal to the radon activity concentration, for the laboratory system, was calculated. Monte Carlo simulations demonstrated the advantages of the gross counting procedures using gamma scintillation detectors in field underground high-frequency radon monitoring.Significant developments in the understanding and manipulation of microbial metabolism have enabled the use of engineered biological systems toward a more sustainable energy and materials economy. While developments in metabolic engineering have primarily focused on the conversion of carbohydrates, substantial opportunities exist for using these same principles to extract value from more heterogeneous and toxic waste streams, such as those derived from lignin, biomass pyrolysis, or industrial waste. Funneling heterogeneous substrates from these streams toward valuable products, termed biological funneling, presents new challenges in balancing multiple catabolic pathways competing for shared cellular resources and engineering against perturbation from toxic substrates. Solutions to many of these challenges have been explored within the field of lignin valorization. Despite recent progress in image-to-image translation, it remains challenging to apply such techniques to generate high-quality medical images. This paper proposes a novel framework named Multi-Cycle GAN, which uses the Pseudo-Cycle Consistent module to control the consistency of generation and the domain control module to provide additional identical constraints. Besides, we design a new generator named Z-Net to improve the accuracy of anatomy details. Extensive experiments show that Multi-Cycle GAN outperforms state-of-the-art CT synthesis methods such as Cycle GAN, which improves MAE to 0.0416, ME to 0.0340, PSNR to 39.1053.Global warming imposes a major threat to plant growth and crop production. In some plants including Arabidopsis thaliana, elevated temperatures induce a series of morphological and developmental adjustments, termed thermomorphogenesis to facilitate plant cooling under high-temperature conditions. Plant thermal response is suppressed by histone variant H2A.Z. At warm temperatures, H2A.Z is evicted from nucleosomes at thermo-responsive genes, resulting in their expression changes. However, the mechanisms that regulate H2A.Z eviction and subsequent transcription changes are largely unknown. Here, we show that the INO80 chromatin remodeling complex (INO80-C) promotes thermomorphogenesis and activates the expression of thermo-responsive and auxin-related genes. INO80-C associates with PHYTOCHROME-INTERACTING FACTOR 4 (PIF4), a potent regulator in thermomorphogenesis, and mediates temperature-induced H2A.Z eviction at PIF4 targets. Moreover, INO80-C directly interacts with COMPASS-like and transcription elongation factors to promote active histone modification Histone H3 lysine 4 trimethylation (H3K4me3) and RNA Polymerase II (RNA Pol II) elongation, leading to the thermal induction of transcription. Notably, transcription elongation factors SPT4 and SPT5 are required for the H2A.Z eviction at PIF4 targets, suggesting the cooperation of INO80-C and transcription elongation in H2A.Z removal. Our results demonstrate that the (PIF4)-(INO80-C)-(COMPASS-like)-(transcription elongator) module controls plant thermal response, and establish a link between H2A.Z eviction and active transcription.Post-translational modifications (PTMs), including phosphorylation and persulfidation, regulate the activity of SNF1-RELATED PROTEIN KINASE2.6 (SnRK2.6). Here, we report how persulfidations and phosphorylations of SnRK2.6 influence each other. The persulfidation of cysteine C131/C137 alters SnRK2.6 structure and brings the serine S175 residue closer to the aspartic acid D140 that acts as ATP-γ-phosphate proton acceptor, thereby improving the transfer efficiency of phosphate groups to S175 to enhance the phosphorylation level of S175. Interestingly, we predicted that S267 and C137 were predicted to lie in close proximity on the protein surface and found that the phosphorylation status of S267 positively regulates the persulfidation level at C137. Analyses of the responses of dephosphorylated and depersulfidated mutants to abscisic acid and the H2S-donor NaHS during stomatal closure, water loss, gas exchange, Ca2+ influx, and drought stress revealed that S175/S267-associated phosphorylation and C131/137-associated persulfidation are essential for SnRK2.6 function in vivo. In light of these findings, we propose a mechanistic model in which certain phosphorylations facilitate persulfidation, thereby changing the structure of SnRK2.6 and increasing its activity.Vaccine hesitancy-defined by the World Health Organization (WHO) as a "delay in acceptance or refusal of vaccines despite availability of vaccination services"-is not a recent phenomenon. More problematic are the means by which concerns over vaccine hesitancy are communicated by a new landscape of digital communication, generating what has been described as an "infodemic" in which an overabundance of information-both factual and misinformation-contributes to hesitancy. In this review, we discuss the background and current drivers of vaccine hesitancy and the evidence base for strategies to combat this. We highlight the important role the allergy/immunology community could have in working to mitigate vaccine hesitancy, particularly with respect to the current coronavirus disease 2019 (COVID-19) pandemic.
The coronavirus disease 2019 (COVID-19) pandemic dramatically increased the number of patients requiring treatment in an intensive care unit or invasive mechanical ventilation worldwide. Delirium is a well-known neuropsychiatric complication of patients with acute respiratory diseases, representing the most frequent clinical expression of acute brain dysfunction in critically ill patients, especially in those undergoing invasive mechanical ventilation. Among hospitalized patients with COVID-19, delirium incidence ranges from 11% to 80%, depending on the studied population and hospital setting.

We retrospectively studied consecutive hospitalized adult (≥18 y) patients with confirmed COVID-19 pneumonia from March 15 to July 15, 2020, in a tertiary-care hospital in Mexico City. Delirium was assessed by the attending physician or trained nurse, with either the Confusion Assessment Method for the intensive care unit or the Confusion Assessment Method brief version, according to the appropriate diagnostic tool for each hospital setting.
