As a prime synthetic peptide, KDI, is responsible for the neurite outgrowth of human embryonic neurons. In this study, we have designed, modified a KDI derivative and synthesized by replacing isoleucine (I) with Pro (P) amino acid at C-terminal to enhance its potency towards neurite growth. -Cys-Gly-Cys (-CGC) N2S2 motif was also incorporated in the present design for peptide radiolabeling. The modified peptide showed a better binding with the desired 3T1M receptor for neurite growth. The peptide was synthesized using solid phase peptide synthesis and Fmoc-strategy with more than 80% yield. The receptor binding studies of 99mTc-N2S2-KDP in Neuro2A cell lines showed Kd value in 31 nM range and the complex showed appreciable brain uptake in mice. The results on human SH-SY5Y indicate that the unlabeled N2S2-KDP may perhaps be useful for neurite growth in neurodegenerative disorder.This study aimed to obtain tyrosinase inhibitors for treating hyperpigmentation. A series of cinnamyl ester analogues were designed and synthesized with cinnamic acid (CA) and peaonol compounds. The safety, melanin content and inhibitory effects of all target compounds were evaluated. In the enzymatic activity test, the inhibitory rate of compounds 8, 13 and 14 had stronger inhibitory activity with the IC50 values of 20.7 μM, 13.98 μM and 15.16 μM, respectively than the positive drug kojic acid (IC50 with 30.83 μM). The cytotoxicity evaluation showed that compounds 13 and 14 have higher safety than the other compounds to the proliferation of B16F10 cells. The result of the melanocyte test supported that compound13 has stronger cellular tyrosinase inhibitory activity than kojic acid and arbutin at 100 μM and 200 μM. The enzyme kinetics mechanism revealed that compound 13 was a non-competitive inhibitor while compounds 8 and 14 were mixed inhibitors. For the experiments of melanin content and tyrosinase activity in the B16F10 melanona cells, the inhibition rates of compounds 8, 14 and 13 were with 19.62%, 20.59% and 23.83%, respectively. In addition, compound 13 revealed the highest inhibitory activity to tyrosinase in the melanocyte with inhibition rates of 23.83%, which was better than kojic acid and arbutin (19.21% and 20.45%) at the same concentration. In the anti-melanogenesis experiment, compounds 8 and 13 had better anti-melanin effects than kojic acid from 25 μM to 100 μM. In summary, the results indicated that compounds 8, 13 and 14 had better tyrosinase inhibitory activity and anti-melanogenesis activity. Especially, the compound 13 has potentiality to develop novel tyrosinase inhibitors and whitening agents. The docking studies results revealed that the functional group of compound 13 mostly depends on the phenolic hydroxyl moiety, and its hydroxyl group did not insert into the active site of tyrosinase, which was in agreement with the results of the kinetics study.The secreted Mycobacterium tuberculosis (Mtb) protein tyrosine phosphatase B (MptpB) is an essential virulence factor required for the intracellular survival of Mtb within host macrophages. MptpB has become a promising target for the development of novel anti-tuberculosis (TB) drugs. In this study, two new fusarielins, fusarielins M (1) and N (2), and a biogenetically related known compound, fusarielin G (3) were isolated from the marine-derived fungus Fusarium graminearum SYSU-MS5127. Their inhibitory effects on MptpB were evaluated. Among these compounds, fusarielin M substantially inhibited MptpB with a half-maximal inhibitory concentration (IC50) of 1.05 ± 0.08 μM, and an inhibition constant (Ki) of 1.03 ± 0.39 μM. Surface plasmon resonance analysis was used to characterize the interaction between fusarielin M and MptpB in vitro. Fusarielin M also exhibited cellular activity in blocking MptpB-mediated Erk1/2 and p38 inactivation in macrophages. Importantly, fusarielin M (20 μM) substantially reduced intracellular mycobacterial growth within macrophages, causing a 62% reduction in the bacterial burden. The binding mode of fusarielin M was further explored via molecular docking which suggested that fusarielin M binds to the active site of MptpB, forming a hydrogen bond with the side chain of Asp165; this is unique in the P-loop of MptpB compared to conventional human PTPs. The contact between fusarielin M and Asp165 in the catalytic loop provides a potential basis for inhibitor selectivity. Therefore, fusarielin M shows great potential as an anti-TB drug candidate.Anorexia nervosa (AN) is a devastating psychiatric disorder characterized by extreme restriction of food intake and low body weight, both associated with significant medical and psychological morbidity. REE are currently mined from virgin ores while REE-rich contaminated soil is left untreated in the environment. Soil remediation strategies are needed that concomitantly cleanup soil and harvest metals that contribute to process circular economy. In this review we aim to (i) define REE concentrations in contaminated soils as well as (ii) identify soil remediation techniques used in remediating REE from soils, emphasizing the ones that extract REE. Current literature lists REE polluted soils in the vicinities of REE mines, coal mines, high traffic roads and agricultural soils (due to REE association with phosphate fertilizers). We first list the conventional separation methods used in the mining industry and their main strategies in extracting/precipitating REE. Solvent extraction is the most commonly conventional method used followed by electrodeposition of REE at high temperatures. We then highlight soil remediation techniques that are used to treat REE. These techniques can be separated into two types the ones that (a) stabilize REE in soils, and the ones that (b) extract REE from soils. Bioremediation, soil amendments and others offer stabilization of REE, eventually creating a legacy problem since REE keep accumulating in the soil. Soil remediation techniques that achieve REE extraction are a step closer to resource recovery, contributing to the circularity of REE. Techniques such as phytoremediation, soil washing and electrokinetic treatment show promising extraction results.The present study was conducted to enhance the biosurfactant production yield of Pseudomonas sp. CQ2 isolated from the Chongqing oilfield (China). Besides, the capability of biosurfactant and underlying mechanism for remediation of heavy metal contaminated soil was also investigated. Our results suggested that maximum biosurfactant production (40.7 g/L) was attained at 35 °C by using soybean oil and ammonium nitrate as carbon and nitrogen sources with pH 7, rotational speed of 175 rpm and inoculation ratio of 3%). The removal efficiencies of 78.7, 65.7 and 56.9% for Cd, Cu and Pb respectively were achieved at optimized bioleaching conditions (pH 11, soil/solution ratio 301 and non-sterilized soil), comparative tests between common chemical surfactants (SDS, Tween-80) and biosurfactants demonstrated the larger removal capacity of biosurfactants. Through SEM-EDX, it was found that the granular material disappeared, the content of Cd, Cu and Pb decreased significantly, and the soil surface became smooth with hole formation after soil washing following bioleaching. ATR-FTIR results showed that the carboxyl functional groups in biosurfactants could chelate heavy metals. These results indicated that biosurfactants from Pseudomonas sp. link2 CQ2 could effectively eliminate Cd, Cu, and Pb from soil.The inactivation mechanism of pathogenic microorganisms in water needs to be comprehensively explored in order to better guide the development of an effective and green disinfection method for drinking water safety. Here, metal-free modified g-C3N4 was prepared and used to inactivate two typical bacteria (namely, Gram-positive E. coli and Gram-negative S. aureus) in water under visible light from a comparative perspective. These two bacteria could be inactivated in the presence of modified g-C3N4 within 6 h of visible light, but their inactivation kinetics were quite different. E. link3 coli were inactivated slowly in the early disinfection stage and rapidly in the later disinfection stage, whereas S. aureus were inactivated steadily during the entire disinfection process. Moreover, the impacts of important water parameters (pH, salt, temperature, and water matrix) on photocatalytic inactivation of E. coli and S. aureus were also distinct. In addition, scavenger experiments indicated that superoxide radicals played the most important role in E. coli inactivation, while both superoxide and hydroxyl radicals were important for S. aureus inactivation. Quantitative changes in fatty acids, potassium ions, proteins and DNA of the bacterial suspensions suggested that the higher resistance of E. coli in the early inactivation stage could be originated from the difference in the phospholipid repair system in cell membrane structures. This study can provide new insights into research and development of a safe and effective disinfection technology for drinking water.A tube-feeding model for administering microplastic (MP, Ø = 30 μm) spheres to fish larvae was employed to quantify the uptake of hydrophobic organic contaminants (HOCs) into the larval body through a single administration of MP. Polychlorinated biphenyl-153 (PCB-153) was used as a representative HOC that can be sorbed to MP in the sea. Atlantic herring (Clupea harengus) larvae (34-51 days post-hatching) were selected as the animal model. The herring larvae were tube-fed a single load of up to 200 polystyrene or polyethylene MP spheres spiked with 14C-labelled PCB-153, and the control larvae were tube-fed an isotonic solution without MP. At the time of sampling (24 h post feeding), some larvae had evacuated all MP spheres from the gut, while others still had MP remaining in the gut. In larvae with a significant number of MP spheres still present in the gut, whole-body scintillation counting (including the MP in the gut lumen) showed elevated levels of the tracer compared to those in the control fish larvae. For larvae in which all or almost all MP had been evacuated by the time of sampling, the tracer levels of the whole body were not significantly different compared to those for the control fish larvae.
