Gastrointestinal (GI) and metabolic function are frequently altered in Parkinson's disease (PD). Although enteric nervous system anatomopathological alterations have previously been reported in the 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) monkey model of PD, the resulting gastric emptying and intestinal permeability functional parameters are unknown. The current exploratory study was, thus, designed to investigate these GI functional factors and insulin resistance in the MPTP-treated monkey.
Eight rhesus macaque monkeys (4 controls and 4 MPTP-treated) received the oral acetaminophen absorption test to measure gastric emptying, the oral FITC-dextran absorption test to investigate intestinal permeability, and the intravenous glucose tolerance test to assess insulin resistance. Constipation was evaluated using the Bristol stool scale.
None of the tests, acetaminophen absorption, FITC-dextran absorption or glucose tolerance, showed a difference between control and MPTP-treated monkeys. MPTP-treated monkeys did present signs of transit acceleration.

While the MPTP monkey model reliably displays motor and certain non-motor symptoms of PD, the current study did not demonstrate the GI symptoms associated with PD.The lipophilic compounds product (LCP), which was isolated and purified from Eucalyptus viminalis plants, has shown earlier broad antimicrobial and anti-inflammatory activities. To study secondary compounds responsible for the pharmacological activities, chemical composition of the LCP was studied with application of ultra-performance liquid chromatography combined with photodiode array detector and high-resolution Q Exactive Orbitrap mass spectrometer (UPLC-PDA-HRMS/MS). There were found thirty two compounds twenty phloroglucinol derivatives (isopentyl diformyl phloroglucinol, macrocarpals, sideroxylonals, etc.), eight ursane type triterpenoids (loxanic acid, dehydroursolic acid lactone, dehydroursolic acid lactone acetate, two isomers of p-coumaroyl-dehydroursolic acid lactone and two isomers of feruloyl-dehydroursolic acid lactone), sequiterpenoid (S)-β-macrocarpene and three unknown phenolics. The major compounds of the LCP were pharmacologically active macrocarpals A and B, dehydroursolic acid lactone and its derivatives. It is supposed that previously discovered antimicrobial and anti-inflammatory activities of the LCP is due to the high contents of these secondary compounds.The aim of this study is to develop an accurate and reliable numerical model of the coastal Talar aquifer threatened by seawater intrusion by developing an ensemble meta-model (MM). In comparison with previous methodologies, the developed model has the following superiority (1) Its performance is enhanced by developing ensemble MMs using four different meta-modelling frameworks, i.e., artificial neural network, support vector regression, radial basis function, genetic programing and evolutionary polynomial regression; (2) The accuracy of different MMs based on 16 integration of four meta-modeling frameworks is compared; and (3) the effect of aquifer heterogeneity on the MM. The performance of the proposed MM was assessed using an illustrative case aquifer subject to seawater intrusion. The obtained results indicate that the ensemble MM that combines all four meta-modeling frameworks outperformed the GP and ANN models, with a correlation coefficient of 0.98. Moreover, the proposed MM using nonlinear-learning ensemble of SVR-EPR achieves a better and robust forecasting performance. Therefore, it can be considered as an accurate and robust simulator to predict salinity levels under different abstraction patterns in variable density flow. The result of uncertainty analyses reveals that robustness value and pumping rate are inversely proportional and scenarios with a robustness measure of about 12% are more reliable.We reported an experimental and theoretical spectroscopic studies of (2Z,5Z)-5-(4-nitrobenzylidene)-3-N (2-methoxyphenyl)-2-N' (2-methoxyphenylimino) thiazolidin-4-one (C24H19N3O5S) molecule, using FT-IR, NMR spectroscopy, and density functional theory (DFT) via time-dependent schema (TD-DFT) respectively. The molecular inter-contacts were explored using Hirshfeld surfaces (HS) analysis method. Vibrational frequencies, gauge-independent atomic orbital (GIAO)1H and13C NMR chemical shift values and frontier molecular orbitals (FMOs) have been calculated from the optimized structure of the molecule by DFT/B3LYP functional with 6-31G(d, p) basis set. Our theoretical results show a good agreement with the experimental data. The calculated UV-visible spectrum employing TD-DFT shows electronic transitions at 388 nm and 495 nm. To get insight on the charge interaction happening inside the molecule, HOMO and LUMO were scrutinized and their calculated energy gap was found to be 2.96 eV. The molecular docking was analyzed via interplay study ofacetyl cholinesterase, and Butyrylcholinesterase using molecular docking methodology.
Obesity is a risk factor for different chronic conditions. Over the years, obesity has become a pandemic and it is therefore important that effective diagnostic tools are developed. Obesity is a measure of adiposity and it has become increasingly evident that anthropometric measures such as body mass index (BMI) used to estimate adiposity are inadequate. This study therefore examined the ability of different anthropometric measurements to diagnose obesity within a cross-section of Ghanaian women.
We obtained anthropometric measurements and used that to generate derived measures of adiposity such as body adiposity index (BAI) and conicity index. Furthermore we also measured adiposity using a bioimpedance analyser. Associations between these measurements and percentage body fat (%BF) were drawn in order to determine the suitability of the various measures to predict obesity. The prevalence of obesity was determined using both %BF and BMI.
BMI, Waist and hip circumference and visceral fat (VF) were positivpriate population based cut-off points if anthropometric measurements are to be employed in diagnosing obesity within a particular population.Dimension reduction is often a preliminary step in the analysis of data sets with a large number of variables. Most classical, both supervised and unsupervised, dimension reduction methods such as principal component analysis (PCA), independent component analysis (ICA) or sliced inverse regression (SIR) can be formulated using one, two or several different scatter matrix functionals. Scatter matrices can be seen as different measures of multivariate dispersion and might highlight different features of the data and when compared might reveal interesting structures. Such analysis then searches for a projection onto an interesting (signal) part of the data, and it is also important to know the correct dimension of the signal subspace. These approaches usually make either no model assumptions or work in wide classes of semiparametric models. Theoretical results in the literature are however limited to the case where the sample size exceeds the number of variables which is hardly ever true for data sets encountered in bioinformatics. In this paper, we briefly review the relevant literature and explore if the dimension reduction tools can be used to find relevant and interesting subspaces for small-n-large-p data sets. We illustrate the methods with a microarray dataset of prostate cancer patients and healthy controls.Blastocystis is a parasite commonly found in the gut of humans and animals; there are 22 known subtypes (STs). STs 1-9 and 12 have been found in humans. This parasite has a faecal-oral route of transmission; its high infection prevalence in developing countries is due to poor hygiene practices, exposure to infected animals, and intake of contaminated water or food. Its pathogenicity has not been established, because it has been found in symptomatic and asymptomatic patients. The goal of this study was to analyze the frequency of Blastocystis and its subtypes (1, 2, 3, 4, 5, and 7), and assess the relationship between these subtypes and abdominal pain and distension. 202 university students participated in this study. A questionnaire was applied to assess the gastrointestinal symptoms, and subsequently the students were asked to provide faecal samples. The presence of parasites was determined by optical microscopy. Blastocystis-positive samples had their DNA extracted and end-point PCR was performed to corroborate the presence of Blastocystis and determine its subtypes. Among the samples, 47.03% were positive according to PCR analysis. The most prevalent subtypes were ST3 (29.79%), ST4 (16.84%), and ST1 (14.89%). We found a relationship between ST1 and abdominal pain (OR = 0.196; CI = 0.0533-0.7318; p = 0.015), and between ST4 and abdominal distension (OR = 0.2928; CI = 0.1017-0.8429; p = 0.023). However, the presence of this parasite and the probable relationship with gastrointestinal symptoms suggest the need to determine its role within intestinal microbiota in order to confirm whether its eradication is really necessary or not.Mechanistic modeling constitutes a powerful tool to unravel complex biological phenomena. This study describes the construction of a mechanistic, dynamic model for grapevine plant growth and canopy biomass (vigor). To parametrize and validate the model, the progeny from a cross of Ramsey (Vitis champinii) × Riparia Gloire (V. riparia) was evaluated. Plants with different vigor were grown in a greenhouse during the summer of 2014 and 2015. Selisistat mw One set of plants was grafted with Cabernet Sauvignon. Shoot growth rate (b), leaf area (LA), dry biomass, whole plant and root specific hydraulic conductance (kH and Lpr), stomatal conductance (gs), and water potential (Ψ) were measured. Partitioning indices and specific leaf area (SLA) were calculated. The model includes an empirical fit of a purported seasonal pattern of bioactive GAs based on published seasonal evolutionary levels and reference values. The model provided a good fit of the experimental data, with R = 0.85. Simulation of single trait variations defined the individual effect of each variable on vigor determination. The model predicts, with acceptable accuracy, the vigor of a young plant through the measurement of Lpr and SLA. The model also permits further understanding of the functional traits that govern vigor, and, ultimately, could be considered useful for growers, breeders and those studying climate change.Laurel (Laurus nobilis L.) is a plant species from Lauraceae family, and is native to the Mediterranean region. The goal of this study was to compare chemical composition of laurel leaves and antibacterial activity of its essential oil (EO) from wild-grown trees in Greece and Georgia. The laurel leaves from the two native habitats had dissimilar concentrations of phenolic acids. Of the conjugated flavonols and flavons, kaempferol (1981.3 μg/g) and apigenin (1433.6 μg/g) were the major representatives in the leaves from Greece, while luteolin (839.1 μg/g) and kaempferol (688.1 μg/g) were the major ones in the leaves from Georgia, respectively. The EO content was 1.42% and 4.54% in the leaves from Greece and Georgia, respectively. The main EO constituents of the Greek laurel plants were 1,8-cineole (30.8%), α-terpinyl acetate (14.9%), α-terpineol (8.0%), sabinene (7.9%), and terpinen-4-ol (6.0%).
