Synchrotron-generated microbeam radiation therapy (MRT) represents an innovative preclinical type of cancer radiation therapy with an excellent therapeutic ratio. Beyond local control, metastatic spread is another important endpoint to assess the effectiveness of radiation therapy treatment. Currently, no data exist on an association between MRT and metastasis. Here, we evaluated the ability of MRT to delay B16F10 murine melanoma progression and locoregional metastatic spread.
We assessed the primary tumor response and the extent of metastasis in sentinel lymph nodes in 2 cohorts of C57BL/6J mice, one receiving a single MRT and another receiving 2 MRT treatments delivered with a 10-day interval. We compared these 2 cohorts with synchrotron broad beam-irradiated and nonirradiated mice. In addition, using multiplex quantitative platforms, we measured plasma concentrations of 34 pro- and anti-inflammatory cytokines and frequencies of immune cell subsets infiltrating primary tumors that received either 1 or 2 MRT treatments.
Two MRT treatments were significantly more effective for local control than a single MRT. Remarkably, the second MRT also triggered a pronounced regression of out-of-radiation field locoregional metastasis. Augmentation of CXCL5, CXCL12, and CCL22 levels after the second MRT indicated that inhibition of melanoma progression could be associated with increased activity of antitumor neutrophils and T-cells. Indeed, we demonstrated elevated infiltration of neutrophils and activated T-cells in the tumors after the second MRT.
Our study highlights the importance of monitoring metastasis after MRT and provides the first MRT fractionation schedule that promotes local and locoregional control with the potential to manage distant metastasis.

To develop an automated lung tumor segmentation method for radiation therapy planning based on deep learning and dual-modality positron emission tomography (PET) and computed tomography (CT) images.
A 3-dimensional (3D) convolutional neural network using inputs from diagnostic PETs and simulation CTs was constructed with 2 parallel convolution paths for independent feature extraction at multiple resolution levels and a single deconvolution path. At each resolution level, the extracted features from the convolution arms were concatenated and fed through the skip connections into the deconvolution path that produced the tumor segmentation. Our network was trained/validated/tested by a 311 split on 290 pairs of PET and CT images from patients with lung cancer treated at our clinic, with manual physician contours as the ground truth. A stratified training strategy based on the magnitude of the gross tumor volume (GTV) was investigated to improve performance, especially for small tumors. Multiple radiation oncegy generated clinically useful lung cancer contours that were highly acceptable on physician review.

To date, there is growing evidence that calcium channel blockers (CCBs) reduce oxidative stress and development of inflammation in the nervous system. Therefore, the aim of the present study was to evaluate the impact of such CCBs as Nifedipine, Verapamil, and MK-801 on the expression of peroxisome proliferator-activated receptor (Pparγ), i.e. ROS-related and inflammation-related proteins, in mouse astrocytes exposed in vitro to the VGVAPG peptide. While previous studies intensively evaluated air pollution episodes and extreme heat events, a limited number of studies comprehensively assessed atmospheric hot-and-polluted-episodes (HPE) - an episode with simultaneously high levels of air pollution and temperature - which have potential adverse synergic impacts on human health. This study focused on the Pearl River Delta (PRD) region of China due to its high temperature in summer and poor air quality throughout a year. We employed geostatistical downscaling to model meteorology at a spatial resolution of 1 km, and applied a machine learning algorithm (XGBoost) to estimate a high-resolution (1 km) daily concentration of particulate matter with an aerodynamic diameter ≤2.5 μm (PM2.5) and ozone (O3) for June to October over 20 years (2000-2019). Our results indicate an increasing trend (∼50%) in the frequency of HPE occurrence in the first decade (2000-2010). Conversely, the annual frequency of HPE occurrence reduced (16.7%), but its intensity increased during the second decade (2010-2019). The northern cities in the PRD region had higher levels of PM2.5 and O3 than their southern counterparts. During HPEs, regional daily PM2.5 exceeded the World Health Organization (WHO) and Chinese guideline levels by 75% and 25%, respectively, while the O3 exceeded the WHO O3 standard by up to 69%. Overall, 567,063 (95% confidence interval (CI) 510,357-623,770) and 52,231 (95%CI 26,116-78,346) excessive deaths were respectively attributable to exposure to PM2.5 and O3 in the PRD region. Our findings imply the necessity and urgency to formulate co-benefit policies to mitigate the region's air pollution and heat problems.Microplastics have been proven to be hotspots of bacterial pathogens and antibiotic resistance genes (ARGs). The enrichment of ARGs in microplastisphere, the specific niche for diverse microbial communities attached to the surface of microplastic, has attracted worldwide attention. By collecting 477 pairs of ARG abundance data belonging to 26 ARG types, based on the standardized mean difference (SMD) under the random effect model, we have performed the first meta-analysis of the ARG enrichment on microplastics in aquatic environments in order to quantitatively elucidate the enrichment effect, with comparison of non-microplastic materials. It was found that ARGs enriched on the microplastics were more abundant than that on the inorganic substrates (SMD = 0.26) and natural water environments (SMD = 0.10), but lower abundant than that on the natural organic substrates (SMD = -0.52). Furthermore, microplastics in freshwater tended to have a higher degree of ARG enrichment than those in saline water and sewage. The biofilm formation stage, structure, and component of microplastisphere may play a significant role in the enrichment of ARGs.Microplastics (MPs), plastic particles less then 5 mm in diameter, have become an emerging ubiquitous concern for the environment. Rivers are the primary pathways that transport MPs from the land to the ocean; however, standardized methodologies for in-situ sampling in freshwater environments remain undefined. Notably, uncertainties in MP sampling methods lead to errors in estimating MP discharge through rivers. In the present study, the inter-sample variance of plankton net-obtained MP concentrations for two urban rivers in Japan was investigated. Numerical concentrations, expressed in particles·m-3, revealed that variance s2 was proportional to the mean m of replicated estimates of numerical concentrations. A derived statistical model suggested that river MPs disperse according to purely random processes; that is, Poisson point processes. Accordingly, a method was established to project the "precision," the ratio of the standard error to m, of numerical concentrations based on the number of net sampling repetitions. It was found that the mean of two replicates maintained sufficient precision of less then 30% for conditions with high concentrations of ≥3 particles·m-3. Projected precisions under different levels of MP concentrations are also presented to help design future field campaigns.
